Theta-toxin of Clostridium perfringens. I. Purification and some properties.

Theta-Toxin, an oxygen-labile hemolysin produced by Clostridium perfringens, was purified 3300 fold from culture filtrate by successive chromatography on DEAE-Sephadex A-50 and Sephadex G-150. The purified toxin gave two distinct bands in disc electrophoresis, while the same material, after mild reduction with dithiothreitol, yielded a single band, indicating that the purified theta-toxin contained, as well as a reduced, active form, an oxidized, inactive form of toxin. These two forms of the toxin had a similar, if not identical molecular size. The purified preparation gave a single band in a

Prevalence of farmer's lung in Scotland: a pilot survey.

In a survey of the farming population of Orkney, Ayrshire, and East Lothian the prevalence of farmer's lung was estimated at 86 per 1,000 in both Orkney and Ayrshire and 23 per 1,000 in East Lothian. If cases with a negative farmer's lung hay (F.L.H.) precipitin test are excluded these figures are reduced to 43, 36, and nil respectively, but those for Orkney and Ayrshire are still about 20 times higher than any figure previously reported for the prevalence of farmer's lung in Britain.Regional variations in prevalence are probably related both to climatic conditions and to differences in agricu

Immunological determination of muscle-type enolase in the serum as a diagnostic test for myocardial infarction.

An antiserum raised in the chicken against purified rat skeletal muscle enolase neutralized 86% of the enolase activity of human heart muscle and 8.7% +/- 4.8% (mean +/- S.D.) of the enolase activity in normal human serum. The fraction of enolase inhibited by the antiserum, designated as 'immunologically reacting muscle enolase', promptly rose after myocardial infarction in the serum of affected subjects, reaching peak values in excess of 40% in all but one of 23 subjects. Only one subject with ischaemic heart disease, but without infarction, and 2 with miscellaneous conditions, among 117 such

New perspectives on coccidioidomycosis.

Coccidioidomycosis is a growing problem in the endemic regions of Arizona and California. The reasons for its increase are unclear, but may relate to an influx of nonsusceptible individuals into the endemic regions, construction and dust, and climate. Most cases of coccidioidal infection are completely asymptomatic, but approximately 40% present as a pulmonary process that may be difficult to distinguish from a bacterial community-acquired pneumonia. This has led to underdiagnosis of coccidioidomycosis and inappropriate antibacterial therapy. Serology has been the mainstay of diagnosis, but it

Assessment of four serological techniques in the immunological diagnosis of farmers' lung disease.

Farmers' lung disease (FLD) is a pulmonary disease that results from repeated inhalation of antigens from mouldy hay or straw. The objective of this prospective study was to assess the reliability of four serological techniques in FLD diagnosis. Sera from 15 consecutive patients with FLD, 15 healthy control farmers and 30 urban controls were analysed using four serological techniques [electrosyneresis (ES), Ouchterlony double diffusion (DD), ELISA and Western blot (WB)] with four antigens (Absidia corymbifera, Eurotium amstelodami, Wallemia sebi and Saccharopolyspora rectivirgula). In the auth

[Blood feeding sources and peridomiciliation of Lutzomyia longipalpis (Lutz & Neiva, 1912) (Psychodidae, Phlebotominae)].

A precipitin test was employed to study the alimentary tract content of Lutzomyia longipalpis in the intra- and peridomiciliary environments in the municipality of Raposa, Maranh o State, a transmission area for visceral leishmaniasis or kala azar. Out of 2,240 female sandflies captured, 547 (24.4%) had fed on vertebrate blood, with the following proportions: avian (87.9%); rodent (47.2%); human (42.4%); canine (27.6%); opossum (26.6%); and equine (22.5%). Based on a survey of 120 human dwellings, chickens were found to be the most common domestic animals in the peridomicile (28.3%), followed

Maternal group B streptococcal (GBS) genital tract colonization at term in women who have asymptomatic GBS bacteriuria.

OBJECTIVE: To determine the rate of positive group B streptococcus (GBS) cultures at 35-37 weeks gestation in women who have first trimester asymptomatic GBS bacteriuria. METHODS: Pregnant women with asymptomatic first trimester GBS bacteriuria had genital cultures for GBS performed at 35-37 weeks gestational age. Serotyping was performed by the standard Lancefield capillary precipitin method. RESULTS: Fifty-three women with positive urine cultures had genital cultures performed at 35-37 weeks. Sixteen of the 53 (30.2%; 95% confidence interval: 18.4-44.3%) third trimester vaginal cultures we

[Saprophytic and invasive pulmonary aspergillosis].

The characteristics of chest x-ray films in 19 patients with saprophytic pulmonary aspergillosis secondary to acid-fast bacilli infection were reported. The saprophytic form, exemplified by the classic fungus ball or mycetoma, results from the growth of fungal mycelia within a pre-existing area of destroyed lung, typically a pre-existing cavity such as that resulting from tuberculosis. Aspergillus fumigatus was detected in 52.6% of sputum cultures from 19 patients. Aspergillus precipitin test was positive in 68.4% of patients. Thickening of cavitary walls was first found in 85% of chest x-ray
