Effects of Vitamin D supplementation or deficiency on metabolic phenotypes in mice of different sexes.

Vitamin D is a steroid hormone precursor that was initially recognized for its important roles in calcium-phosphate homeostasis and bone health. However, the resent prevalence of vitamin D deficiency has highlighted its non-skeletal function, such as its important role in regulating endogenous metabolism. The aim of the present study was to examine the roles of vitamin D supplementation or deficiency on metabolic phenotypes in both male and female mice by using targeted metabolomics analysis. Six weeks old C57BL/6 mice of different sexes were fed with standard chow diet (1000 IU/kg vitamin D3

Metabolic adaptations to interrupted glycosaminoglycan recycling.

Lysosomal storage diseases (LSD) are metabolic disorders characterized by accumulation of undegraded material. The mucopolysaccharidoses (MPS) are LSDs defined by the storage of glycosaminoglycans. Previously, we hypothesized that cells affected with LSD have increased energy expenditure for biosynthesis because of deficiencies of raw materials sequestered within the lysosome. Thus, LSDs can be characterized as diseases of deficiency as well as overabundance (lysosomal storage). In this study, metabolite analysis identified deficiencies in simple sugars, nucleotides, and lipids in the livers o

Hyaluronan biosynthesis by class I streptococcal hyaluronan synthases occurs at the reducing end.

Previous studies reached different conclusions about whether class I hyaluronan synthases (HASs) elongate hyaluronic acid (HA) by addition to the reducing or the nonreducing end. Here we used two strategies to determine the direction of HA synthesis by purified class I HASs from Streptococcus equisimilis and Streptococcus pyogenes. In the first strategy we used each of the two UDP-sugar substrates separately to pulse label either the beginning or the end of HA chains. We then quantified the relative rates of radioactive HA degradation by treatment with beta-glycosidases that act at the nonredu

Browning and decomposed products of model orange juice.

A model solution of orange juice containing sugars, ascorbic acid, and citric acid was prepared and its browning during storage was examined. The solution gradually turned brown. Ascorbic acid (AsA) most contributed to the browning. Citric acid and such amino acids as Arg and Pro promoted the browning. DTPA, a strong chelator, inhibited the browning. 3-Hydroxy-2-pyrone (3OH2P), 5-hydroxymethylfurfural (HMF), furfural, 5-hydroxymaltol, and 2-furoic acid were identified as decomposed products in the stored solution. When 3OH2P was stored, the solution turned slightly brown. Furfural solution add

Glycoproteins secreted by sublingual glands of the echidna (Tachyglossus aculeatus).

The portion of echnida sublingual glycoproteins which could be solubilized was fractionated by electrophoresis in polyacrylamide gel and by filtration on a column of Sephadex G-100. Analyses for carbohydrate and amino acid are given and are compared with those of the submaxillary mucins of cattle, dog, pig and sheep. The molar ratio of galactosamine to sialic acid was roughly 1:1 in the echidna fractions, the sialic acid was N-acetyl-neuraminic acid, and there was very little neutral sugar. Proline was relatively high in amount, and isoleucine, half-cystine, methionine, leucine, tyrosine, and

Comprehensive Evaluation of Agronomic Traits and Nutritional Composition in Summer-Sown Vegetable Soybean Varieties from Shanghai, China.

Shanghai-native vegetable soybean (Glycine max [L.] Merril) landraces are valuable germplasms, but their systematic evaluation for agronomic and nutritional traits remains insufficient. This study aimed to assess their phenotypic and nutritional diversity to explore their potential for breeding and meeting dietary needs. Twenty-nine local landraces and one control cultivar ('Qingsu 7') were evaluated for key agronomic traits, yield components, nutritional traits, and isoflavone profiles, using hierarchical clustering, principal component analysis (PCA), and correlation analysis. The role of short-chain fatty acids in immunity, inflammation and metabolism.

Short-chain fatty acids (SCFAs) are carboxylic acids with carbon atom numbers less than 6, which are important metabolites of gut microbiome. Existing research shows that SCFAs play a vital role in the health and disease of the host. First, SCFAs are the key energy source for colon and ileum cells, and affect the intestinal epithelial barrier and defense functions by regulating related gene expression. Second, SCFAs regulate the function of innate immune cells to participate in the immune system, such as macrophages, neutrophils and dendritic cells. Third, SCFAs can also regulate the different

Comparative analysis of fecal phenolic content between normal and obese rats after oral administration of tea polyphenols.

Tea polyphenols (TP) have many health benefits, but most are metabolized into low molecular-weight phenolic acids after oral administration. In the present study, the absorption, metabolism, and excretion of catechins in rats fed a normal chow diet and in obese rats fed a high-fat and high-sugar (HFHS) diet were compared. After a ten-day oral administration of TP (500 mg per kg bw), the plasma levels of (-)-epigallocatechin gallate (EGCG) and (-)-gallocatechin gallate (GCG) in obese rats were significantly lower than those in the normal group. In obese rats, the fecal levels of EGCG, (-)-epica

Impacts of organic loading rate fluctuations and division of labor on sugar-based chain elongation revealed through metatranscriptomics.

Medium-chain carboxylates (MCCs) can be produced using open, mixed cultures of microorganisms in a process termed "chain elongation." Chain elongating bacteria can increase ATP yield by producing six-carbon caproate rather than four-carbon butyrate. Therefore, requiring chain elongating bacteria to maximize ATP yield for cell synthesis may be a way to increase production of the more valuable caproate. To test this, duplicate bioreactors were operated, and the impact of organic loading rate (OLR) fluctuations were assessed with glucose and xylose as substrates in media that did not include amin

Exploring correlations between soy sauce components and the formation of thermal contaminants during low-salt solid-state fermentation.

Soy sauce is a traditional seasoning in Asia and provides a unique flavor to food. However, some harmful Maillard reaction products (MRPs) were inevitably formed during the manufacturing process. Fermentation is a critical step of soy sauce manufacturing and has a significant impact on MRPs formation. Therefore, this study investigated the formation of some characteristic MRPs (e.g., furan, carboxymethyl lysine (CML), 5-hydroxymethylfurfural (5-HMF), α-dicarbonyl compounds) and their correlation with major quality indicators (e.g., free amino acids, reducing sugar, total acid, ammonia nitrogen

Silica-Based Stationary Phase with Surface Bound N-Acetyl-glucosamine for Hydrophilic Interaction Liquid Chromatography.

A hydrophilic silica-based stationary phase with surface bound N-acetylglucosamine (GlcNAc-silica) was prepared in house and characterized physically via Fourier transform infrared (FTIR) analysis and thermogravimetric analysis (TGA) and chromatographically over a wide range of mobile phase compositions. While both FTIR and TGA confirmed the attachment of the GlcNAc ligands to the silica surface, the chromatographic evaluation of GlcNAc-silica with polar and slightly polar standard solutes (e.g., sugars, nucleic acid fragments, phenolic, and benzoic acid derivatives) yielded the typical hydrop

Characterizing the chemical and sensory profiles of traditional American meads.

The compositional and sensorial profiles of traditional American meads were determined using standard enological, volatile, and descriptive analyses. Forty-one commercial meads produced by 35 meaderies across 20 states were selected to encompass a broad product range. The meads were analyzed for ethanol content, residual sugar, pH, titratable acidity, acetic acid, and free and total sulfur dioxide. Forty-three volatile compounds (alcohols, esters, acids, terpenes, aldehydes, aromatic hydrocarbons, etc.) were tentatively identified using a nontargeted HS-SPME-GC-MS method. Ethyl octanoate, phen

Separation, isolation, and amino acid composition of the albumin-like fraction from psoriatic scales.

The albumin-like fraction of psoriatic scales was isolated either directly by slab gel electroporesis or after previous treatment with polyethylene glycol6000. After both isolation procedures the amino acid composition was determined. Both proteins were rich of glutamic acid, glycine, and alanine. On the other hand both contained very small amounts of cysteine and tyrosine. Only after the direct isolation procedure the amino-sugar galactosamine could be detected.

Digestion and fermentation characteristics of the polysaccharides from Polygalae Radix processed by Glycyrrhizae Radix et Rhizoma and its effects on gut microbiota and metabolites.

Polygalae Radix Processed by Glycyrrhizae Radix et Rhizoma (GPR) contains polysaccharides (PGPR) as key active ingredients, yet their oral digestive fate and gut microbiota interactions remain elusive. In this study, we employed in vitro digestion and fermentation models to investigate PGPR's structural stability, fermentation properties, and modulatory effects on gut microbial communities and metabolites. Digestion experiments revealed no significant alterations in PGPR's molecular weight, free monosaccharides, reducing sugar (RS) content, or Fourier transform infrared spectral characteristic
