Aspergillus flavus (Link) toxins reduces the fitness of dengue vector Aedes aegypti (Linn.) and their non-target toxicity against aquatic predator.

Mosquito that accountable for dispersal of dengue fever is Aedes aegypti Linn. and considered to be a chief vector for dengue especially in South Asian countries. Aspergillus flavus is considered to be wild growing green yellow colonies and synthesis highly regulating aflatoxins (B1, B2, G1 and G2) as a secondary metabolite. Mycotoxins of A. flavus showed its efficacy against III and IV instars of Ae. aegypti with more than 90% mortality at the prominent dosage of 2 × 108 conidia/ml. The proximate lethal concentrations (LC50 and LC90) of mycotoxins against third and fourth instars was 2 × 105 …

Zearalenone exposure and the mammalian gut microbiome: a systematic review of the literature.

Mycotoxins are fungal-derived metabolites found in the global food supply, particularly in cereal grains and processed foods. Zearalenone (ZEN), one of the more common mycotoxins, widely contaminates human food supplies at concentrations above established maximum contamination levels. However, the effects of ZEN exposure on human health have only begun to be explored, with most studies on this topic focusing on its endocrine-disrupting effects in the reproductive system. ZEN is readily absorbed following oral administration, making the gastrointestinal (GI) tract and its microbiota primary tar…

A portable electrochemical platform for biorecognition-free detection of ochratoxin A and ascorbic acid for food safety and quality monitoring.

Even low concentrations of mycotoxins in food and agricultural products can be extremely hazardous for the health of both humans and livestock animals. Ascorbic acid, commonly referred to as vitamin C, can potentially reduce the levels of reactive oxygen species (ROS) generated by mycotoxin exposure. For the first time, this study reports the development of a biorecognition molecule-free electrochemical sensor that can simultaneously detect both the mycotoxin ochratoxin-A (OTA) and ascorbic acid (AA). Here, we present a paper-based electrode modified with tellurium oxide (TeO2) sheets and nick…

Study on the Dynamic Changes in Fungal Communities During the Storage of Polygalae Radix and the Antifungal Effects of Peppermint Essential Oil.

Polygalae Radix, a traditional Chinese medicine for insomnia and memory disorders, is highly susceptible to fungal contamination and mycotoxin production (especially by Aspergillus flavus) during storage, compromising its safety and efficacy. Therefore, in this study, high-throughput sequencing was employed to evaluate the dynamic changes in fungal communities during the storage of Polygalae Radix and to analyze common mycotoxin-producing genera. Furthermore, the inhibitory effects of peppermint essential oil (PEO) on A. flavus were assessed through fumigation treatments, combined with colony

Exposure assessment of the European adult population to deoxynivalenol - Results from the HBM4EU Aligned Studies.

Mycotoxins are natural toxins produced by fungi that may cause adverse health effects thus constituting a public health concern. Deoxynivalenol (DON), a mycotoxin affecting the immune system and causing intestinal disorders, was selected as a priority under the European Human Biomonitoring Initiative (HBM4EU). Urinary total DON levels (tDON) of 1270 participants from six countries were used to characterize the internal exposure of the adult European population and identify the most relevant determinants of exposure. tDON concentrations' P50 and P95 were in the range of 0.41-10.16 µg/L (0.39-9.…

Zearalenone Adsorbent Based on a Lyophilized Indigenous Bacterial Lactobacillus plantarum Strain as Feed Additive for Pigs: A Preliminary Study In Vivo.

Feed contamination with naturally occurring mycotoxins is an unavoidable condition of significant concern in intensive productions. The presence of high concentrations of zearalenone >1 ppm in the diet can cause major reproductive disorders, particularly in swine. In order to reduce the consequences of intoxication, mycotoxin adsorbents are incorporated into feed. In the present study, zearalenone adsorption capacity of a lyophilized indigenous strain of Lactobacillus plantarum (L4; previously isolated from pig's rectal swabs) was first evaluated in vitro. A preliminary study in vivo was then …

Fumonisin B1 contamination of maize and experimental acute fumonisin toxicosis in pigs.

The fumonisin B1 content of 69 visibly mouldy and 23 mould-free maize samples grown in Hungary in 1993-1995 was determined by high-performance liquid chromatography (HPLC). Fumonisin B1 was found to occur in 70-73% of the mouldy samples. The mycotoxin level increased from year to year: the highest fumonisin B1 concentration was 75.1 mg/kg. The samples that were mould-free on visual inspection showed a much lower prevalence of fumonisin B1 contamination (30%) and contained fumonisin B1 in markedly lower concentrations (average, 1.52 mg/kg; maximum concentration, 5.1 mg/kg). Using the Fusarium m

In vitro metabolism of fumonisin B1 by ruminal microflora.

Fumonisin B1 (FB1) is a mycotoxin produced by Fusarium moniliforme and F. proliferatum. Little is known of its metabolic fate after oral ingestion in ruminants, but these animals are reported to be tolerant towards FB1. The metabolism of this mycotoxin was evaluated following incubation (1 microg/ml) in ruminal fluid for up to 72 h, in the presence or absence of alfalfa as a substrate for microbial growth, using a model rumen (sealed flask, anaerobic conditions, exclusion of light, gentle agitation, 39 degrees C). The decrease in FB1 concentration and the production of short-chain fatty acids

Listeriosis in diacetoxyscirpenol-treated mice.

Mice were treated with the trichothecene mycotoxin diacetoxyscirpenol (DAS) and subsequently were inoculated intraperitoneally with Listeria monocytogenes. The effect of the mycotoxin on the course of the infection was monitored by observing the resultant mortality and the bacterial content of the spleens from inoculated mice. Mice given 3 mg of DAS/kg of body weight, PO, at days -2 and -1 before inoculation had increased mortality and splenic Listeria counts. In these mice, thymus weights were reduced, and lymphocytes were depleted from the thymus cortex and from splenic lymphoid follicles an

Hepato- and cardiotoxicity of xanthoascin, a new metabolite of A. candidus Link, to mice. I. Blood chemistry and histological changes in mice.

Aspergillus candidus Link, one of the commonest constituents of cereal mycoflora, produces two kinds of mycotoxins, terphenyllin and xanthoascin, which show different chemical and toxicological properties. The latter, xanthoascin, caused severe hepatic injury with jaundice and focal or confluent necrosis of hepatocytes, when given to mice in doses of 6 mg/kg b.w. or higher by a single subcutaneous injection. With higher doses above 15 mg/kg, myocardial degeneration and necrosis was induced after a week or two in addition to the hepatic injury. Vacuolation of the nuclei of the alveolar intersti…

Phytotoxins Produced by Diplodia and Other Fungal Species Involved in Oak (Quercus) Diseases.

Forests are an indispensable resource for human existence in the different regions of the world. Forests are often concentrated in a few parts and vary in size. The most important and extensive green reserve on Earth is the Amazon rainforest. Unfortunately, the forests are continually suffering massive losses due to abiotic stresses and deforestation to obtain other arable lands and microbial diseases, particularly those caused by pathogenic fungi. Pathogenic fungi play an important role in inducing oak decline, which is one of the most relevant forest diseases globally. The pathogens also aff

Orellanine: From Fungal Origin to a Potential Future Cancer Treatment.

Fungal metabolites represent an underutilized resource in the development of novel anticancer drugs. This review will focus on the promising fungal nephrotoxin orellanine, found in mushrooms including Cortinarius orellanus (Fools webcap). Emphasis will be placed on its historical significance, structural features, and associated toxicomechanics. Chromatographic methods for analysis of the compound and its metabolites, its synthesis, and chemotherapeutic potential are also discussed. Although orellanine's exceptional selectivity for proximal tubular cells is well documented, the mechanics of it

A novel gene cluster in Fusarium graminearum contains a gene that contributes to butenolide synthesis.

The development of expressed sequence tag (EST) databases, directed transformation and a sequenced genome has facilitated the functional analysis of Fusarium graminearum genes. Extensive analysis of 10,397 ESTs, derived from thirteen cDNA libraries of F. graminearum grown under diverse conditions, identified a novel cluster of eight genes (gene loci fg08077-fg08084) located within a 17kb region of genomic sequence contig 1.324. The expression of these genes is concomitantly up-regulated under growth conditions that promote mycotoxin production. Gene disruption and add-back experiments followed

Ochratoxin A in human sera in the area with endemic nephropathy in Croatia.

Ochratoxin A (OA) is nephrotoxic fungal metabolite (mycotoxin) occurring in foodstuffs. The compound is causally associated with mycotoxin porcine nephropathy, a disease comparable with a human kidney disease called endemic nephropathy (EN). In this paper we presented results obtained over a 10-year period in the hyperendemic village Kaniza, and in control villages where no clinical cases of nephropathy had been found. In the hyperendemic village Kaniza and non-endemic villages the incidence of OA in human blood was up to 4.5% (range 2-50 ng/ml) and up to 2.4% (range 2-10 ng/ml), respectively.

Antifungal and antimycotoxin activity of plant-synthesized silver nanoparticles (AgNPs): A novel approach to combat food contamination.

Fungi and mycotoxins hugely contaminate food and feed commodities and consequently contribute to adverse effects on health and the economy. While various measures have been adopted to control food contamination, the potential application of nanotechnology to control fungi and mycotoxins in food seems promising; nevertheless, it has not been sufficiently explored. This study investigated the antifungal effect of plant-mediated silver nanoparticles (AgNPs) form Garcinia kola, Carica papaya, Achillea millefolium, Ocimum gratissimum, and Perilla frutescens against ten food-borne fungi. Furthermore…
