Effects of topical analgesics on the pressor response evoked by muscle afferents.

PURPOSE: Pressor responses are reflexly evoked by the activation of groups III and IV muscle afferents, which are also known to mediate nociceptive responses. In this experiment, the effects of analgesic balm (AB) application on these responses were investigated without the interference of other types of anesthesia or effects from the higher brain. METHODS: Heart rate (HR), blood pressure, and end-tidal CO(2) were monitored in midcollicularly decerebrated cats. Static contractions (30 s) of hindlimb muscles were evoked by electric stimulation of L7 and S1 ventral roots. After control runs, a

Electric Field Effect on Inhibiting the Co-fibrillation of Amyloid Peptides by Modulating the Aggregation Pathway.

With the revelation of the close link between Alzheimer's disease (AD) and type II diabetes (T2D) and the possible assembly of multiple amyloid peptides therein, it is critical to understand and regulate the co-fibrillation pathway between related amyloid peptides. Here, we show experimentally and theoretically that electric field (EF) inhibited hybrid amyloid fibrillation of β-amyloid peptide (Aβ) and human islet amyloid peptide (hIAPP) by modulating the hetero-aggregation pathway. Experimental results confirm that the β-sheet secondary structure of amyloid peptides would be disrupted under s

Applied potential tomography: possible clinical applications.

Applied potential tomography (APT) or electrical impedance imaging has received considerable attention during the past few years and some in vivo images have been produced. This paper reviews the current situation in terms of what in vivo results have been and are likely to be obtained in the near future. Both static and dynamic imaging are possible and these two areas are dealt with separately. Features of the existing in vivo imaging system are good tissue contrast, high-speed data collection, good sensitivity to resistivity changes, low spatial resolution, low cost and no known hazard. It i

Local electrostatics governs tryptophan oxidation and enables rational stability engineering in antibodies.

Oxidation of tryptophan (Trp) residues in therapeutic antibody complementarity-determining regions (CDRs) can impair binding affinity, stability, and developability, yet the molecular determinants of site-specific susceptibility remain incompletely understood. Here, we present a structure-informed machine learning framework trained on Trp oxidation profiles from 187 monoclonal antibodies to identify the physicochemical drivers of oxidation risk. Beyond solvent accessibility, the dominant known predictor, we identify local residue-level electrostatic potential (Epot) as a strong independent mod

Coupled Hydrogen-Bond-Electrostatic Recognition of Phosphatidylglycerol Drives the Design of Resistance-Suppressing Miniature Peptidomimetics.

The emergence of multidrug-resistant (MDR) pathogens has urged us to find new antimicrobial strategies. Phosphatidylglycerol (PG) is an attractive bacterial-specific lipid target but is targeted by only one clinical agent, daptomycin. Yet daptomycin, like most reported PG binders, binds PG through an imprecise hydrophobic-electrostatic mode, necessitating a relatively large molecular size. This requirement, together with its strict Ca2+ dependence, significantly limits its efficacy. Here, we report bis-pyridinium amides (BisPAs), a rationally designed class of small molecules capable of precis

Engineering the Stereoselectivity of Short-Chain Dehydrogenases/Reductases for "Difficult-to-Reduce" Ketones Reduction via Electrostatic Tuning and Pocket Confinement.

Chiral alcohols derived from "difficult-to-reduce" ketones are key precursors in pharmaceutical intermediates and high-value fine chemicals. Despite advances in alcohol dehydrogenases (ADHs), the stereoselective reduction of "difficult-to-reduce" ketones remains a major challenge. In this study, we identified 10 short-chain dehydrogenases/reductases (SDRs) through gene mining using tetrahydrofuran-3-one and tetrahydrothiophene-3-one as substrates. Structure-guided mutagenesis combined with docking and molecular dynamics simulations revealed key residues regulating enantiomer selectivity. The A

Filling process-induced tribo-charging of lubricated and non-lubricated gelatine and HPMC capsules.

Tribo-charging can negatively affect pharmaceutical processes and the quality of pharmaceutical products. This study investigates electrostatic charging during the filling of hard capsules intended to be used in dry powder inhalers (DPIs). Gelatin and HPMC capsules without and with external lubricants (sodium lauryl sulfate, carnauba wax, and magnesium stearate) were subjected to automatic powder filling at two filling speeds and two humidities. Tribo-charging was compared among capsules and correlated with filling parameters using a lactose and 1 % budesonide formulation. Further, we evaluate

Determining the Electrostatic Contributions of GTPase-GEF Complexes on Interfacial Drug Binding Specificity: A Case Study of a Protein-Drug-Protein Complex.

Understanding the factors that contribute to specificity of protein-protein interactions allows for design of orthosteric small molecules. Within this environment, a small molecule requires both structural and electrostatic complementarity. While the structural contribution to protein-drug-protein specificity is well characterized, electrostatic contributions require more study. To this end, we used a series of protein complexes involving Arf1 bound to guanine nucleotide exchange factors (GEFs) that are sensitive or resistant to the small molecule brefeldin A (BFA). By comparing BFA-sensitive

Theoretical and experimental spectroscopic studies and analysis for wave function on N-phenylmorpholine-4-carboxamide benzene-1,2-diamine with computational techniques.

The present study focuses on structural and chemical analyses of N-phenylmorpholine-4-carboxamide benzene-1,2-diamine (PMCBD) using quantum computational methods. The calculated bond angle, length, and dihedral angle between atoms were compared with measured values. The observed and stimulated FT-IR (Fourier Transform Infrared Spectroscopy) spectra parameters for vibrational wavenumbers and their associated PED (Potential Energy Distribution) values in percentage have been obtained from VEDA4 software. The electronic transitions of PMCBD were discussed by TD-SCF/DFT/B3LYP based on the 6-311++G

A model for pH coupling of the SARS-CoV-2 spike protein open/closed equilibrium.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), causative agent of the coronavirus disease 2019 (COVID-19) pandemic, is thought to release its RNA genome at either the cell surface or within endosomes, the balance being dependent on spike protein stability, and the complement of receptors, co-receptors and proteases. To investigate possible mediators of pH-dependence, pKa calculations have been made on a set of structures for spike protein ectodomain and fragments from SARS-CoV-2 and other coronaviruses. Dominating a heat map of the aggregated predictions, three histidine residue

First-line palliative systemic therapy alternated with electrostatic pressurised intraperitoneal aerosol chemotherapy (oxaliplatin) for isolated unresectable colorectal peritoneal metastases: protocol of a multicentre, single-arm, phase II study (CRC-PIPAC-II).

INTRODUCTION: Despite its increasing use, first-line palliative systemic therapy alternated with electrostatic pressurised intraperitoneal aerosol chemotherapy with oxaliplatin (ePIPAC-OX), hereinafter referred to as first-line bidirectional therapy, has never been prospectively investigated in patients with colorectal peritoneal metastases (CPM). As a first step to address this evidence gap, the present study aims to assess the safety, feasibility, antitumour activity, patient-reported outcomes, costs and systemic pharmacokinetics of first-line bidirectional therapy in patients with isolated

Role of Electrostatic Interactions in Calcitonin Prefibrillar Oligomer-Induced Amyloid Neurotoxicity and Protective Effect of Neuraminidase.

Salmon calcitonin is a good model for studying amyloid behavior and neurotoxicity. Its slow aggregation rate allows the purification of low molecular weight prefibrillar oligomers, which are the most toxic species. It has been proposed that these species may cause amyloid pore formation in neuronal membranes through contact with negatively charged sialic acid residues of the ganglioside GM1. In particular, it has been proposed that an electrostatic interaction may be responsible for the initial contact between prefibrillar oligomers and GM1 contained in lipid rafts. Based on this evidence, the

Monitoring diclofenac adsorption by organophilic alkylpyridinium bentonites.

Organoclays have been applied as efficient adsorbents for pharmaceutical pollutants from aqueous solution. In this work, dodecylpyridinium chloride (C12pyCl) and hexadecylpyridinium chloride (C16pyCl) cationic surfactants were used for the preparation of organobentonites destined for diclofenac sodium (DFNa) adsorption, an anionic drug widely detected in wastewater. The organofunctionalization of the clay samples was performed under microwave irradiation at 50 °C for 5 min with surfactant amounts of 100% and 200% in relation to the cation exchange capacity (CEC) of the pristine bentonite. Identification of Extracellular DNA-Binding Proteins in the Biofilm Matrix.

We developed a new approach that couples Southwestern blotting and mass spectrometry to discover proteins that bind extracellular DNA (eDNA) in bacterial biofilms. Using Staphylococcus aureus as a model pathogen, we identified proteins with known DNA-binding activity and uncovered a series of lipoproteins with previously unrecognized DNA-binding activity. We demonstrated that expression of these lipoproteins results in an eDNA-dependent biofilm enhancement. Additionally, we found that while deletion of lipoproteins had a minimal impact on biofilm accumulation, these lipoprotein mutations incre

Phospholipid dependent mechanism of smp24, an α-helical antimicrobial peptide from scorpion venom.

Determining the mechanism of action of antimicrobial peptides (AMPs) is critical if they are to be developed into the clinical setting. In recent years high resolution techniques such as atomic force microscopy (AFM) have increasingly been utilised to determine AMP mechanism of action on planar lipid bilayers and live bacteria. Here we present the biophysical characterisation of a prototypical AMP from the venom of the North African scorpion Scorpio maurus palmatus termed Smp24. Smp24 is an amphipathic helical peptide containing 24 residues with a charge of +3 and exhibits both antimicrobial a

Modification of polychlorinated phenols and evaluation of their toxicity, biodegradation and bioconcentration using three-dimensional quantitative structure-activity relationship models.

Three-dimensional quantitative structure-activity relationship (3D-QSAR) models were established by comparative molecular field analysis (CoMFA) and comparative molecular similarity indices analysis (CoMSIA). Experimental toxicity data in Poecilia reticulata (pLC50) and physico-chemical properties for 12 polychlorinated phenols were used as dependent and as independent variables, respectively. Among the 12 polychlorinated phenols, nine were randomly selected and used as a training set to construct the 3D-QSAR models through the SYBYL-X software to predict the pLC50 values of the remaining 8 po

Estimation of electrokinetic and hydrodynamic global properties of relevant amyloid-beta peptides through the modeling of their effective electrophoretic mobilities and analysis of their propensities to aggregation.

Neuronal activity loss may be due to toxicity caused by amyloid-beta peptides forming soluble oligomers. Here amyloid-beta peptides (1-42, 1-40, 1-39, 1-38, and 1-37) are characterized through the modeling of their experimental effective electrophoretic mobilities determined by a capillary zone electrophoresis method as reported in the literature. The resulting electrokinetic and hydrodynamic global properties are used to evaluate amyloid-beta peptide propensities to aggregation through pair particles interaction potentials and Brownian aggregation kinetic theories. Two background electrolytes

Self-assembly of gold nanoparticles on poly(allylamine hydrochloride) nanofiber: a new route to fabricate "necklace" as single electron devices.

Fabrication of a percolating conductive device exhibiting "necklace-like morphology" is being reported utilizing a new route. This device was fabricated by exploiting the electrostatic self-assembly of citrate capped negatively charged Au nanoparticles (NPs) (60 nm diameter) over positively charged poly(allylamine hydrochloride) (PAH) fibrous scaffold and followed by synthesis of small Au NPs (∼10 nm) on the PAH surface. These 10 nm Au NPs were selectively synthesized over the PAH fiber surface using the surface catalyzed reduction of Au precursor (HAuCl4), leading to a continuous conducting n

Comprehensive analysis of the effects of CdSe quantum dot size, surface charge, and functionalization on primary human lung cells.

The growing potential of quantum dots (QDs) in applications as diverse as biomedicine and energy has provoked much dialogue about their conceivable impact on human health and the environment at large. Consequently, there has been an urgent need to understand their interaction with biological systems. Parameters such as size, composition, surface charge, and functionalization can be modified in ways to either enhance biocompatibility or reduce their deleterious effects. In the current study, we simultaneously compared the impact of size, charge, and functionalization alone or in combination on
