Novel Plasmodium falciparum metabolic network reconstruction identifies shifts associated with clinical antimalarial resistance.

BACKGROUND: Malaria remains a major public health burden and resistance has emerged to every antimalarial on the market, including the frontline drug, artemisinin. Our limited understanding of Plasmodium biology hinders the elucidation of resistance mechanisms. In this regard, systems biology approaches can facilitate the integration of existing experimental knowledge and further understanding of these mechanisms. RESULTS: Here, we developed a novel genome-scale metabolic network reconstruction, iPfal17, of the asexual blood-stage P. falciparum parasite to expand our understanding of metaboli…

An Integrated Workflow for Culturing and AI-Based Profiling of Mouse Intestinal Epithelium.

Two-dimensional (2D) culture systems are powerful in vitro tools that have revolutionized the study of intestinal stem cell biology. These culture systems derived from mouse intestinal and colonic epithelia enable the modeling of tissue architecture, barrier function, and disease mechanisms under physiologically relevant conditions. This protocol details methods for isolating and maintaining mouse intestinal crypts as 2D monolayers. Monolayers are established via enzymatic dissociation and seeding onto ECM-functionalized hydrogels. This protocol also describes a deep learning pipeline that per

Tryptophan-Kynurenine Pathway in COVID-19-Dependent Musculoskeletal Pathology: A Minireview.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) causes coronavirus disease 2019 (COVID-19), affecting multiple organ systems, including the respiratory tract and lungs. Several studies have reported that the tryptophan-kynurenine pathway is altered in COVID-19 patients. The tryptophan-kynurenine pathway plays a vital role in regulating inflammation, metabolism, immune responses, and musculoskeletal system biology. In this minireview, we surmise the effects of the kynurenine pathway in COVID-19 patients and how this pathway might impact muscle and bone biology.

Synaptopathy, circuitopathy and the computational biology of Huntington's disease.

Huntington's disease (HD) is one of the most common tandem repeat disorders and presents as a unique trilogy of cognitive, psychiatric and motor symptoms. One of the major mysteries of HD is why it selectively affects specific neuronal populations. A new article in BMC Biology provides a piece in the puzzle of pathogenesis. By demonstrating the delicate relationship between cortical and striatal neurons, it provokes broader questions of how we might understand HD as a disorder of synapses, neural circuits and systems biology.

Harnessing plant agriculture to mitigate climate change: A framework to evaluate synthetic biology (and other) interventions.

Plant agriculture contributes substantially to global greenhouse gas emissions, yet it also offers powerful opportunities for climate change mitigation. Here, we focus on how to identify and prioritize synthetic biology strategies to reduce emissions and sequester carbon through plant-based interventions. Effective solutions must process large volumes of carbon, be scalable, yield a positive life-cycle balance, and be economically viable, technically feasible, and deployable in field conditions without undue damage to what remains of nature on Earth. Using Fermi estimation, we quantify the per

Redox-responsive nanomedicine beyond glutathione: harnessing reactive oxygen species and emerging endogenous triggers for precision drug delivery.

Redox-responsive nanomedicine has emerged as a promising approach for site-selective drug delivery by leveraging pathological redox imbalances. Nevertheless, the majority of systems predominantly depend on glutathione (GSH) as a universal trigger, an assumption that frequently fails to account for the complexity and heterogeneity of endogenous redox biology across various diseases. This review critically evaluates redox-responsive nanomedicine beyond GSH, emphasizing emerging endogenous redox triggers such as reactive oxygen species (ROS), thioredoxin systems, NADPH-dependent pathways, hypoxia

Development of long-term oyster explant cultures enable sustained tissue-specific activity and reveal novel cellular behaviours.

Molluscs are ecologically and economically important, yet progress in understanding their cellular biology has been limited by the lack of reliable cell culture systems. Here, we establish reproducible long-term in vitro and ex vivo cultures from multiple tissues of the European flat oyster, Ostrea edulis, including heart, visceral mass, mantle, and gill. Explanted tissues remained intact and exhibited tissue-specific function for over two months, with whole hearts maintaining rhythmic contractions for up to nine months. Cultures revealed cellular plasticity, including haemocyte granule turnov
