[A case of primary reticulum cell sarcoma of the brain with uveitis (author's transl)].

In 1972, Neault and his co-workers reported seven cases of uveitis associated with intracranial reticulum cell sarcoma. Recently we have experienced a similar case for the first time in Japan. A 32-year-old woman registered on March 10, 1971, complaining of blurred vision in the left eye for about two weeks. By ophthalmic examination, left posterior uveitis was diagnosed but the etiology was unknown. Treatment with corticosteroids was begun, but her left eye continued to fail in spite of the treatment. In August, 1971, she complained of weakness of left arm and leg, and in October, she suffere…

Identification of endoplasmic reticulum stress-related biomarkers in asthma and asthma exacerbation.

BACKGROUND: Endoplasmic reticulum (ER) stress plays a crucial role in airway inflammation and other asthma-related processes. This study aims to identify potential ER stress-related biomarkers in asthma. METHODS: Sputum transcriptome data (GSE76262) from 118 moderate-to-severe asthma patients and 21 healthy controls were analyzed to identify ER stress-related differentially expressed genes (DEGs). Functional enrichment, weighted gene co-expression network analysis (WGCNA), and protein-protein interaction (PPI) network construction were performed. Machine learning algorithms (LASSO logistic re…

Mechanism of endoplasmic reticulum stress: Insights for potential therapeutic benefits in burns.

Burns are injuries to the skin and deep tissues caused by thermal effects, which can trigger inflammation, necrosis, and systemic pathological reactions, often leading to life-threatening complications such as shock, infection, and multi-organ failure. Endoplasmic reticulum stress (ERS) is a key driving factor for tissue damage after burns. Burns are proposed to induce ERS through multiple mechanisms, including protein denaturation, inflammatory cytokine storms, and metabolic disturbances, Under stress conditions, cells can activate three key sensors, IRE1, PERK, and ATF6, to fully activate th…

MYOM1 deficiency leads to dilated cardiomyopathy by disrupting the homeostasis of sarcoplasmic reticulum.

MYOM1, a major component of the vertebrate myofibrillar M band, binds myosin, titin, and light meromyosin, has been linked to cardiomyopathy … cular level, transcriptomic and immunoblot analyses demonstrated downregulation of key sarcoplasmic reticulum regulators, including RYR2 and SERCA2. Consistent with these findings, calcium imaging documented impaired calcium conduction velocity and blunted intracellular calcium transients, indicating sarcoplasmic reticulum dysfunction. Our study provided si …

Sarcolipin is a Conformation-Dependent Regulator of the Sarcoplasmic Reticulum Calcium Pump SERCA.

Muscle relaxation is enabled by the sarco-endoplasmic reticulum Ca2+-ATPase (SERCA), which removes calcium from the cytosol and returns it to the lumen of the sarcoplasmic reticulum. During transport, SERCA adopts a variety of conformational states which differ in their structure and affinity for substrates. The transition between these states is mediated by calcium and ATP binding and the formation of an aspartyl-phosphate intermediate, which enables the transport of calcium across the membrane. SERCA function is highly regulated because of the importance of calcium in processes such as muscl…

Endoplasmic Reticulum Stress as a Stage-Dependent Regulatory Hub in Rheumatoid Arthritis.

Rheumatoid arthritis (RA) is a chronic autoimmune disease characterized by persistent synovial inflammation and progressive joint destruction. Beyond its canonical role in maintaining proteostasis, growing evidence indicates that endoplasmic reticulum (ER) stress acts as a dynamic regulatory hub integrating inflammatory signaling, metabolic reprogramming, and cell fate control within the RA synovium. Activation of the unfolded protein response (UPR) sensors-IRE1α, PERK, and ATF6-initially promotes adaptive compensation aimed at restoring ER homeostasis. However, sustained or maladaptive signal…

Clearance and secretion of α-synuclein by RTN3L-mediated endoplasmic reticulum autophagy.

The misfolding and aggregation of α-synuclein (α-syn), an abundant synaptic protein, leads to the pathogenesis of Parkinson's disease and related synucleinopathies. The cell-to-cell propagation of seeding-competent α-syn is initiated by unconventional protein secretion, yet the physiological pathway(s) underlying this process remain poorly defined. Here we show that α-syn secretion in human cells is mediated by Reticulon-3L (RTN3L)-dependent endoplasmic reticulum autophagy (ER-phagy), a conserved protein quality-control pathway that safeguards ER protein homeostasis. We also demonstrate that R…

Remodelling of smooth endoplasmic reticulum of Müller cells in aged human retina.

The smooth endoplasmic reticulum (SER) is a system of interconnected cytoplasmic tubules with various subdomains, which can remodel in response to certain stimuli, as in the retinal pigment epithelium. Earlier studies reported Müller cells (MC) to contain the SER tubules as the main subdomain. A routine examination of postmortem human retinas (54-94 years; N = 8) revealed diverse SER subdomains in aged MC. Besides the presence of tubules with a distinct lumen, MC possessed tubules that were rather compressed, with partial to fully obliterated lumen. The other SER subdomains detected were lamel…

Identification and validation of endoplasmic reticulum autophagy-related potential biomarkers in periodontitis.

Periodontitis is a chronic inflammatory disease that mainly occurs in the dental supporting tissues. Endoplasmic reticulum autophagy (ER-phagy) is a new type of selective autophagy. The main function of ER-phagy is to degrade excess ER membranes or toxic protein aggregates, control the volume of ER, and maintain cell homeostasis. This study utilized bioinformatics to identify and validate potential ER-phagy-related biomarkers for periodontitis. The relationship between immune cell infiltration and periodontitis as well as potential biomarkers was analyzed, to identify new targets for the diagn…

BPS exposure triggered endoplasmic reticulum (ER) stress, evidenced by elevated levels of ER stress-related proteins (XBP1-s, ATF4, and IRE1α), ultrastructural ER abnormalities observed via transmission electron microscopy, and increased immunostaining of IRE1α and ATF4 in RIF endometrial tiss …

The contribution of the Golgi and the endoplasmic reticulum to calcium and pH homeostasis in Toxoplasma gondii.

The cytosolic Ca2+ concentration of all cells is highly regulated demanding the coordinated operation of Ca2+ pumps, channels, exchangers, a … hangers that localize to the Golgi apparatus. We localized TgCAXL1 to the Golgi and the endoplasmic reticulum (ER) of T. gondii and validated its role as a Ca2+/proton exchanger by yeast complementation. Characterization of a knock-out mutant for TgCAXL1 (Δcaxl) underscored the role of TgCAXL1 in Ca2+ storage by the ER and acidic stores, most likely the Gol …

Regulation of endoplasmic reticulum stress and ferroptosis by rutin in a rat model of periodontitis.

Periodontitis is a bacteria-induced chronic inflammatory disease affecting tooth-supporting structures, involving endoplasmic reticulum (ER) stress and ferroptosis with a potential link between them. This study investigated the effects of rutin, a plant-derived flavonoid with diverse bioactivities, such as anti-inflammatory and antioxidant properties, on periodontitis in a rat model by modulating ER stress and ferroptosis pathways. Periodontitis was induced in rats using silk ligatures around molars. Rutin (0, 100, or 200 mg/kg) was administered orally for seven consecutive days, starting one …
