Human placental conditioned medium reverses apparent commitment to differentiation of human promyelocytic leukemia cells (HL60).

Using a system of sequential daughter cell transfers in semisolid medium, we have analyzed self-renewal and differentiation of human promyelocytic leukemia cells (HL60) in presence of all-trans retinoic acid and human placental conditioned medium (HPCM). We find that retinoic acid induces coordinated losses of self-renewal potential which are followed by phenotypic differentiation. The latter occurs as an all-or-none event and is reversible in the presence of HPCM. Thus, HL60 cells that apparently had terminally differentiated (as estimated by the ability to reduce nitroblue tetrazolium [NBT])

EphB2 activation is required for ependymoma development as well as inhibits differentiation and promotes proliferation of the transformed cell.

Our intracranial implantation mouse model of ependymoma clearly demonstrates overexpression of the ephrin receptor EphB2 in Ink4a/Arf((-/-)) supratentorial embryonic neural stem cells (STeNSCs) to be essential for transformation and disease development; however the requirement for and consequence of receptor activation on transformation and neural stem cell function were not examined. We definitively illustrate the necessity for receptor activation in cellular transformation and the importance of implantation site and microenvironment in directing ependymoma development. In vitro assays of Eph

Statins, stem cells, and cancer.

The statins (3-hydroxy-3-methylglutaryl coenzyme A reductase inhibitors) were proven to be effective antilipid agents against cardiovascular disease. Recent reports demonstrate an anticancer effect induced by the statins through inhibition of cell proliferation, induction of apoptosis, or inhibition of angiogenesis. These effects are due to suppression of the mevalonate pathway leading to depletion of various downstream products that play an essential role in cell cycle progression, cell signaling, and membrane integrity. Recent evidence suggests a shared genomic fingerprint between embryonic

Niches in cancer: from primary tumors to metastatic disease.

Cancer progression does not occur in isolation: malignant-cell states and fates are shaped by specialized microenvironments, or cancer niches. Within primary tumors, vascular, stromal, immune and neural interactions create local hypoxic, metabolic and mechanical conditions that together influence malignant-cell behavior. These local environments sustain stem-like states, regulate immune surveillance and vascular remodeling, and influence dissemination and treatment response. Tumor-derived extracellular vesicles and other systemic signals connect primary tumors to distant organs, conditioning t

The diagnostic potential of nanobodies in acute myeloid leukemia.

Acute myeloid leukemia (AML) is a heterogeneous hematologic cancer, which is caused by clonal expansion of immature myeloid precursors, leading to bone marrow failure and poor long-term survival, especially in older patients and those with adverse-risk features. Relapse, primarily from chemotherapy-resistant leukemic stem cells (LSCs) in protective niches, remains the leading cause of mortality despite genomic advancements and targeted therapies like FLT3 inhibitors and all-trans retinoic acid for acute promyelocytic leukemia. Conventional antibody-based approaches, such as gemtuzumab ozogamic

Detection of Intratumoral Immune Cell Antigens in an Immunocompetent Glioblastoma Stem-like Cell Model Using Substrate-Based Immunohistochemistry.

Glioblastoma (GBM) is the most common and lethal primary brain malignancy, lacking effective therapies. It is characterized by a profoundly immunosuppressive tumor microenvironment, driven in part by treatment-resistant GBM stem-like cells (GSCs). Here, we describe reproducible substrate-based single- and dual-color immunohistochemistry (IHC) protocols for detecting tumor-infiltrating immune cell-associated surface and intracellular antigens in formalin-fixed, paraffin-embedded (FFPE) brain tumor sections. The brain tumors were derived from mouse GSCs, which were orthotopically implanted in im

KIAA1199: A Novel Regulator of Chemoresistance in Human Cancer Cells.

KIAA1199 (also known as CEMIP or HYBID), an archived gene listed in the Human Unidentified Gene-Encoded (HUGE) database, has been implicated in tumor progression and metastasis in a wide range of cancers, including breast, cervical, cholangiocarcinoma, pancreatic, colorectal, gastric, hepatocellular, lung, ovarian, prostate, papillary thyroid, and brain cancers. Overexpression of KIAA1199 is significantly correlated with poor prognosis and metastasis and has been clinically associated with chemoresistance. In addition, KIAA1199 may serve as a novel biomarker for cancer diagnosis and prognosis,

Role of chemokines in metastatic niche: new insights along with a diagnostic and prognostic approach.

Chemokines are cytokines that are involved in the movement of leukocytes and the occurrence of immune responses. It has recently been noted that these cytokines play a role in the movement of cancer cells to different parts of the body and create a suitable environment [i.e. (pre) metastatic niche] for their growth and proliferation. We studied the role of chemokines in the metastasis of cancer cells, as well as their involvement in the proliferation and growth of these cells. Relevant literature was identified by a PubMed search (2005-2017) of English language papers using the terms 'chemokin

Expression of Idh1R132H in the Murine Subventricular Zone Stem Cell Niche Recapitulates Features of Early Gliomagenesis.

Isocitrate dehydrogenase 1 mutations drive human gliomagenesis, probably through neomorphic enzyme activity that produces D-2-hydroxyglutarate. To model this disease, we conditionally expressed Idh1R132H in the subventricular zone (SVZ) of the adult mouse brain. The mice developed hydrocephalus and grossly dilated lateral ventricles, with accumulation of 2-hydroxyglutarate and reduced α-ketoglutarate. Stem and transit amplifying/progenitor cell populations were expanded, and proliferation increased. Cells expressing SVZ markers infiltrated surrounding brain regions. SVZ cells also gave rise to

Symbiotic exclusivity between CLOCK and TFPI2 drives stemness and immunosuppression in glioblastoma models.

Glioblastoma (GBM) is a highly aggressive brain tumor characterized by extensive crosstalk between glioblastoma stem cells (GSCs) and immunosuppressive microglia, with our previous work identifying CLOCK and TFPI2 as key regulators of this interaction. Here, we uncover a 'symbiotic exclusivity' pattern between CLOCK and TFPI2, showing that, despite mutually exclusive amplifications, they sustain symbiotic regulatory interactions in GBM. The CLOCK-BMAL1 complex transcriptionally upregulates TFPI2, while TFPI2-driven hypoxia inducible factor 1 α (HIF-1α) signaling activates nuclear factor k B (N

Cancer Stem Cell Quiescence: Molecular and Microenvironmental Evidence for an Actively Regulated Adaptive Survival Strategy.

Cancer stem cells (CSCs) are a therapy-refractory subpopulation that drives tumor persistence, recurrence, and metastasis. A defining characteristic of CSCs is their ability to reversibly exit the cell cycle and enter a quiescent state, thereby escaping cytotoxic therapies that primarily target proliferating cells. Although numerous reviews have examined CSC dormancy and quiescence, less attention has been devoted to recently identified regulators and to the molecular events associated with the transition into quiescence. This narrative review consolidates current knowledge on CSC-specific ent

Primary and secondary cell death in human melanoma xenografts following hyperthermic treatment.

The response to hyperthermic treatment (42.5 degrees C for 60 min) of 5 human malignant melanomas grown in athymic mice (BALB/c/nu/nu/BOM) was studied. Local hyperthermia was given by immersing the tumor-bearing leg of the mice into a thermostatically regulated water bath. Growth delay studies indicated that the melanomas were different in heat responsiveness. The differences were confirmed by measuring the fraction of clonogenic cells in the melanomas as a function of time after treatment. The latter experiment showed that some tumor cells were inactivated during the treatment, while others l

Effects of melatonin on apoptosis and cell differentiation in MCF-7 derived cancer stem cells.

Melatonin is a hormone of the pineal gland that has a wide range of biological effects such as antioxidant, anti-inflammatory, and anti-tumor activity. Previous studies have shown that melatonin also affects survival, proliferation, and apoptosis of the cells. In this study, we investigated the effect of melatonin on apoptosis, self-renewal, and differentiation. For this purpose, MCF-7 and HEK293 cells were subjected to melatonin treatment. Expression of genes related to apoptosis (Bax and Bcl2) and self-renewal and differentiation (Oct4, Sox2, and Nanog) analyzed after the sorting of cancer s

Bulk Sequencing Combined With Single-Cell Sequencing Identifies High Expression of VCAN in Fibroblasts Promoting the Progression of High-Stemness Gastric Adenocarcinoma Cells.

Gastric cancer ranks as the fifth most prevalent malignancy and the fifth leading cause of cancer-related mortality worldwide, with stomach adenocarcinoma (STAD) being the predominant pathological type. Despite a decline in STAD incidence in recent years, factors such as an aging population may contribute to an increase in future cases. Current clinical management of STAD primarily involves surgical resection, radiation, chemotherapy, and emerging immune checkpoint blockade (ICB) therapies. However, the limited efficacy of ICB monotherapy in most patients with STAD highlights the need for nove

[Hepatic neoductules].

Proliferation of preexisting bile ducts, ductular metaplasia of hepatocytes and proliferation and differentiation of liver stem cells are discussed in the pathogenesis of neoductular structures in the liver. Under the condition of experimental bile duct obstruction and in extrahepatic bile duct stenosis neoductular structures are first the result of proliferation and sprouting of preexisting ducts and cholangioles. Especially in later stages of cholestasis but also in other chronic progredient liver diseases such as chronic alcoholic liver disease and chronic active hepatitis periportal hepato

Dedifferentiation-driven oncogenic stemness promotes tumor-sustaining adaptability in the intestinal epithelium.

Intestinal tumorigenesis can occur via two distinct routes: bottom-up tumorigenesis occurs from mutations sustained in the Lgr5⁺ stem cells, whereas top-down tumorigenesis is driven by dedifferentiation of epithelial cells near the intestinal lumen. While sporadic human colon adenomas exhibit features of top-down tumorigenesis, their biological determinants remain elusive. Here, using a Smad4 loss-of-function and β-catenin gain-of-function (Smad4LOF:β-cateninGOF) mouse model, we demonstrate that dedifferentiation-derived oncogenic stem cells sustain tumorigenesis more effectively than endogeno

Single-cell and bulk transcriptomics uncovers PRKD2-driven tumor stemness and progression in multiple myeloma.

Multiple myeloma (MM) remains incurable, as most patients relapse or become refractory (RRMM), and the molecular nexus among clonal evolution, immune escape and therapy response remains unknown. Bulk RNA-seq from 24 MM to 6 matched marrows (subset of 132) plus single-cell datasets were analysed by differential expression, pseudotime, WGCNA, enrichment, immune deconvolution and GDSC drug screens. PRKD2 was knocked down (KD) or over-expressed (OE) in H929, 8226 and U266 cells. Viability (CCK-8) and apoptosis (Annexin V/7-AAD) were assessed after 48 h of axitinib (1.25-10 µM). PRKD2 was the sole

The propensity of invasive circulating tumor cells (iCTCs) in metastatic progression and therapeutic responsiveness.

Circulating tumor cells (CTCs) are important clinical indicators of metastatic progression and treatment efficacy. However, because of their low number and heterogeneity, reliable patient-derived CTC models are not readily available. We report here the isolation and characterization of the invasive population of CTCs, iCTCs, from blood of 10 patients with epithelial ovarian cancer (EOC) and one pancreatic cancer patient based on the avidity of tumor cells toward an artificial collagen-based adhesion matrix (CAM), in comparison with tumor progenitor (TP) cells isolated from tumor cell lines, tu
