A culture procedure for rat third molars suitable for nutritional-developmental studies is described. Unerupted third molars from 12-day-old rats were cultured in BGJb media containing 20 per cent rat serum and supplemented with 25 mM HEPES buffer, 25 mg ascorbic acid, 20 mg L-glutamine, 12 mg penicillin G and 10 mg streptomycin sulphate per 100 ml of media. Molars were cultured at the liquid-gas interphase using a 50 per cent O2, 45 per cent N2, 5 per cent CO2 gas mixture at 10 lb-psig (pounds per square inch guage). Molar cultures were maintained successfully for 9-14 days without evidence o

The developing tooth comprises diverse and highly specialized cell populations that work together to maintain proper form and function. Elucidating the interactions among these cells and their surrounding microenvironment is critical for understanding the regulatory mechanisms underlying normal tooth development. Perturbations in these processes can result in congenital disorders such as tooth agenesis, dentinogenesis imperfecta, and amelogenesis imperfecta. Despite the substantial progress enabled by single-cell RNA sequencing (scRNA-seq) in revealing cellular heterogeneity, it does not prese

BACKGROUND: Interstitial brachytherapy is an effective and organ-preserving radiotherapeutic modality for pediatric and adolescent patients with head and neck tumors, offering favorable outcomes with minimal impact on surrounding normal tissues. However, the long-term effects of brachytherapy on dental development in young patients remain underexplored, despite clinical observations of tooth anomalies in tumor survivors. This study aimed to provide longitudinal outcomes of tooth developmental anomalies in pediatric and adolescent tumor survivors who underwent head and neck interstitial brachyt

The purpose of this retrospective cross-sectional study conducted in a Spanish population, was to determine whether dental development is delayed in paediatric patients with agenesis. The comparative analysis of the average difference between chronological and dental age was carried out to assess the validity of maturation tables for the Spanish population. The study examined the potential influence of the following variables: sex, number of agenesis, location of the agenesis. CONCLUSION: Dental development in the studied sample representing Spanish population, is notably more delayed in chil

Dental follicle cells (DFCs) activate and recruit osteoclasts for tooth development and tooth eruption, whereas DFCs themselves differentiate into osteoblasts to form alveolar bone surrounding tooth roots through the interaction with Hertwig's epithelial root sheath (HERS). Also during tooth development, parathyroid hormone-related peptide (PTHrP) is expressed surrounding the tooth germ. Thus, we aimed to investigate the effect of PTHrP (1-34) on bone resorption and osteogenesis of DFCs in vitro and in vivo. In vitro studies demonstrated that DFCs cocultured with HERS cells expressed higher le

MicroRNAs (miRNAs) are a class of small, non-coding RNAs that provide an efficient pathway for regulation of gene expression at a post-transcriptional level. Tooth development is regulated by a complex network of cell-cell signaling during all steps of organogenesis. Most of the congenital dental defects in humans are caused by mutations in genes involved in developmental regulatory networks. Whereas the developmental morphological stages of the tooth development already are thoroughly documented, the implicated genetic network is still under investigation. The involvement of miRNAs in the reg

INTRODUCTION: Hypodontia is the most frequent developmental anomaly of the orofacial complex, and its detection in prenatal ultrasound may indicate the presence of congenital malformations, genetic syndromes and chromosomal abnormalities. To date, only a few studies have evaluated the histological relationship of human tooth germs identified by two-dimensional (2D) ultrasonography. In order to analyze whether two-dimensional ultrasonography of tooth germs may be successfully used for identifying genetic syndromes, prenatal ultrasound images of fetal tooth germs obtained from a Portuguese popul

OBJECTIVE: The aim of this study was to investigate the prevalence of selected dental variations in association with infraocclusion, as well as determining the effects of infraocclusion on dental development and tooth size, in singletons and twins.
DESIGN: Two samples were analysed. The first sample comprised 1454 panoramic radiographs of singleton boys and girls aged 8-11 years. The second sample comprised dental models of 202 pairs of monozygotic and dizygotic twins aged 8-11 years. Adobe Photoshop CS5 was used to construct reference lines and measure the extent of infraocclusion (in mm) of

AIM: To report a rare case of arrested root formation of permanent incisors in a young boy with mixed dentition and its management.
BACKGROUND: Traumatic injury to a primary tooth may damage the underlying permanent tooth germ and affect its development. The extent of the malformation depends upon the developmental stage of the permanent tooth and the intensity of trauma. Discoloration and hypoplasia of the crown, dilaceration, root angulation and disturbances in eruption are commonly seen developmental disturbances following trauma. However, partial or complete arrest of root formation is a

Periodontitis is a highly prevalent inflammatory disease that results in damage to the tooth-supporting tissues, potentially leading to tooth loss. Periodontal tissue regeneration is a complex process that involves the collaboration of two hard tissues (cementum and alveolar bone) and two soft tissues (gingiva and periodontal ligament). To date, no periodontal-regenerative procedures provide predictable clinical outcomes. To understand the rational basis of regenerative procedures, a better understanding of the events associated with the formation of periodontal components will help to establi

Intrusion of permanent teeth is one of the most severe types of traumatic injuries. Different treatment strategies have been proposed, and the best approach for each case depends on the stage of root development, severity of the intrusive luxation, presence of alveolar fracture and number of intruded teeth. The purpose of this paper is to describe the treatment management of 2 cases of severely intruded immature permanent maxillary central incisors in 7 1/2- and 8-year-old children. In case 1, the traumatized tooth was treated by surgical repositioning, while in case 2 watchful waiting for spo

Clastic cells are responsible for mineralized tissue resorption. Bone resorbing cells are called osteoclasts; however, they are able to resorb mineralized dental tissues or calcified cartilage and then they are called odontoclasts and chondroclasts, respectively. They derive from mononuclear precursors of the monocyte-macrophage lineage from hemopoietic tissue, reach target mineralized tissues and degrade them under many different physiologic or pathologic stimuli. Clastic cells play a key role in calcium homeostasis, and participate in skeletal growth, tooth movement, and other physiological

The transforming growth factor-beta (TGF-beta) family of cytokines consists of multi-functional polypeptides that regulate a variety of cell processes, including proliferation, differentiation, apoptosis, extracellular matrix elaboration, angiogenesis, and immune suppression, among others. In so doing, TGF-beta plays a key role in the control of cell behavior in both health and disease. In this report, we review what is known about the mechanisms of activation of the peptide, together with details of TGF-beta signal transduction pathways. This review summarizes the evidence implicating TGF-bet

An etiologic connection between palatally ectopic canines and small and missing teeth is well established in the literature. Additionally, it has been observed that patients with palatally ectopic canines have a delayed dental development. The present study was designed to examine the validity of this latter observation. We radiographically assessed the subjects' dental ages using criteria of tooth calcification, rather than tooth eruption pattern. A similar determination was made in relation to subjects in whom buccally ectopic canines were present. The experimental group consisted of panoram

Coronal resorption of maxillary canines is a possible complication of long standing impaction. Degeneration of the enamel epithelium allows the connective tissues to come into direct contact with enamel. Irregular replacement of the pericoronal tissues and resorbed enamel by bone characterizes the histological picture. Inflammatory changes are rare. Resorption of enamel is slow and usually symptomless so that reported cases have usually been incidental radiographic findings in older patients. This report presents the early arrested development, ankylosis and resorption of the crown of a maxill

Enamel and dentin thicknesses were measured in permanent tooth crowns of 47,XXY (Klinefelter syndrome) males. In 47,XXY males, enamel thickness in maxillary central incisors was significantly greater than that in control males or females, and dentin thickness in incisors and canines was significantly greater than that in control females, but smaller than that in normal males. These findings confirm the concept that human dental growth is affected by sex chromosomes. Amelogenesis is promoted by both X and Y chromosome genes, and sexual dimorphism in average tooth size is evidently determined by
