Impact of Photoreceptor Loss on Retinal Circuitry.

Our sense of sight relies on photoreceptors, which transduce photons into the nervous system's electrochemical interpretation of the visual world. These precious photoreceptors can be disrupted by disease, injury, and aging. Once photoreceptors start to die, but before blindness occurs, the remaining retinal circuitry can withstand, mask, or exacerbate the photoreceptor deficit and potentially be receptive to newfound therapies for vision restoration. To maximize the retina's receptivity to therapy, one must understand the conditions that influence the state of the remaining retina. In this re

Multimodal high-resolution retinal imaging using a camera-based DMD-integrated adaptive optics flood-illumination ophthalmoscope.

We demonstrate the feasibility of a multimodal adaptive optics flood-illumination ophthalmoscope, able to provide both bright-field and dark-field images (such as phase contrast). The multimodality was made possible by integrating a digital micromirror device (DMD) at the illumination path to project a sequence of complementary high-resolution patterns into the retina. Through a versatile post-processing method that digitally selects backscattered or multiply scattered photons, we were able: (1) to achieve up to four-fold contrast increase of bright-field images when imaging the photoreceptor

Physiological and pathological roles of FATP-mediated lipid droplets in Drosophila and mice retina.

Increasing evidence suggests that dysregulation of lipid metabolism is associated with neurodegeneration in retinal diseases such as age-rel … an FATP1 was non-toxic and promoted mitochondrial energy metabolism in RPCs and non-autonomously in photoreceptor cells. In contrast, the inhibition of LD accumulation by dFatp knockdown suppressed neurodegeneration in Aats-metFB Drosophila mutants, which carry elevated levels of reactive oxygen species (ROS). This suggests that abnormal turnover of LD may be toxic f …

An allosteric interleukin-1 receptor modulator mitigates inflammation and photoreceptor toxicity in a model of retinal degeneration.

BACKGROUND: Inflammation and particularly interleukin-1β (IL-1β), a pro-inflammatory cytokine highly secreted by activated immune cells during early AMD pathological events, contribute significantly to retinal neurodegeneration. Here, we identify specific cell types that generate IL-1β and harbor the IL-1 receptor (IL-1R) and pharmacologically validate IL-1β's contribution to neuro-retinal degeneration using the IL-1R allosteric modulator composed of the amino acid sequence rytvela (as well as the orthosteric antagonist, Kineret) in a model of blue light-induced retinal degeneration. METHODS:

The versatile roles of retinal pigment epithelium in the pathophysiology of retinitis pigmentosa.

Retinitis pigmentosa (RP) is a group of hereditary retinal diseases that lead to progressive vision loss, with most disease-causing genes expressed in rod photoreceptors and a smaller fraction in retinal pigment epithelium (RPE) cells. The RPE and photoreceptor cells share a symbiotic relationship characterized by close spatial and functional interactions that play a pivotal role in vision. Although the role of RPE is fundamental to the retina, its involvement in retinal pathogenesis, and, in particular, in RP remains underappreciated. In this review, we summarize morphological alterations in …

Evaluating Human Rod Photoreceptor Function Using Pixelwise Intensity-Based Optoretinography.

PURPOSE: More than 50 inherited retinal diseases are known, with rod photoreceptor functional loss serving as an early indicator in nearly half of them. The "optoretinogram" is a relatively new assay that detects optical changes in cells in response to stimuli. This tool has excellent potential for providing insights into the earliest functional changes of rod photoreceptors, with the potential to assist in the early detection, monitoring, and treatment of retinal diseases. METHODS: In this work, we obtained intensity-based optoretinograms (iORGs) from rod photoreceptors using an adaptive opt

Ordered Chaos: Photoreceptor Mosaics in the Larval and Adult Turquoise Killifish (Nothobranchius furzeri).

An organism's vision is dependent on the photoreceptor types in the eye and how these cells are organized across the retina. The turquoise killifish (Nothobranchius furzeri) is an annual killifish species that lives in the turbid waters of ephemeral pools. Although turquoise killifish are gaining popularity as a model, we know little about their photoreceptors. Here, the turquoise killifish retina was investigated for photoreceptor subtype identity and mosaic organization at the adult and larval stages to assess specialization for their light environment. Turquoise killifish were determined to

Light and electron microscopic observations on retinal neurons of red-tail shark (Epalzeorhynchos bicolor H. M. Smith, 1931).

The structure of photoreceptors (PR) and the arrangement of neurons in the retina of red-tail shark were investigated using light and electron microscopy. The PR showed a mosaic arrangement and included double cones, single cones (SC), and single rods. Most cones occur as SC. The ratio between the number of cones and rods was 3:1.39 (±0.29). The rods were tall that reached the pigmented epithelium. The outer plexiform layer (OPL) showed a complex synaptic connection between the horizontal and photoreceptor terminals that were surrounded by Müller cell processes. Electron microscopy showed that

Characterization of zebrafish rod and cone photoresponses.

Zebrafish is a popular species widely used in vision research. The zebrafish retina has one rod and four cone subtypes (UV-, blue-, green-, and red-sensitive) with 40%-rod 60%-cone ratio, making it suitable for comparable studies of rods and cones in health and disease. However, the basic photoresponse properties of the four zebrafish cone subtypes have not been described yet. Here, we established a method for collecting flash photoresponses from zebrafish rods and cones by recording membrane current with a suction electrode. Photoreceptor subtypes could be distinguished based on their charact

Eye features and retinal photoreceptors of the nocturnal aardvark (Orycteropus afer, Tubulidentata).

The nocturnal aardvark Orycteropus afer is the only extant species in the mammalian order Tubulidentata. Previous studies have claimed that it has an all-rod retina. In the retina of one aardvark, we found rod densities ranging from 124,000/mm² in peripheral retina to 214,000/mm² in central retina; the retina of another aardvark had 163,000 - 245,000 rods/mm². This is moderate in comparison to other nocturnal mammals. With opsin immunolabelling we found that the aardvark also has a small population of cone photoreceptors. Cone densities ranged from about 300 to 1,300/mm² in one animal, and fro

Differential composition of DHA and very-long-chain PUFAs in rod and cone photoreceptors.

Long-chain PUFAs (LC-PUFAs; C20-C22; e.g., DHA and arachidonic acid) are highly enriched in vertebrate retina, where they are elongated to very-long-chain PUFAs (VLC-PUFAs; C 28) by the elongation of very-long-chain fatty acids-4 (ELOVL4) enzyme. These fatty acids play essential roles in modulating neuronal function and health. The relevance of different lipid requirements in rods and cones to disease processes, such as age-related macular degeneration, however, remains unclear. To better understand the role of LC-PUFAs and VLC-PUFAs in the retina, we investigated the lipid compositions of wh

Neural Repertoire Behind the World's Most Complex Retina: Neuroanatomy of the Stomatopod Lamina.

The lamina is the first optic neuropil and visual information integration station in crustaceans and insects, containing synaptic connections between photoreceptors and first-order interneurons. The lamina circuitry in mantis shrimp (stomatopods) is both interesting and complex, as there are 16 different types of photoreceptors contained within the mid-band region of the eye. Using serial block-face scanning electron microscopy, we have reconstructed photoreceptor terminals and lamina interneurons in two major superfamilies with different visual ecologies. Neurons follow the same general patte

The role of neurotrophic factors in retinal ganglion cell resiliency.

Many retinal diseases are characterized by direct or indirect retinal ganglion cell (RGC) neurodegeneration. In glaucoma and optic nerve neuropathies, RGCs are the primary affected cells, whereas in photoreceptor dystrophies, RGC loss is secondary to the death of rods and cones. The death of RGCs in either case will irreversibly cause loss of vision, as RGCs are the sole output neurons of the retina. RGC neurodegeneration affects certain neurons preferentially, resulting in subpopulations of resilient and susceptible cells. Neurotrophins (NTs) are known to mediate neuronal survival through the

Importance of Autoimmune Responses in Progression of Retinal Degeneration Initiated by Gene Mutations.

Inherited retinal diseases (IRDs) are clinically and genetically heterogeneous rare disorders associated with retinal dysfunction and death of retinal photoreceptor cells, leading to blindness. Among the most frequent and severe forms of those retinopathies is retinitis pigmentosa (RP) that affects 1:4,000 individuals worldwide. The genes that have been implicated in RP are associated with the proteins present in photoreceptor cells or retinal pigment epithelium (RPE). Asymmetric presentation or sudden progression in retinal disease suggests that a gene mutation alone might not be responsible …

Retinal dysplasia in mice lacking p56lck.

The product of the proto-oncogene p56lck is a non-receptor tyrosine kinase member of the Src family. It is found in T cells (Marth et al., 1985, 1988) and in the mouse brain (Omri et al., 1996; Van Tan et al., 1996). In this report, we describe experiments showing that Lck is present in the mouse retina neurons. Lck gene expression was identified after isolating and sequencing the specific 5' and 3' part of the cDNA obtained by RT-PCR. In adult retina Lck immunoreactivity was most abundant in photoreceptor cells and within the outer plexiform layers. Staining was also observed in the inner nuc…

Comparative transcriptomic insights into the evolution of vertebrate photoreceptor types.

To explore the molecular similarities and potential evolutionary origins of vertebrate photoreceptor types, we analyzed single-cell and -nucleus transcriptomic atlases from six vertebrate species: zebrafish, chicken, lizard, opossum, ground squirrel, and human. Comparative analyses identified conserved transcriptional signatures for the five ancestral photoreceptor types: red, blue, green, and UV cones, as well as rods. We further identified and validated molecular markers of the principal and accessory members of the tetrapod double cone. Comparative transcriptomics suggests that the principa

Physiological studies of the interaction between opsin and chromophore in rod and cone visual pigments.

The visual pigment in vertebrate photoreceptors is a G protein-coupled receptor that consists of a protein, opsin, covalently attached to a chromophore, 11-cis-retinal. Activation of the visual pigment by light triggers a transduction cascade that produces experimentally measurable electrical responses in photoreceptors. The interactions between opsin and chromophore can be investigated with electrophysiologial recordings in intact amphibian and mouse rod and cone photoreceptor cells. Here we describe methods for substituting the native chromophore with various chromophore analogs to investiga…

Mathematical analysis of photoreceptor changes in conditions of separation from the underlying retinal pigment epithelium.

Photoreceptors (PR) are responsible for absorbing and converting light into electrical signals necessary to create vision. To accomplish that, they are in an intimate relationship and attached to the underlying retinal pigment epithelium (RPE). Separation of the PR from the underlying RPE is seen in many retina disorders and leads to PR cell death and subsequent vision loss. Separation of PR from the RPE (detachment) interferes with the normal phagocytosis and recycling of PR outer segments by the RPE, and disrupts nutrient and metabolite delivery, including glucose. All these factors contribu

Unique ultrastructural organization of human rod photoreceptors.

Rod and cone photoreceptor cells are specialized neurons responsible for transforming the information reaching the eyes in the form of photons into the language of neuronal activity. Rods are the most prevalent photoreceptor type, primarily responsible for light detection under conditions of limited illumination. Here we demonstrate that human rods have a morphological organization unique among all described species, whereby the cell soma extends alongside the light-sensitive outer segment compartment to form a structure we have termed the "accessory inner segment." These structures have two s…
