PP1, PP2A and PP2B Interplay in the Regulation of Sperm Motility: Lessons from Protein Phosphatase Inhibitors.

Male fertility relies on the ability of spermatozoa to fertilize the egg in the female reproductive tract (FRT). Spermatozoa acquire activated motility during epididymal maturation; however, to be capable of fertilization, they must achieve hyperactivated motility in the FRT. Extensive research found that three protein phosphatases (PPs) are crucial to sperm motility regulation, the sperm-specific protein phosphatase type 1 (PP1) isoform gamma 2 (PP1γ2), protein phosphatase type 2A (PP2A) and protein phosphatase type 2B (PP2B). Studies have reported that PP activity decreases during epididymal

Epididymal α-l-fucosidase and its possible role in remodelling the surface of bull spermatozoa.

The mammalian epididymis provides an appropriate environment for sperm maturation. During the epididymal transit, spermatozoa undergo biochemical and morphological changes that lead to the acquisition of the fertilizing capacity. In this study we analysed the fucosylation status of membrane glycoproteins in the spermatozoa obtained from different regions of the bull epididymis. High amounts of fucose were detected on caput spermatozoa (R.F.I. = 1010 ± 20.35), mostly located in the post-acrosome zone. A significant decrease in the fucose levels was detected toward the cauda (R.F.I. = 540.5 ± 49…

Characterization of recombinant murine binder of sperm protein homolog 1 and its role in capacitation.

Sperm capacitation is a maturation step that is deemed to be essential for sperm to fertilize an oocyte. A family of proteins, the binder of sperm (BSP), are known to bind choline phospholipids on sperm membranes and promote capacitation in bulls and boars. Recently, BSP-homologous genes have been identified in the epididymal tissues of human (BSPH1) and mouse (Bsph1, Bsph2). The aim of this study was to determine the binding characteristics of the murine binder of sperm protein homolog 1 (BSPH1) and evaluate its effects on sperm capacitation. Since it is not possible to purify the native BSP

Cytosolic and Acrosomal pH Regulation in Mammalian Sperm.

As in most cells, intracellular pH regulation is fundamental for sperm physiology. Key sperm functions like swimming, maturation, and a unique exocytotic process, the acrosome reaction, necessary for gamete fusion, are deeply influenced by pH. Sperm pH regulation, both intracellularly and within organelles such as the acrosome, requires a coordinated interplay of various transporters and channels, ensuring that this cell is primed for fertilization. Consistent with the pivotal importance of pH regulation in mammalian sperm physiology, several of its unique transporters are dependent on cytosol

Soma-sperm communication during the journey to fertilization: addressing challenges and opportunities†.

Male infertility is a significant concern in both human medicine and livestock. As it is mainly impacted by sperm quality, understanding the intrinsic characteristics of sperm that contribute to infertility is crucial. Male fertility relies on the sperm's ability to reach the site of fertilization, fertilize the oocyte, and support early embryonic development. Thus, classical sperm parameters such as motility, morphology, and the integrity of the plasma membrane, acrosome, and DNA are critical indicators of sperm quality. Recently, molecular sperm features have also been recognized as key comp

A morphometric comparison of the nuclear morphology of fresh and frozen-thawed human zona-bound and unbound sperm.

The role of sperm nuclear morphology in zona binding and the effect of cryopreservation on sperm nuclear morphology have been investigated using objective criteria. The nuclear dimensions of fresh and frozen-thawed sperm heads that had been prepared by the swim-up (SU) technique, and bound to human oocytes in vitro, were measured using a computer-assisted image analyzer. These were compared with each other and also to sperm in the SU samples and those that had not bound to the zona pellucida. The unbound (UB) sperm and SU sperm had very similar nuclear morphology. In contrast, the nuclear morp

Region-specific expression of androgen and growth factor pathway genes in the rat epididymis and the effects of dual 5alpha-reductase inhibition.

Dihydrotestosterone (DHT) is the primary androgen acting in the epididymis, the site of sperm maturation. Previously, we showed that the treatment of male rats with PNU157706, an inhibitor that acts on both isoforms of 5alpha-reductase to prevent DHT formation, has effects on the expression of genes implicated in processes that create the optimal luminal microenvironment required for sperm maturation, and on sperm maturation itself. However, signaling pathways involved in regulating or mediating DHT actions in the epididymis remain largely unknown. The goals of this study were to determine the…

Soluble adenylyl cyclase of sea urchin spermatozoa.

Fertilization, a key step in sexual reproduction, requires orchestrated changes in cAMP concentrations. It is notable that spermatozoa (sperm) are among the cell types with extremely high adenylyl cyclase (AC) activity. As production and consumption of this second messenger need to be locally regulated, the discovery of soluble AC (sAC) has broadened our understanding of how such cells deal with these requirements. In addition, because sAC is directly regulated by HCO(3)(-) it is able to translate CO₂/HCO(3)(-)/pH changes into cAMP levels. Fundamental sperm functions such as maturation, motili

Molecular, biochemical, and cellular characterization of epididymal ADAMs, ADAM7 and ADAM28.

The mammalian epididymis is critical for sperm to acquire motility and fertilizing capacity. This maturation process involves the interaction of epididymal secretory proteins with sperm. We analyzed mouse a disintegrin and metalloprotease (ADAMs) 7 and 28 expressed specifically or predominantly in the epididymis. We found that these ADAM genes are expressed in an epididymal region-specific manner and their gene expression is regulated by both androgen and testicular factors (ADAM7) or only testicular factors (ADAM28). We identified an ADAM28 transcript isoform that lacks the transmembrane doma

[Correlation between functional and ultrastructural status of spermatozoa (based on transmission electron microscopy and sperm-hyaluronan binding assay)].

INTRODUCTION: Standard diagnostic testing for male infertility includes a spermogram in accordance with current World Health Organization (WHO) recommendations. However, a basic ejaculate examination is insufficient to identify the causes of unsuccessful conceiving attempts, early pregnancy terminations, and ineffective ART protocols. Modern methods include transmission electron microscopy (TEM) of sperm, which allows visualization of their ultrastructural elements, and the HBA (Hyaluronan Binding Assay) test, which assesses the functional maturity of sperm. The clinical significance of these

A mammalian epididymal protein with remarkable sequence similarity to snake venom haemorrhagic peptides.

Following spermatogenesis in the testis, mammalian spermatozoa pass into the epididymis, where they undergo changes which confer on them forward motility and the ability to recognize and penetrate the egg. Many of these maturation events involve androgen-regulated epididymal proteins which become associated with the sperm membrane, and/or effect changes to integral sperm membrane proteins. Here we report the sequence of an 89 kDa androgen-regulated protein from rat (Rattus norvegicus) and monkey (Macaca fascicularis) epididymis that is synthesized exclusively in the caput region and is localiz

Differential testicular gene expression in seasonal fertility.

Spermatogenesis is an essential precursor for successful sexual reproduction. Recently, there has been an expansion in the knowledge of the genes associated with particular stages of normal, physiological testicular development and pubertal activation. What has been lacking, however, is an understanding of those genes that are involved in specifically regulating sperm production, rather than in maturation and elaboration of the testis as an organ. By using the reversible (seasonal) fertility of the Syrian hamster as a model system, the authors sought to discover genes that are specifically inv

Formation of a complex between TMEM217 and the sodium-proton exchanger SLC9C1 is crucial for mouse sperm motility and male fertility.

Sperm motility is essential for male fertility and is tightly controlled by signaling events in the flagellum. Slc9c1 encodes a sperm-specific Na+/H+ exchanger (sNHE/SLC9C1) that localizes to the flagellum and is indispensable for sperm motility and male fertility. SLC9C1 is unique among Na+/H+ exchangers in that it possesses a voltage-sensing domain (VSD), the physiological function of which remains poorly understood in mammals. Here, by analyzing coevolving genes with Slc9c1, we identified Tmem217, which encodes a transmembrane protein that is localized in the sperm flagellum. Knockout (KO)

Identification of bovine CD52-like molecule by monoclonal antibody IVA-543: distribution of CD52-like molecule in the bull genital tract.

The bovine maturation-associated sperm membrane antigen CD52-like molecule has been analysed using a mouse anti-sperm monoclonal antibody developed against bull spermatozoa. The antigen recognised by monoclonal antibody IVA-543 was detected on blood mononuclear cells (including lymphocytes and monocytes) and on a minor population of polymorphonuclear leukocytes. The bovine CD52-like molecule is secreted by the epididymal epithelium and then it is inserted into the sperm membrane during the epididymal transport in the distal part of epididymis. The CD52-like molecule was absent from spermatozoa
