Flow cytometric analysis of the phenotypic distribution of splenic lymphocytes in zinc-deficient adult mice.

Many immunodeficiency states create an imbalance in the ratio of T cells to B cells or in the subsets of lymphocytes found within these two major classes. A 30-d period of suboptimal intake of zinc often caused a 50% decrease in the total number of splenic lymphocytes in young adult mice. One- and two-color flow cytometric analysis was made of the residual splenocytes of the zinc-deficient mice to determine whether the deficiency had also altered the composition or phenotypic distribution of any of the major subsets of T cells or B cells. Marginally zinc-deficient mice had a normal ratio of T

TCR Signals Controlling Adaptive Immunity against Toxoplasma and Cancer.

T cells play a crucial role in adaptive immunity by recognizing and eliminating foreign pathogens and abnormal cells such as cancer cells. T cell receptor (TCR), which is expressed on the surface of T cells, recognizes and binds to specific antigens presented by major histocompatibility complex (MHC) molecules on antigen-presenting cells (APCs). This activation process leads to the proliferation and differentiation of T cells, allowing them to carry out their specific immune response functions. This chapter outlines the TCR signaling pathways that are common to different T cell subsets, as wel

T cell repertoire in mice with alopecia areata.

It has become clear that skin infiltrating autoreactive CD4+ T helper cells play a crucial role in the initiation of alopecia areata. However, the natures of the pathogenic T cell clones as well as of the skin antigen they recognize remain obscure. Here, we analyzed the T cell receptor repertoire expressed in the spleen of diseased mice. We consistently observed the dominant expansion of a limited set of T cell clones expressing Vbeta8.2/Jbeta2.5 T cell receptor rearrangement. We conclude that T cell response in mice alopecia areata is markedly oligoclonal; a feature that may permit the design

A reappraisal of the contribution of Th9 cells and interleukin-9 to the pathogenesis of rheumatoid arthritis.

Interleukin-9 (IL-9), the principal cytokine of Th9 cells, influences a vast range of functions due to the widespread expression of its receptor on various immune and non-immune cell types. Th9 cells and IL-9 play a detrimental role in the pathogenesis of some autoimmune diseases, such as rheumatoid arthritis (RA). Elevated Th9 cell numbers and higher IL-9 levels were observed in the peripheral blood and synovial tissues of patients with RA and animals with collagen-induced arthritis (CIA), correlating with disease activity. IL-9 can contribute to the pathogenesis of RA through multiple mechan

CD8+ T Cells You Should Know about in Autoimmunity: Current Paradigms of T Cell Pathogenesis in Autoimmune Disease.

PURPOSE OF REVIEW: CD8 T cells comprise a large portion of cells in inflamed tissues in many autoimmune diseases, yet their roles in autoimmune pathogenesis have been unclear. RECENT FINDINGS: Newer studies have demonstrated that CD8 T cells perform many effector functions that may play a vital role in autoimmune disease pathogenesis. In some autoimmune diseases, such as type 1 diabetes and polymyositis, classical cytotoxic T lymphocytes are thought to play a driving role, but in most tissues affected by autoimmune disease, granzyme K-expressing CD8 T cells are the most abundant. These cells

Effects of low-dose rapamycin on lymphoid organs of mice prone and resistant to accelerated senescence.

Aging is a complex, natural, and irreversible phenomenon that subjects the body to numerous changes in the physiological process, characterized by a gradual decline in the organism's homeostatic mechanisms, closely related to immunosenescence. Here, we evaluated the regulation of immunosenescence in lymphoid organs of senescence-accelerated prone 8 (SAM-P8) and senescence-accelerated resistant 1 (SAM-R1) mice treated with a low dose of rapamycin (RAPA). Mice were treated with a dose of 7.1 µg/kg RAPA for 2 months and had body mass and hematological parameters analyzed prior and during treatmen

Comparison and subsets analysis of peripheral CD4+T cells in patients with psoriasis and psoriatic arthritis.

Psoriatic arthritis (PsA) is a disease that transformed from psoriasis (PsO), and its underlying mechanisms are still not fully understood. Overactivation of the immune system is a key factor driving inflammatory diseases. Our goal is to define the unbalanced subsets of peripheral blood CD4 +T cells between PsO and PsA patients. Blood samples from 43 patients (23 PsA and 20 PsO) and 36 healthy donors (HD) were studied. Peripheral blood mononuclear cells (PBMC) were separated from blood and underwent fluorescent staining to assess CD4+T cell subsets by flow cytometry. We found that frequencies

Marked skewing of entire T-cell memory compartment occurs only in a minority of CMV-infected individuals and is unrelated to the degree of memory subset skewing among CMV-specific T-cells.

BACKGROUND: Chronic CMV infection drives the clonal expansion and accumulation of terminally differentiated, dysfunctional CMV-specific T-cells. CMV infection also appears to accelerate the differentiation of non-CMV-specific T-cells; however, the extent of this phenomenon is unclear. METHODS: The distribution of CD4 and CD8 T-cells into four memory subsets determined by CD45RA and CCR7 expression was analyzed in 96 CMV-infected (CMV+) and 81 CMV-uninfected (CMV-) older individuals. In CMV+ individuals, the distribution of IFN-γ producing CMV-specific T-cells into the same subsets was analyze

Altered T-cell subset repertoire affects treatment outcome of patients with myelofibrosis.

Phenotypic characterization of T cells in myelofibrosis (MF) is intriguing owing to increased inflammation, markedly elevated pro-inflammatory cytokines, and altered distribution of T-cell subsets. Constitutive activation of Janus kinase-2 (JAK2) in the majority of MF patients contributes to the expression of the programmed cell death protein-1 (PD1) and T-cell exhaustion. We wondered whether T-cell activation affects treatment outcome of patients with MF and sought to determine whether the JAK1/2 inhibitor ruxolitinib affects the activation of T-cell subsets. T cells from 47 MF patients were

Alterations in Th17 and Treg Lymphocyte Subset in Workers Occupationally Exposed to Lead.

Occupational exposure to lead (Pb) may have a deleterious effect on health of the workers. Among the various physiological systems, the immune system is one of the most susceptible targets of lead. Previous studies have been inconclusive in establishing the effect of Pb on the immune system. With this background, the aim of our study was to determine the effect of occupational Pb exposure on workers' immune parameters. A total of 110 individuals who were occupationally exposed to Pb and 97 apparently healthy non-exposed individuals were recruited in this study. Blood lead levels (BLL) were det

Demonstration of virus-specific cytotoxic T lymphocytes in liver tissue in hepatitis A--a model for immunopathological reactions.

The pathogenetic mechanism leading to liver tissue injury in hepatitis caused by hepatitis A virus is unclear. We have randomly established T cell clones from liver biopsies from 4 patients with hepatitis A. A total of 578 clones was phenotypically analyzed. Whereas during the acute phase of disease CD8+ clones dominated over CD4+ clones, from a biopsy taken late after onset of clinical syndromes more CD4+ than CD8+ clones were obtained. Interestingly, in a patient with a second exacerbation of the disease, more than 20% of all clones had the CD3+ WT31- CD4- CD8- gamma delta TCR+ phenotype. Va

[Immunological study of the T- and B-lymphocytes in normal cows and in chronic lympholeukemia].

T- and B-lymphocyte populations from lymphoid organs and tissue of normal cattle and cattle with chronic lymphocytic leukaemia (CLL) were studied. Comparative studies of surface properties, quantitative parameters and heterogeneity of main T- and B-cell populations were performed. It must be noted that proliferation of B-lymphocytes, bearing surface IgM in blood, lymph nodes and spleen is closely connected with the progression of leukaemic process. An increased number of B-lymphocytes (2-3 times) with the receptors for complement was found. The proportion of T mu and T gamma cell subsets in CL

"What do we know about regulatory T cells and endometriosis? A systematic review".

Endometriosis is a benign, chronic inflammatory disease that presents alterations in immune response that can be detected in eutopic endometrium, peritoneal fluid and peripheral blood of affected women. Regulatory T (TReg) cells are a subpopulation of T lymphocytes specialized in immune regulation that seem to participate in the development of endometriosis, by suppressing the immune response and favoring the establishment of lesions. Our aim was to review the scientific literature that evaluates TReg cell phenotypes in the context of endometriosis. PRISMA statement for systematic reviews was

New Predictor of Organ Failure in Acute Pancreatitis: CD4+ T Lymphocytes and CD19+ B Lymphocytes.

OBJECTIVE: Lymphocytes are one of the main effector cells in the inflammatory response of acute pancreatitis (AP). The purpose of the study was to evaluate whether peripheral blood lymphocyte (PBL) subsets at admission change during AP based on clinical outcomes and to explore whether these changes vary by aetiology of AP. Hence, we performed a prospective study to find a predictor in lymphocyte subsets that might allow easier, earlier, and more accurate prediction of clinical outcomes. METHODS: Patients with AP were enrolled from December 2017 to June 2018 at the First Affiliated Hospital of

Effect of glioblastoma and brain radiotherapy on T-lymphocyte subpopulations in rodents.

INTRODUCTION: Although lymphopenia is linked to immune suppression favoring tumor growth, the effect of different radiation types on specific T-lymphocyte subsets remains unclear. Among the T-lymphocyte subpopulations, CD8+-lymphocytes serve as key effectors in cancer immunity. This study aimed to examine the changes in T-lymphocyte subpopulations in both tumor-free and glioblastoma-bearing mice following brain-irradiation. METHODS: The study was divided into two main phases. First, C57BL/6 mice, with or without glioblastoma (GL261 cells), received hemispheric brain-irradiation or no-treatmen…

Effect of iron loading on peripheral blood lymphocyte subsets and on circulating cytokine levels in iron-depleted hemodialysis patients receiving erythropoietin.

BACKGROUND/AIMS: High doses of iron are recommended intravenously in iron-depleted hemodialysis (HD) patients receiving recombinant erythropoietin (EPO). Iron deficiency and mainly iron overload impair cellular and humoral immune response mechanisms. Imbalances in T cell subsets are common findings in disorders of iron metabolism. The aim of this study was to evaluate the effect of iron load on peripheral blood lymphocytes subsets and on circulating cytokine levels in HD iron depleted patients, treated with EPO. METHODS: We studied 19 stable adult HD patients, 12 males, with a mean age 59 +/-

Decreased CD3 + CD4+ T lymphocyte count predicts 28-day all-cause mortality in severe AECOPD patients admitted to the ICU: a retrospective cohort study.

Studies report a high 28-day all-cause mortality rate in patients with acute exacerbation of chronic obstructive pulmonary disease (AECOPD). The specific prognostic value of lymphocyte subsets for critically ill patients remains unclear. We aimed to evaluate the association between lymphocyte subsets and 28-day all-cause mortality in patients with AECOPD admitted to the intensive care unit (ICU). We conducted a retrospective cohort study on patients with severe AECOPD admitted to our hospital from January 2022 to December 2025. Comparisons were made between the survivors and non-survivors. Azithromycin targets the CD27 pathway to modulate CD27hi T-lymphocyte expansion and type-1 effector phenotype.

Macrolide antibiotic azithromycin is widely used in clinical practice to treat respiratory tract infections and inflammatory diseases. However, its mechanism of action is not fully understood. Given the involvement of the CD27 pathway in the pathophysiology of various T-lymphocyte-mediated inflammatory, autoimmune, and lymphoproliferative diseases, we examined the impact of AZM on CD27 regulation and potential consequences on CD4+ and CD8+ T-cell phenotypes. Using cellular immunology approaches on healthy donors' peripheral blood mononuclear cells, we demonstrate AZM-mediated downregulation of

Lymphocyte subpopulations in pediatric age. Definition of reference values by flow cytometry.

In the present paper we have analyzed the age-related changes in the distribution and numbers per cubic millimeter of the circulating lymphocyte subpopulations. Lymphocyte subsets have been examined by monoclonal antibodies specific for surface antigens (namely CD2, CD3, CD4, CD8, CD16, CD20 and CD57) and flow cytometry in lysed peripheral blood of normal neonates (1-3 days old), 1-6 month, 7-12 month, 1-2 year old babies and 3-5 year, 6-10 year old children and young people up to 17 years of age. The results confirm the lymphocytosis at birth and at later stages of life and indicate that majo
