Nitric oxide-mediated suppression of 2,3-bisphosphoglycerate synthesis: therapeutic relevance for environmental hypoxia and sickle cell disease.

Though hemoglobin (Hb) is best known for transporting oxygen and metabolic wastes throughout the circulatory system, this erythrocyte protein also acts as a hypoxic sensor, its oxygen saturation dependent on the oxygen partial pressure (pO(2)) which varies throughout the vasculature. The production and transport of the endogenous vasodilator nitric oxide (NO) by Hb is dependent on Hb's oxygen saturation, thereby allowing the protein to auto-regulate blood flow efficiency to meet the relative demands of respiring tissues. Erythrocyte concentrations of 2,3-bisphosphoglycerate (BPG), an enhancer

Stent thrombosis in the era of drug eluting stents.

Despite a marked reduction in restenosis and the need for repeat revascularization procedures with the use of drug-eluting stents (DES), the risk for stent thrombosis remains of serious concern. Although the safety profiles of DES dose not seem to differ from those of bare metal stent (BMS) in the acute and subacute phases following coronary intervention, recent data suggest a potential increase of thrombotic events late after DES deployment. The main factors associated with late stent thrombosis remain elusive. Delayed re-endothelialization, hypersensitivity reaction, technical and mechanical

Expression of the p53 homologue p63alpha and DeltaNp63alpha in the neoplastic sequence of Barrett's oesophagus: correlation with morphology and p53 protein.

BACKGROUND: While loss of p53 function is a key oncogenic step in human tumorigenesis, mutations of p53 are generally viewed as late events in the metaplasia-dysplasia-adenocarcinoma sequence of Barrett's oesophagus. Recent reports of a series of genes (p63, p73, and others) exhibiting close homology to p53 raise the possibility that abnormalities of these p53 family members may exert their influence earlier in the sequence. AIM: Following recent characterisation of expression of p63 and a major isoform DeltaNp63 by generation of an antiserum that recognises p63 isoforms, but not p53, our aim

Simulated kangaroo care in very preterm infants does not reduce physiological instability: the COSYBABY randomised controlled cross-over trial.

INTRODUCTION: Infants who are born very preterm experience frequent episodes of physiological instability including apnoea, oxygen desaturation and bradycardia due to immaturity of the pulmonary and nervous systems. Parental contact, such as kangaroo care, may reduce physiological instability. However, there may be long periods when parents cannot be with their baby. The BABYBE SYSTEM® is a medical device designed to simulate kangaroo care. METHODS: We conducted a randomised cross-over trial to determine whether episodes of apnoea and other episodes of physiological instability were reduced w

Differences in physiological indicators between twin and singleton pregnancy.

OBJECTIVE: It has been widely reported that there are physiological differences between twin and singleton pregnancies, as evidenced by physiological indicators examined during prenatal period. However, not all physiological indicators have been evaluated. Physiological indicators of twin pregnancies still refer to the standards of single pregnancies and lack precise variation intervals. Therefore, our study compared and analyzed the differences in physiological indicators between twin and singleton pregnancies. METHODS: Retrospective cohort data of deliveries at Peking University Third Hospi

From elective protocols to emergency reality: rethinking adipose tissue harvesting for stromal vascular fraction applications.

BACKGROUND: Adipose tissue is a readily accessible source of stromal vascular fraction (SVF) and adipose-derived stem/stromal cells, widely used in regenerative medicine. Current harvesting and processing standards are derived almost exclusively from elective surgical settings performed under controlled physiological conditions. However, expanding applications in trauma and neurosurgery increasingly require adipose tissue procurement in emergency settings, where systemic inflammation, hypoperfusion, and time constraints may alter tissue biology. The biological impact of emergency harvesting re

Dissecting the host transcriptome and microbiota responses to viral hemorrhagic septicemia virus (VHSV) in the turbot (Scophthalmus maximus) intestine: insights from moribund and asymptomatic phenotypes.

Viral infections profoundly influence host physiology, not only by triggering complex immune responses but also by reshaping the associated microbiota. Conversely, the microbiota can modulate antiviral defenses and disease outcomes through its interactions with the host immune system. This dynamic triad -virus, microbiota, and host immunity-has emerged as a central determinant of infection severity and recovery. Yet, despite growing evidence from mammalian models, its functional implications in fish remain poorly understood. Here, we investigated the effects of viral hemorrhagic septicemia vir

Laparoscopic Ovum Pick-up For In Vivo Oocyte Retrieval In Alpacas.

Alpacas are increasingly incorporated into reproductive and biomedical research due to distinctive physiological and immunological characteristics that extend their relevance beyond agricultural applications. As camelids, alpacas exhibit induced ovulation, prolonged follicular dynamics, asymmetric uterine physiology, and a unique immune system characterized by heavy-chain-only antibodies. These features support their use in comparative and translational research, particularly in reproductive biology and genetic engineering. Laparoscopic ovum pick-up (LOPU) is a minimally invasive approach for

From primates to people: Mapping host-microbiome-health relationships in aging.

The human microbiome profoundly influences host physiology, metabolism, and immune function. A balanced microbial ecosystem supports immune homeostasis and health, whereas dysbiosis contributes to diverse diseases, including metabolic, autoimmune, neurodegenerative, and infectious disorders. Aging, the greatest risk factor for chronic disease, is characterized by hallmarks such as mitochondrial dysfunction, genomic instability, and "inflammaging," a state of chronic low-grade inflammation. Emerging evidence links age-associated microbial dysbiosis to inflammaging and systemic decline, as micro

The heart responds to pressure via the lung: Dive depth alters cardiac control in bottlenose dolphins.

Marine mammals face the dual challenge of foraging underwater while relying on limited oxygen stores, making efficient oxygen management essential for survival. The dive response, which includes bradycardia and peripheral vasoconstriction, helps conserve oxygen for critical organs, such as the heart and brain. While the heart rate (HR) is known to decline with dive duration, the independent effect of dive depth, often confounded by longer dives, remains unclear. To isolate this effect, we measured HR and heart rate variability (HRV) in seven bottlenose dolphins (Tursiops truncatus) during volu

Cadmium exposure triggers adaptive physiological and molecular responses that undermine development in Bactrocera dorsalis.

The oriental fruit fly, Bactrocera dorsalis (Diptera: Tephritidae), an important invasive fruit and vegetable pest, exhibits a broad host range that includes citrus, mango, and banana. Its larvae bore into fruit tissues, causing severe economic losses. Cadmium (Cd²⁺) is classified among the most toxic heavy metal contaminants, capable of accumulating in plants and biomagnifying through food chains, thereby threatening biodiversity and human health. This study assessed the effects of Cd²⁺ stress on B. dorsalis larval growth and development, alongside changes in key enzymatic activities and tran

Pleiotropic roles of the MATE transporter CD20030 in Clostridioides difficile: linking multidrug resistance to oxidative stress defense and virulence regulation.

BACKGROUND: The multidrug-resistant pathogen Clostridioides difficile (C. difficile) presents a persistent clinical threat. While Multidrug and Toxic Compound Extrusion (MATE) transporters are recognized as xenobiotic efflux pumps, their pleiotropic roles in pathogen physiology, particularly in stress adaptation and virulence regulation, remain largely unexplored. Understanding how C. difficile adapts and thrives in the face of host defenses and antimicrobial pressures, potentially influencing gut microbiome dynamics, is crucial for combating C. difficile infection. METHODS: We functionally c

Exploring the evolutionary divergence of cyclic di-nucleotide signaling in diverse mycobacterial species.

Cyclic dinucleotides (CDNs), such as cyclic di-AMP (c-di-AMP) and cyclic di-GMP (c-di-GMP), are key second messengers that regulate fundamental bacterial processes, including cell wall synthesis, biofilm formation, antibiotic resistance, stress response, and virulence. These pathways are particularly relevant in major pathogens like Mycobacterium tuberculosis. Increasing evidence highlights the pathogenic potential of non-tuberculous mycobacteria (NTM), originally environmental species that are emerging as significant human pathogens. Understanding the evolution of CDN signaling may therefore
