Inhibitory effect of a bioactivity-guided fraction from Rheum undulatum on the acid production of Streptococcus mutans biofilms at sub-MIC levels.

Rheum undulatum root has been used traditionally in Korea for the treatment of dental diseases. The purpose of this study was to separate a fraction from R. undulatum showing anti-acid production activity against Streptococcus mutans biofilms and identify the main components in that fraction. Methanol extract of R. undulatum root and its fractions were prepared. To select a fraction exhibiting anti-acid production activity, suspension glycolytic pH-drop assay was performed. Among the fractions tested, dichloromethane fraction exhibited the strongest activity in a dose-dependent manner. To exam…

The sloABCR operon of Streptococcus mutans encodes an Mn and Fe transport system required for endocarditis virulence and its Mn-dependent repressor.

Streptococcus mutans belongs to the viridans group of oral streptococci, which is the leading cause of endocarditis in humans. The LraI family of lipoproteins in viridans group streptococci and other bacteria have been shown to function as virulence factors, adhesins, or ABC-type metal transporters. We previously reported the identification of the S. mutans LraI operon, sloABCR, which encodes components of a putative metal uptake system composed of SloA, an ATP-binding protein, SloB, an integral membrane protein, and SloC, a solute-binding lipoprotein, as well as a metal-dependent regulator, S…

Bioabsorbable Cold Plasma-Generated Nitric Oxide Gas and Plasma-Activated Nitric Oxide Water: Inactivation of Oral Planktonic and Biofilm-Forming Streptococcus mutans.

Gas plasma exposure and plasma-activated water (PAW) containing long-lived reactive oxygen species (H2O2) and nitrogen species (NO2 ‒, NO3 ‒) generated by a multielectrode cylindrical dielectric barrier discharge plasma effectively inactivated oral bacteria Streptococcus mutans, opportunistic pathogen Staphylococcus aureus, and foodborne or intestinal Escherichia coli strains, which were used as comparator organisms. Gas plasma exposure to the water alters the water pH, H2O2, and NOx (Combined NO2 ‒ & NO3 ‒) concentrations. Gas Plasma (NOx and O3) achieved a 4-6 log reduction of bacteria withi…

Inhibitory efficacy of cyclo(L-leucyl-L-prolyl) from mangrove rhizosphere bacterium-Bacillus amyloliquefaciens (MMS-50) toward cariogenic properties of Streptococcus mutans.

Since Streptococcus mutans is the principal etiologic agent causing dental caries, by encompassing an array of unique virulence traits, emerging treatment strategies that specifically target the virulence of this pathogen may be promising as alternative approaches compared to conventional antibiotic therapy. In this perspective, we investigated chloroform extract of cell-free culture supernatant from mangrove rhizosphere bacterium Bacillus amyloliquefaciens (MMS-50) in terms of anticariogenic properties of S. mutans, without suppressing its viability. Crude chloroform extract of MMS-50 was sub…

Targeting mixed-species biofilms of Streptococcus mutans and Candida albicans on clear aligners: boron-based natural substance combinations for enhanced antibiofilm efficacy.

In orthodontic treatment, insufficiently cleaned clear-aligners can serve as reservoirs for biofilms. This study evaluates in-vitro biofilm models and investigates the antibiofilm efficacy of boron-based formulations with natural substances. Minimum inhibitory concentrations (MICs) were determined by microdilution, and Fractional inhibitory concentration by checkerboard methods. Biofilm inhibition was assessed at sub-MIC levels, and mature biofilm-eradication at supra-MICs. Time-kill assays were performed on mixed-biofilms of Streptococcus mutans and Candida albicans on clear-aligners. The MIC…

A mixed-bacteria ecological approach to understanding the role of the oral bacteria in dental caries causation: an alternative to Streptococcus mutans and the specific-plaque hypothesis.

For more than 100 years, investigators have tried to identify the bacteria responsible for dental caries formation and to determine whether their role is one of specificity. Frequent association of Lactobacillus acidophilus and Streptococcus mutans with caries activity gave credence to their being specific cariogens. However, dental caries occurrence in their absence, and the presence of other bacteria able to produce substantial amounts of acid from fermentable carbohydrate, provided arguments for non-specificity. In the 1940s, Stephan found that the mixed bacteria in dental plaque produced a…

Evaluation of the Antimicrobial Efficacy of Sodium Hypochlorite, Ozone, Diode Laser, and Photodynamic Therapy Against Enterococcus faecalis and Streptococcus mutans: An In-Vitro Study.

PURPOSE: Persistent bacterial contamination of the root canal system, particularly by Enterococcus faecalis (E. faecalis) and Streptococcus mutans (S. mutans), remains a major obstacle in endodontic therapy. Sodium hypochlorite (NaOCl) is widely recognized as the gold-standard irrigant due to its broad-spectrum antimicrobial properties. However, its cytotoxicity has prompted the exploration of alternative or adjunctive disinfection methods, including ozone therapy, diode lasers, and photodynamic therapy (PDT). This study aimed to evaluate and compare the antimicrobial efficacy of NaOCl, ozone,…

Cell Response Toward Inflammation of DPSCs Co-Cultured With Streptococcus mutans in the Presence of Alginate/Hydroxyapatite-Based Scaffolds Enriched With Silver Nanoparticles.

Managing bone defects is challenging, with autologous grafts being the most effective treatment. Biomaterials like alginate/hydroxyapatite (Alg/Hap) composites are increasingly used due to their biocompatibility and osteoconductive properties. Graft implantation in the oral cavity may trigger inflammatory responses, such as periodontitis, pulpitis, or caries, due to biomaterial recognition as non-self and the presence of pathogens like Streptococcus mutans. Conjugating Alg/Hap composites with antimicrobial silver nanoparticles (nAg) offers a strategy to counteract oral inflammation caused by m…

[Research Progress on Drug Intervention to Inhibit Dental Plaque Biofilm Formation by Streptococcus mutans Based on the Concept of Ecological Prevention of Dental Caries].

Dental caries is the local destruction of hard tooth tissue caused by acidic byproducts generated by cariogenic bacteria, primarily Streptococcus mutans, which ferment free sugars in the presence of host factors, dietary components, and environmental conditions. A main feature of dental caries is the formation of dental plaque biofilm, which significantly improves the resistance of bacteria to drugs and host immunity. Traditional anti-caries drugs mainly exert anti-biofilm functions indirectly through antibacterial activities. However, they tend to interfere with the symbiotic microbiota while…
