From Dysbiosis to Healthy Skin: Major Contributions of Cutibacterium acnes to Skin Homeostasis.

Cutibacterium acnes is the most abundant bacterium living in human, healthy and sebum-rich skin sites, such as the face and the back. This bacterium is adapted to this specific environment and therefore could have a major role in local skin homeostasis. To assess the role of this bacterium in healthy skin, this review focused on (i) the abundance of C. acnes in the skin microbiome of healthy skin and skin disorders, (ii) its major contributions to human skin health, and (iii) skin commensals used as probiotics to alleviate skin disorders. The loss of C. acnes relative abundance and/or clonal d…

Probiotics and gut microbiota modulation: implications for skin health and disease management.

The gut microbiota, consisting of a varied population of microorganisms in the digestive tract, is essential for sustaining overall human health, encompassing skin health. This review explored the intricate relationship between gut microbiota and various skin disorders, investigating the pathways through which gut dysbiosis may have impacted the development and progression of these conditions. We focused on the impact of gut microbiota on atopic dermatitis, psoriasis, acne vulgaris, acne rosacea, and melanoma. The review highlighted the potential of probiotics as a therapeutic strategy for mod

Microbiome-active drug delivery systems (MADDS): Leveraging microbial stimuli for controlled drug release.

The human microbiome comprises diverse microbial communities that inhabit tissues and biofluids throughout the body, including the gastrointestinal tract, lungs, vagina, and skin. These sites create dynamic microenvironments rich in enzymes, metabolites, and chemical gradients that act both as biological barriers and as localised targets for drug delivery. This review provides an overview of Microbiome-Active Drug Delivery Systems (MADDS), an emerging class of platforms that exploit microbial stimuli for site-specific therapeutic release. Unlike conventional systems that simply coexist with th

Longitudinal multi-omics analysis of host microbiome architecture and immune responses during short-term spaceflight.

Maintenance of astronaut health during spaceflight will require monitoring and potentially modulating their microbiomes. However, documenting microbial shifts during spaceflight has been difficult due to mission constraints that lead to limited sampling and profiling. Here we executed a six-month longitudinal study to quantify the high-resolution human microbiome response to three days in orbit for four individuals. Using paired metagenomics and metatranscriptomics alongside single-nuclei immune cell profiling, we characterized time-dependent, multikingdom microbiome changes across 750 samples

Characterisation of the intestinal microbiota of commercially farmed saltwater crocodiles, Crocodylus porosus.

The Australian saltwater crocodile (Crocodylus porosus) industry began commercially in the 1980s, producing skins for export and crocodile meat as a by-product. Industry research has thus far focused on strategies to improve production efficiency. In the current study, we utilised 16S rRNA sequencing to characterise the intestinal microbiome of Australian saltwater crocodiles. Samples were collected from 13 commercially farmed crocodiles from six sample sites along the length of the intestinal tract. The results indicate a similar microbiome composition to that found in the freshwater alligato

Role of microbes in the pathogenesis of neuropsychiatric disorders.

Microbes inhabit different anatomical sites of the human body including oral cavity, gut, and skin. A growing literature highlights how microbiome variation is associated with human health and disease. There is strong evidence of bidirectional communication between gut and brain mediated by neurotransmitters and microbial metabolites. Here, we review the potential involvement of microbes residing in the gut and in other body sites in the pathogenesis of eight neuropsychiatric disorders, discussing findings from animal and human studies. The data reported provide a comprehensive overview of the

Emerging Novel Therapies for the Treatment of Psoriasis: A Narrative Review.

Psoriasis is a chronic autoimmune and autoinflammatory disorder defined by abnormal skin cell turnover and inflammation, resulting in the formation of plaques on the skin. Although biologic therapies targeting interleukin (IL)-17 and IL-23 have significantly improved the treatment landscape for moderate-to-severe psoriasis, they are not effective for all patients. This highlights the need for additional therapeutic strategies. In recent years, exploring novel treatment avenues such as targeting IL-21, small nucleolar RNA (snoRNA) Snora73, the gut microbiome, and natural remedies have shown inc

Integrated microbiome and metabolome profiling in prepubertal girls with vulvar lichen sclerosus.

UNLABELLED: Vulvar lichen sclerosus (VLS) is a chronic anogenital disease with long-term risks of genital disfigurement and squamous cell cancer. The involvement of the gut-skin axis in prepubertal VLS remains unclear. We conducted a cross-sectional study of 43 prepubertal VLS patients and 46 healthy controls, performing fecal 16S rRNA sequencing and serum LC-MS to characterize gut microbiome-metabolome associations and identify altered microbial and metabolic signatures. We further assessed their exploratory classification performance using random forest models. No significant differences in

The Role of the Microbiome in Lichen Sclerosus: Pathophysiological Insights and Therapeutic Implications.

Lichen sclerosus (LS) is a chronic inflammatory and fibrotic dermatosis of unclear etiology, traditionally considered an autoimmune disorder. Emerging evidence suggests that microbiome dysbiosis may contribute to disease pathogenesis by modulating local immune responses and tissue remodeling. This narrative review synthesizes current data on the skin, genital, and gut microbiome in LS, focusing on potential mechanistic links between microbial imbalance and immune activation. Available studies consistently show alterations in microbial composition; however, findings remain heterogeneous because

Microbiome Responses to Oral Fecal Microbiota Transplantation in a Cohort of Domestic Dogs.

Fecal microbiota transplants (FMTs) have been successful at treating digestive and skin conditions in dogs. The degree to which the microbiome is impacted by FMT in a cohort of dogs has not been thoroughly investigated. Using 16S rRNA gene sequencing, we document the changes in the microbiome of fifty-four dogs that took capsules of lyophilized fecal material for their chronic diarrhea, vomiting, or constipation. We found that the relative abundances of five bacterial genera (Butyricicoccus, Faecalibacterium, Fusobacterium, Megamonas, and Sutterella) were higher after FMT than before FMT. Feca

More Than Skin Deep: Understanding the Skin-Bone Axis.

The skin's role as a mirror of internal disease has long been established, with clear associations to several internal organs. However, its relationship with bone is less well known. There are clear physiological links between the two organ systems, with their shared factors crucial to maintain structural integrity and homeostasis. Collagen, vitamin D homeostasis, aging, medications, and the gut microbiome are all factors that impact both skin and bone. It is of utmost importance that the skin-bone axis be adequately understood to support a healthy aging process. In this review, we discuss the

A Rare Case of Demodicosis Following Treatment With Oral Fluconazole.

Demodex folliculorum and Demodex brevis are commensal human ectoparasites that reside within or near hair follicles and have been highly associated with rosacea-like papulopustular skin eruptions. We present an interesting case of recurrent, iatrogenic demodicosis in a 56-year-old man. We suspect this to have been triggered by antifungal therapy given it occurred twice closely following azole treatment. We propose that oral antifungals in the setting of immunosuppression can alter the skin microbiome, facilitating Demodex proliferation.

The human microbiome as a source of novel bioactive natural products: structures, bioactivities, and biosynthetic insights.

The human microbiome, comprising trillions of microorganisms in distinct anatomical locations such as the gut, oral cavity, skin, and vagina, has emerged as a source of bioactive natural products with diverse scaffolds. Through co-evolution with the host, the human microbiome produces small molecules tailored to physicochemical environments that contribute to immune regulation, epithelial barrier maintenance, pathogen defense, and neurochemical signaling. Recent advances in metagenomics, single-cell genomics, synthetic biology, and integrated omics approaches have enabled rapid discovery and s

Nasal Microbial Community Shifts Following Treatment in Chronic Rhinitis: An Observational Study.

OBJECTIVES: Nasal microbiome helps maintain mucosal homeostasis and immune function. Previous studies suggested the pathogenesis of chronic rhinitis and the nasal microbiome were related. This study aimed to evaluate the treatment effect on the nasal microbiome in chronic rhinitis through analyzing microbial diversity and composition in pre- and post-treatment status. METHODS: In participants diagnosed with chronic rhinitis, nasal microbiome samples were collected before and after treatment. Treatment regimens included intranasal corticosteroid spray, oral antihistamines, and/or oral leukotri

Staphylococcus epidermidis Cicaria, a Novel Strain Derived from the Human Microbiome, and Its Efficacy as a Treatment for Hair Loss.

The skin tissue of the scalp is unique from other skin tissues because it coexists with hair, and many differences in microbial composition have been confirmed. In scalp tissues, hair loss occurs due to a combination of internal and external factors, and several studies are being conducted to counteract this. However, not many studies have addressed hair loss from the perspective of the microbiome. In this study, subjects with hair loss and those with normal scalps were set as experimental and control groups, respectively. In the experimental group, hair loss had progressed, and there was a la

Mendelian randomization analysis unveils causal relationships between skin microbiota and osteomyelitis.

BACKGROUND: Osteomyelitis results principally from endo-/exogenous bacterial infections, whose incidence that complicating bone injuries reaches approximately 30%, with the risk of disability and teratogenicity. Skin microbiota has been found to be clinically linked to osteomyelitis, but substantial evidence is lacking. METHODS: Mendelian randomization (MR) was applied to unveil the causality between human skin microbiome and osteomyelitis. Genetic data of 1,656 skin microbiota samples were obtained from genome-wide association studies (GWAS), with the outcome of osteomyelitis from UK Biobank…

Topical Application of Indole-3-Acetic Acid, Present in S. epidermidis Supernatant, Alleviates Atopic Dermatitis in Mice at Least via the Aryl Hydrocarbon Receptor Signalling Pathway.

Dysbiosis of the skin microbiome, characterised by Staphylococcus aureus overgrowth and imbalance of commensals such as Staphylococcus epidermidis (S. epidermidis), is closely associated with atopic dermatitis (AD). However, the therapeutic relevance of defined S. epidermidis-associated indole metabolite, especially indole-3-acetic acid (IAA), in AD-like inflammation remains incompletely characterised. Here, we investigated the role of IAA, a tryptophan-derived metabolite enriched in the culture supernatant of the tested S. epidermidis strain, in AD-like inflammation. Public transcriptomic ana…

Effects of common cosmetic ingredients on growth of dominant human skin inhabitant Cutibacterium acnes.

OBJECTIVES: Cosmetic products, which typically contain multiple ingredients such as ethanol, glycerol and Tween 80 (polysorbate 80), may influence the homeostasis of the skin microbiome. However, the effect of common ingredients on the anaerobic members of human skin microbiome is poorly studied. In this study, we directly evaluated the effects of common cosmetic ingredients on a dominant human skin anaerobe Cutibacterium acnes using in vitro techniques. MATERIALS AND METHODS: Five cosmetic ingredients were added to seven C. acnes strains, including type strains of the three subspecies (types…
