The release of intracellular peroxidase (EPO) was investigated in order to evaluate rat eosinophil activation by various immunoglobulin (Ig) isotypes. After successive incubations with purified rat IgG1, IgG2a, IgG2b, IgG2c, IgE, or IgM and their respective anti-Ig antisera, eosinophils released significant amounts of EPO (up to 26% of the intracellular content) only in the case of Ig with anaphylactic activities (IgG2a and IgE). The transport of horseradish peroxidase (HRP) from the nasal cavity to the olfactory bulb was examined in rat. HRP was present primarily in the olfactory nerve and glomerular layer. In some animals the glomeruli were densely filled with product while in others there was considerable interglomerular variation in density. A significant inactivation of red blood cell glutathione peroxidase (25% less than the physiological value) was observed after exposure of intact erythrocytes to 2 mM divicine (an autoxidizable aminophenol from Vicia faba seeds) and 2 mM ascorbate for 3 h at 37 degrees C. Neurological endemic cretinism is highly prevalent in severe endemic goiter areas. Often associated to euthyroid goiter, it is probably related to iodine deficiency. Reactions of ferric horseradish peroxidase with hydrogen cyanide and hydrogen peroxide were studied as a function of pressure. Activation volumes are small and differ in sign (delta V = 1.7 +/- 0.5 ml/mol for peroxidase + HCN and -1.5 +/- 0.5 ml/mol for peroxidase + H2O2). The temperature dependence of cyanide binding to horseradish peroxidase was also determined. A comparison is made of relevant parameters for cyanide binding and compound I formation.

The sources of innervation of the internal structures of the eye in monkey and rabbit were investigated by injecting horseradish peroxidase (HRP) intraocularly and monitoring its retrograde transport. Conditions were sought to increase the yield of HCN from L-histidine incubated with L-amino acid oxidase (L-amino acid:oxygen oxidoreductase (deaminating), EC 1.4.3.2) from snake venom, and horseradish peroxidase (donor:hydrogen-peroxide oxidoreductase, EC 1.11.1.7).
