Cognition and Evolution of Movement Disorders of FOXG1-Related Syndrome.

FOXG1-related syndrome is a rare neurodevelopmental encephalopathy characterized by early onset hyperkinetic movement disorders, absent language, autistic features, epilepsy, and severe cognitive impairment. However, detailed evaluation of cognition and evolution of movement disorders over time have not been clearly described before. In this study, we performed whole-exome sequencing in a cohort with unknown severe encephalopathy and movement disorders, with/without autistic behaviors. We identified FOXG1 mutations in three patients. One of them had a novel mutation that has not been described

An updated literature review on maternal-fetal and reproductive disorders of Toxoplasma gondii infection.

BACKGROUND: Toxoplasma gondii infection is one of the most prevalent infectious disease with worldwide distribution. Congenital toxoplasmosis is annually responsible for 1.20 million disability-adjusted life years around the world, but often it is overlooked many countries. METHODS: We performed an updated review to summarize the current researches on fetal, neonatal and maternal consequences of T. gondii infection and also adverse effects of toxoplasmosis on women reproductive organs. RESULTS: T. gondii infection could be cause of several abnormalities from hydrocephalus, microcephaly, deaf

Interactions between the cannabinoid and dopaminergic systems: evidence from animal studies.

There is a prominent role of the cannabinoid system to control basal ganglia function, in respect to reward, psychomotor function and motor control. Cannabinoid dysregulations might have a pathogenetic role in dopamine- and basal ganglia related neuropsychiatric disorders, such as drug addiction, psychosis, Parkinson's disease and Huntington's disease. This review highlights interactions between cannabinoids, and dopamine, to modulate neurotransmitter release and synaptic plasticity in the context of drug addiction, psychosis and cognition. Modulating endocannabinoid function, as a plasticity

Resurrecting melancholia.

OBJECTIVE: To define melancholia as a distinct mood disorder, identified by unremitting depressed mood, vegetative dysfunction, and psychomotor disturbances, verifiable by neuroendocrine tests, and treatable by electroconvulsive therapy and tricyclic antidepressants. METHOD: A review of the literature of two centuries finds descriptions of severe mood disorders, either depression or mania or circular, defined as 'melancholia.' In the 1980 diagnostic revision (DSM-III), melancholia was relegated to a features specifier only. RESULTS: DSM classification criteria develop heterogeneous patient s

Patient with rheumatoid arthritis and MCA/MR syndrome due to unbalanced der(18) transmission of a paternal translocation t(18;20)(p11.1;p11.1).

A girl with psychomotor retardation and a pattern of minor anomalies was found to have a slightly enlarged short arm of chromosome 18 by conventional GTG-banded chromosome examination. The 18p+chromosome has also been found in the father. FISH studies using chromosome 18-and chromosome 20-specific painting probes confirmed a reciprocal whole arm translocation between chromosomes 18 and 20 in the father, resulting in monosomy of the short arm of chromosome 18 and trisomy of the short arm chromosome 20 in the patient. FISH analysis using a chromosome 18 alpha-satellite-specific probe showed a re

An overactive mind: Investigating racing thoughts in ADHD, hypomania and comorbid ADHD and bipolar disorder via verbal fluency tasks.

BACKGROUND: Adults with Attention Deficit Hyperactivity Disorder (ADHD) often report mental restlessness akin to racing thoughts found in hypomanic and mixed episodes of bipolar disorder (BD). Past research has suggested that racing thoughts in BD can be tackled via process-oriented verbal fluency measures. In ADHD, it is still unknown whether racing thoughts is due to comorbid BD, and its neuropsychological underpinnings remain to be investigated. To this aim, this study investigates process-oriented verbal fluency measures in adults with ADHD compared to adults with BD in a hypomanic episode

Quetiapine withdrawal: A systematic review.

OBJECTIVE: Withdrawal from psychoactive medication such as quetiapine is a well-documented phenomenon. Despite the extensive use of quetiapine, there have been few studies into the presence of discontinuation symptoms. We therefore performed a systematic review of published literature for evidence of quetiapine withdrawal or symptoms associated with discontinuation. METHODS: We searched PubMed, Embase, CINAHL, Medline, Web of Science, PsycINFO for articles containing the terms 'Quetiapine' AND 'withdraw$' OR 'discontinue$'. We included all study types that reported on somatic withdrawal sympt

[Behavioural phenotypes. Biologically determined neuropsychological patterns].

INTRODUCTION AND AIMS: A behavioural phenotype (BP) is a characteristic pattern of motor, cognitive, linguistic and social abnormalities that are associated in a way that is compatible with a biological disorder, while environmental factors are also known to be important in their development. Taking these concepts into account, we have analyzed several entities with acknowledged BP, which were selected according to the frequency of presentation, the compatibility of the association between BP and the underlying disease, and the importance of recognizing the entity, so as to enable suitable the

Understanding and Managing Infantile PHGDH Deficiency: A Case Report.

Phosphoglycerate dehydrogenase deficiency is a rare neurometabolic disorder with clinical features of congenital microcephaly, psychomotor retardation, intractable seizures, and spasticity. We report a 2.5-year-old boy presenting with speech delay, seizures, microcephaly, and hyperactive behavior. Genetic testing detected a likely pathogenic homozygous variant c.1129G>A in the PHGDH gene. Parents were carrier for the detected variant. Biochemical analysis showed low serine and treatment with oral serine and glycine resulted in seizure control, followed by catchup of developmental milestones. T

[Kleine-Levin syndrome].

Kleine-Levin syndrome (KLS) is manifested by periodic hypersomnolence with an increased appetite and psychopathological symptoms (psychomotor agitation, hallucinations, hypersexuality). The duration of the attack ranges from several days to several weeks. A forced awakening can provoke severe aggressive behavior. The disease occurs for an unknown reason, mainly in adolescence and mostly affects males. Lack of awareness among doctors about the disease leads to an incorrect diagnosis, often of a psychopathological disorder. This article presents modern data on the causes, clinical symptoms, diag

[Serotonin syndrome secondary to the association of paroxetine and amitriptyline: a case report].

Serotonin syndrome is an iatrogenic disorder induced by serotonergic drug. It is characterised by a symptomatic polymorphism that may create confusion with other clinical conditions, delaying diagnosis and putting the vital prognosis at risk. In light of a case report and a literature review, we here report the clinical case of a patient presenting with confusion, psychomotor agitation, hyperreflexia and spontaneous myoclonus following simultaneous use of paroxetine and amitriptyline. The diagnosis of serotonin syndrome was made after ruling out other possible diagnoses. The patient was admitt

A novel IRF2BPL truncating variant is associated with endolysosomal storage.

De novo mutations in the IRF2BPL gene have been identified to date in 18 patients presenting with neuromotor regression, epilepsy and variable neurological signs. Here, we report a female child carrying a novel heterozygous truncating variant in IRF2BPL. Following normal development for two and half years, she developed a progressive neurological condition with psychomotor regression, dystonic tetraparesis with hyperkinetic movements, but no overt epilepsy. Skin biopsy revealed enlarged lysosomes containing granular and tubular material, suggestive of a lysosomal storage disorder. This case ex

Subtelomeric 6p monosomy and 12q trisomy in a patient with a 46,XX,der(6)t(6;12)(p25.3;q24.31) karyotype: Phenotypic overlap with Mutchinick syndrome.

We report on a patient with partial monosomy 6p and partial trisomy 12q identified by fluorescent in situ hybridization (FISH) and array-based comparative genomic hybridization (aCGH). She had a complex phenotype characterized by mental retardation (MR), psychomotor developmental delay, speech disorder, hypertelorism, eye anomalies, hearing loss, low-set malformed ears, thin upper lip, heart defect, clinodactyly, pes valgus, and skeletal anomalies. There is phenotypic overlap between our case and Mutchinick syndrome. This is the first report of a combined partial monosomy 6p and partial trisom

Mucolipidosis IV: ultrastructural diagnosis of a recently defined genetic disorder.

In nine cases of a new genetic disorder mucolipidosis IV clinical findings included early or congenital corneal cloudiness and mild to severe psychomotor retardation in all patients. The diagnosis can be established by electron microscopy of the conjunctiva, which along with several other tissues shows typical ultrastructural changes. All of the known patients described were descendants of parents of Jewish-Ashkenazi origin, and it seems that mucolipidosis IV is another "Jewish" genetic disease. Prenatal diagnosis can be made by finding the typical ultrastructural changes in cells of the amnio

Clinical and molecular characterization of patients with adenylosuccinate lyase deficiency.

BACKGROUND: Adenylosuccinate lyase deficiency (ADSLD) is an ultrarare neurometabolic recessive disorder caused by loss-of-function mutations in the ADSL gene. The disease is characterized by wide clinical variability. Here we provide an updated clinical profiling of the disorder and discuss genotype-phenotype correlations. RESULTS: Data were collected through "Our Journey with ADSL deficiency Association" by using a dedicated web survey filled-in by parents. Clinical and molecular data were collected from 18 patients (12 males, median age 10.9 years ± 7.3), from 13 unrelated families. The age

Major Depressive Disorder in a Patient with Systemic Lupus Erythematous, Pulmonary Hypertension, and Hypercoagulation: A Case Report.

Major depressive disorder is characterized by the presence of single or repeated major depressive episodes, which are considered periods of 2 weeks of depressive moods featuring impaired neurovegetative functioning, psychomotor activity, and cognition, as well as suicidal thoughts. Major depressive disorder is commonly associated with other medical conditions, especially chronic and systemic medical illnesses. Cardiovascular diseases are among the most related, especially pulmonary hypertension, a cardiovascular disorder that results in increased pulmonary circulation pressure--with an average

Protein Kinase C β: a New Target Therapy to Prevent the Long-Term Atypical Antipsychotic-Induced Weight Gain.

Antipsychotic drugs are currently used in clinical practice for a variety of mental disorders. Among them, clozapine is the most effective medication for treatment-resistant schizophrenia and is most helpful in controlling aggression and the suicidal behavior in schizophrenia and schizoaffective disorder. Although clozapine is associated with a low likelihood of extrapyramidal symptoms and other neurological side effects, it is well known for the weight gain and metabolic side effects, which expose the patient to a greater risk of cardiovascular disorders and premature death, as well as psycho
