Probiotic intervention mitigates radiation-induced intestinal injury by alleviating oxidative stress in a human gut-on-a-chip.

Gastrointestinal acute radiation syndrome (GI-ARS) poses a critical public health concern, necessitating the development of effective medical countermeasures (MCM). Here, we evaluated the therapeutic potential of a commercially available probiotic formulation using a human gut-on-a-chip model that mimics the physiodynamic intestinal microenvironment. Intestinal epithelial Caco-2 cells were subjected to 8 Gy gamma radiation, targeting the epithelial layer, the culture medium, or both. The irradiated epithelial cells challenged to the irradiated medium resulted in significant DNA damage quantifi

The impact of lactate and lipid metabolism in microglia upon cognitive impairment following radiation-induced brain injury.

BACKGROUND: Radiation-induced brain injury (RIBI) is a serious complication that occurs after cranial radiotherapy. The main manifestations are delayed radiation effects characterized by neuroinflammation and damage to neural stem cell populations. Microglia, the resident immune cells of the central nervous system (CNS), have become key mediators in the pathological process of RIBI. This review aims to systematically elucidate how metabolic reprogramming of lactate and lipid pathways in microglia contributes to chronic neuroinflammation and cognitive impairment following RIBI, and to evaluate

A Huaxian Formula Nanoformulation Confers Protection Against Acute Radiation-Induced Lung Injury by Targeting Ferroptosis.

Acute radiation-induced lung injury (RILI) remains a major dose-limiting complication of thoracic radiotherapy with limited therapeutic options. Here, we develop an inhalable nanoformulation of the traditional Chinese medicine Huaxian Formula (NHXF) with favorable physicochemical stability and enhanced pulmonary delivery properties. In vitro and in vivo studies demonstrate that NHXF attenuates radiation-induced alveolar epithelial injury, accompanied by reductions in oxidative stress and apoptosis. NHXF also suppresses secondary inflammatory responses, as evidenced by decreased pro-inflammator

Nplate (Romiplostim) Restores Blood Counts and Accelerates Wound Healing Following Radiation Combined Injury in Mice.

BACKGROUND: Exposure to ionizing radiation (IR), particularly when combined with additional trauma such as skin wounding (referred to as radiation combined injury, RCI), results in a synergistic pathology that significantly increases morbidity and mortality compared to isolated radiation injury (RI). While Nplate (romiplostim), a thrombopoietin (TPO) receptor agonist, is a Food and Drug Administration (FDA)-approved medical countermeasure for haematopoietic acute radiation syndrome (H-ARS), its efficacy on RCI is unclear. This study evaluated the mitigative effects of Nplate on 30-day survival

A human iPSC-derived 3D spinal cord organoid model to study radiation-induced neural injury.

BACKGROUND: Radiation therapy remains a cornerstone in the treatment of primary and metastatic tumors; however, its efficacy is limited by the spinal cord's high sensitivity to radiation-induced injury. To better understand the mechanisms underlying spinal cord radiosensitivity, we developed a three-dimensional human spinal cord organoid model derived from human-induced pluripotent stem cells using a neuroectodermal differentiation protocol that closely mimics embryonic spinal cord development. METHODS: Mature spinal cord organoids were exposed to a clinically relevant 2 Gy dose of ionizing r

Characterization of a murine tail model for radiation-induced skin injury following localized irradiation at graded doses.

PURPOSE: Radiation-induced skin injury (RISI) is common in both radiation therapy and accidental exposure. An appropriate animal model of RISI is of great significance for understanding its injury mechanism and developing medical countermeasures (MCMs). A reproducible, dose-dependent murine model of RISI was established via localized irradiation of the tail skin with graded doses. MATERIALS AND METHODS: C57BL/6 mice were employed to establish RISI model by irradiating 2 cm section of the mouse tail with 20 Gy, 30 Gy, and 40 Gy of single irradiation with RS2000 Biological Irradiator. Skin inju

Assessment Tools of Radiation Skin Injury in Head and Neck Cancer: A Narrative Review of Preclinical and Clinical Studies.

Radiation skin injury (RSI) is an unavoidable side effect of radiotherapy that delays the treatment process and affects patients' quality of life. However, RSI is easily overlooked in clinical practice with unified assessment methods. Although over 20 types of clinical scales have already been reported, they were never collated and systematically reorganized. In this review, we collected and categorized the RSI scales from the perspective of RSI-rated symptoms, patient self-feeling, the impact on quality of life, and psychological conditions, and summarize the common assessment modalities (bot

Serum Pro-N-Cadherin Correlates With Cardiac Injury in the Radiation Late Effects Cohort of Nonhuman Primates.

BACKGROUND: The delayed effects of radiation exposure on the heart often manifest as cardiac fibrosis and diastolic dysfunction, which can develop years after exposure. However, no Food and Drug Administration-approved serological biomarker is available to assess an individual's risk of developing radiation-related heart disease (RRHD). OBJECTIVES: The authors hypothesize that the risk of RRHD can be assessed using serum pro-N-cadherin (PNC), a promising marker for predicting subclinical heart failure in the general population. METHODS: We examined 46 male nonhuman primates (NHPs) that survi

Circulating Cell-free DNA as a biomarker for radiation-induced injury: From mechanistic insights to clinical translation.

Circulating cell-free DNA (cfDNA) has attracted increasing attention as a minimally invasive biomarker for assessing radiation-induced tissue injury. Radiation exposure can alter cfDNA concentration, fragmentation profiles, and methylation patterns. These changes may reflect cellular damage, tissue origin, and temporal responses to irradiation. During radiotherapy, however, cfDNA can arise from tumor cells, irradiated normal tissues, endothelial cells, immune cells, and hematopoietic cells. This mixed origin complicates the interpretation of radiation-related signals. Current evidence is deriv

Prior exposure to microplastics heightened the susceptibility of small intestine to radiation-induced injury in C57BL/6 mice.

Microplastics (MPs) have been detected in multiple human organs, raising concerns about their potential health risks. The intestinal tract is particularly vulnerable to MPs exposure and accumulation. Radiotherapy often causes side effects such as radiation-induced intestinal injury (RIII). Although previous studies have shown that MPs exacerbate RIII by altering gut microbiota, their effect on the small intestine's intrinsic sensitivity to radiation remains unclear. In this study, C57BL/6 mice were preexposed to MPs for a short period and then irradiated with 4 or 10 Gy to evaluate intestinal

Repetitive transcranial magnetic stimulation alleviates radiation-induced brain injury in rats: involving the inhibition of ferroptosis.

Radiation-induced brain injury (RIBI) is a prevalent complication following radiotherapy for head and neck tumors, and its effective therapeutic strategies are lacking. Ferroptosis, an iron-dependent cell death, has recently emerged as an important mechanism of radiation-induced cell death. Repetitive transcranial magnetic stimulation (rTMS) is a noninvasive neuro-interventional technique with antioxidant and neuroprotective properties. The present study aimed to explore whether rTMS attenuates RIBI by inhibiting ferroptosis. We demonstrated that rTMS alleviated neuronal injury and death in th

Dihydroartemisinin Attenuates Radiation-Induced Lung Injury by Inhibiting the cGAS/STING/NF-κB Signaling Pathway.

Dihydroartemisinin (DHA) is a derivative of artemisinin, which affects inflammation, oxidative stress, and immune regulation. However, the mechanism underlying its effects remains largely unknown. This study aims to explore the mechanism by which DHA affects radiation-induced lung injury (RILI), providing new insights for lung radiotherapy. To elucidate its mechanism of action, C57BL/6 J mice were irradiated with 15 Gy whole chest. RILI was evaluated by qRT-PCR, ELISA, histology, Western blot analysis, immunohistochemistry, and RILI signaling cascade studies. In addition, small interfering RNA

Raspberry ketone alleviates radiation-induced lung injury through the STAT2-P2X7r/NLRP3 signaling pathway.

BACKGROUND: Radiation-induced lung injury (RILI) is a frequent side effect in patients with breast cancer receiving radiotherapy. Currently, there is no optimal radioprotective agent that is widely utilized for RILI treatment. Raspberry ketone (RK) is a natural aromatic compound found in raspberries (Rubus idaeus L.). A recent study by our research revealed that RK has strong anti-inflammatory and anti-fibrosis effects. However, its actual mechanism of action in inflammation-related lung injury remains elusive. PURPOSE: This study aimed to clarify whether RK can alleviate RILI and determine t

Folic Acid Protects Against Radiation-Induced Lung Injury by Suppressing Senescence of Lung Epithelial Cells.

Radiation-induced lung injury (RILI) is a prevalent complication of thoracic tumor radiotherapy, severely compromising treatment efficacy and the patients' quality of life, yet effective prevention or treatment strategies remain elusive. Folic acid (FA), a water-soluble vitamin, plays critical roles in DNA synthesis/repair, cell cycle regulation, epigenetic regulation via methylation, oxidative stress response, and embryonic development. However, its radioprotective role has not been systematically elucidated. This study aimed to investigate its effects and molecular mechanisms during RILI. Th

Inhalable Nano-Astaxanthin for Radiation-Induced Lung Injury via Enhanced Lung Retention and Inflammation Suppression.

Radiation-induced lung injury (RILI) is a major complication of clinical radiotherapy, characterized by oxidative stress, inflammation, and progressive fibrosis. Astaxanthin (ASX), a potent natural antioxidant, can effectively scavenge radiation-induced reactive oxygen species; however, its therapeutic potential is limited by its poor solubility and bioavailability. Here, we develop inhalable chitosan-modified ASX-loaded PLGA nanoparticles (ASX@P@CS) to prevent RILI. This nanoformulation markedly improves the solubility of ASX, while the cationic chitosan modification enables mucin binding and

Trichostatin A ameliorated combined radiation and skin wound injury-induced mortality and hematopoietic suppression in a rat model.

PURPOSE: Effective countermeasure modalities against combined radiation injury, where the consequences of radiation exposure are highly complex with one or more conditions like burns, wounds, or infection, are yet far from reality. The current study aims to evaluate Trichostatin A (TSA), a presumptive radiomitigative molecule, in a rat model of combined radiation and wound injury (CRWI). METHODS: Sprague-Dawley rats (200 ± 10 g) were whole-body exposed to different doses of 60Co-γ-irradiation (4-7Gy) alone and with an excision wound (EW, size 20 ± 2 mm diameter), to optimize CRWI radiation do

Recent advances in the mechanisms, current treatment status, and application of multifunctional biomaterials for radiation-induced skin injury.

Radiation-induced skin injury (RISI) is a prevalent complication following nuclear accidents and radiotherapy for tumors. The associated side effects may include erythema, desquamation, ulceration, and in severe cases, necrosis of certain skin tissues. These adverse reactions significantly impact the quality of life for patients and contribute to both psychological distress and economic burdens. However, there is currently no standardized protocol for the treatment and management of RISI. In comparison to traditional pharmaceuticals, the utilization of biomaterials in addressing radiation-indu
