Functional role of TASK-1 in the heart: studies in TASK-1-deficient mice show prolonged cardiac repolarization and reduced heart rate variability.

TASK-1, a member of the recently identified K2P channel family, is mainly expressed in the heart and the nervous system. TASK-1 is regulated by several physiological and pathological conditions and functions as a background potassium channel. However, there are limited data concerning the significance of TASK-1 in cardiac physiology. We studied the functional role of TASK-1 in the heart by cardiac phenotyping the TASK-1-deficient mouse (TASK-1(-/-)). TASK-1 was predominantly expressed in the ventricles of control animals. Real-time PCR and immunoblot demonstrated that the expression of seven o

Methodology for adapting a co-created early childhood development intervention and implementation strategies for use by frontline workers in India and Guatemala: a systematic application of the FRAME-IS framework.

There is little evidence on optimizing the effectiveness and implementation of evidence-based early childhood development (ECD) interventions when task-shifted to frontline workers. In this Methods Forum paper, we describe our adaptation of the International Guide for Monitoring Child Development (GMCD) for task-shifting to frontline workers in Guatemala and India. In 2021-2022, implementers, trainers, frontline workers, caregivers, and international GMCD experts collaborated to adapt the GMCD for a task shifted implementation by frontline workers. We used an eight-step co-creating process: as

Sequence Effect in Parkinson's Disease Is Related to Motor Energetic Cost.

Bradykinesia is the most disabling motor symptom of Parkinson's disease (PD). The sequence effect (SE), a feature of bradykinesia, refers to the rapid decrement in amplitude and speed of repetitive movements (e.g., gait, handwriting) and is a major cause of morbidity in PD. Previous research has revealed mixed results regarding the role of dopaminergic treatment in the SE. However, external cueing has been shown to improve it. In this study, we aimed to characterize the SE systematically and relate this phenomenon to the energetic cost of movement within the context of cost-benefit framework o

Coincidence detectors: visual processing of a pair of lines and implications for shape discrimination.

The psychophysical data reported here bear on how the boundaries of an object's retinal image are processed in early vision. We propose that the visual system contains a mechanism sensitive to four relationships between two local stimuli some distance apart and that the output of this hypothetical mechanism encodes and labels orthogonally the four relationships. We measured the just-noticeable difference in the orientation difference between two test lines as well as the just-noticeable differences in their mean orientation, mean location, and separation. A pair of noise lines was located betw

"Phase capture" in amblyopia: the influence function for sampled shape.

This study was concerned with what stimulus information humans with amblyopia use to judge the shape of simple objects. We used a string of four Gabor patches to define a contour. A fifth, center patch served as the test pattern. The observers' task was to judge the location of the test pattern relative to the contour. The contour was either a straight line, or an arc with positive or negative curvature. We asked whether phase shifts in the inner or outer pairs of patches distributed along the contour influence the perceived shape. That is, we measured the phase shift influence function. Our r

Development of a novel rodent rapid serial visual presentation task reveals dissociable effects of stimulant versus nonstimulant treatments on attentional processes.

The rapid serial visual presentation (RSVP) task and continuous performance tasks (CPT) are used to assess attentional impairments in patients with psychiatric and neurological conditions. This study developed a novel touchscreen task for rats based on the structure of a human RSVP task and used pharmacological manipulations to investigate their effects on different performance measures. Normal animals were trained to respond to a target image and withhold responding to distractor images presented within a continuous sequence. In a second version of the task, a false-alarm image was included,

Time-on-task and sleep deprivation effects are evidenced in overlapping brain areas.

Both sleep deprivation and extended task engagement (time-on-task) have been shown to degrade performance in tasks evaluating sustained attention. Here we used pulsed arterial spin labeling (pASL) to study participants engaged in a demanding selective attention task. The participants were imaged twice, once after a normal night of sleep and once after approximately 24h of total sleep deprivation. We compared task-related changes in BOLD signal alongside ASL-based cerebral blood flow (CBF) changes. We also collected resting baseline CBF data prior to and following task performance. Both BOLD fM

A task-based approach to assessing lead exposure among iron workers engaged in bridge rehabilitation.

The assessment of worker exposures to airborne contaminants in the dynamic environment present at most construction sites poses considerable challenges to the industrial hygienist. In this study, we applied a task-based approach to the assessment of lead exposure among structural steel iron workers engaged in a large, complex bridge rehabilitation project. We evaluated the usefulness of task-based exposure data for the development of worker protection programs. Task-specific and multitask samples were collected, and operation-specific and 8-hr time-weighted averages were calculated. The task-s

Fixation duration on natural scenes is explained by memory encoding not processing demand.

Before each of around 200,000 eye movements we make each day, the brain decides how long to fixate before shifting gaze to new information. Here we investigate this process using a large-scale scene-viewing experiment (4,080 natural scenes, five participants) that combines magnetoencephalography, eye tracking and a semantic captioning task. Using multivariate analysis of magnetoencephalography source-space patterns, behavioral analyses and artificial neural network (ANN) modeling, we show that longer fixations do not reflect prolonged visual processing but relate to downstream memory encoding.

Ergonomic effects of HMI design variables in dial-type gearshift manipulation.

This study examined the ergonomic effects of HMI design variables determining the manipulation method of dial-type gearshifts. Eight design alternatives were derived by combining two implementation levels for each of three design variables: P-position engagement method (Rotation, Push), Gear indicator location (Up, Left), and Gear position sequence (RND, DNR). Forty-three participants performed gear-shifting tasks using working prototypes of the design alternatives, during which task completion time, number of extra manipulations, perceived workload, perceived safety, and preference were measu

Perinatal mental health among women in Somalia a silent emergency demanding urgent health system action.

Perinatal mental health among women in Somalia represents a critical yet neglected public health emergency shaped by three decades of protracted conflict, gender-based violence, internal displacement, and one of the world's weakest health systems. With a maternal mortality ratio of 621 per 100,000 live births and fewer than 21% of births occurring in health facilities, depression and anxiety affect approximately one in four perinatal women-yet remain entirely absent from routine maternal care. This Perspective examines the structural, policy, and systemic barriers that perpetuate this neglect

Simultaneous brain, brainstem, and spinal cord pharmacological-fMRI reveals involvement of an endogenous opioid network in attentional analgesia.

Pain perception is decreased by shifting attentional focus away from a threatening event. This attentional analgesia engages parallel descending control pathways from anterior cingulate (ACC) to locus coeruleus, and ACC to periaqueductal grey (PAG) - rostral ventromedial medulla (RVM), indicating possible roles for noradrenergic or opioidergic neuromodulators. To determine which pathway modulates nociceptive activity in humans, we used simultaneous whole brain-spinal cord pharmacological-fMRI (N = 39) across three sessions. Noxious thermal forearm stimulation generated somatotopic-activation o

Deep learning: applications in retinal and optic nerve diseases.

Deep learning (DL) represents a paradigm-shifting, burgeoning field of research with emerging clinical applications in optometry. Unlike traditional programming, which relies on human-set specific rules, DL works by exposing the algorithm to a large amount of annotated data and allowing the software to develop its own set of rules (i.e. learn) by adjusting the parameters inside the model (network) during a training process in order to complete the task on its own. One major limitation of traditional programming is that, with complex tasks, it may require an extensive set of rules to accurately

Inequalities in access to and quality of abortion services in Mexico: Can task-sharing be an opportunity to increase legal and safe abortion care?

First-trimester abortion became legal in Mexico City in April 2007. Since then, 216 755 abortions have been provided, initially in hospitals, by specialized physicians using surgical techniques. With time and experience, services were provided increasingly in health centers, by general physicians using medical therapies. Meanwhile, abortion remains legally restricted in the remaining 31/32 Mexican states. Demand and need for abortion care have increased throughout the country, while overall abortion-specific mortality rates have declined. In an effort to ensure universal access to and improved

Neural correlates of attention to emotional facial expressions in dysphoria.

The present study investigated whether dysphoric individuals have a difficulty in disengaging attention from negative stimuli and/or reduced attention to positive information. Sad, neutral and happy facial stimuli were presented in an attention-shifting task to 18 dysphoric and 18 control participants. Reaction times to neutral shapes (squares and diamonds) and the event-related potentials to emotional faces were recorded. Dysphoric individuals did not show impaired attentional disengagement from sad faces or facilitated disengagement from happy faces. Right occipital lateralisation of P100 wa

Iron deficiency in infancy and neurocognitive functioning at 19 years: evidence of long-term deficits in executive function and recognition memory.

Iron deficiency in infancy negatively impacts a variety of neurodevelopmental processes at the time of nutrient insufficiency, with persistent central nervous system alterations and deficits in behavioral functioning, despite iron therapy. In rodent models, early iron deficiency impairs the hippocampus and the dopamine system. We examined the possibility that young adults who had experienced chronic, severe, iron deficiency as infants would exhibit deficits on neurocognitive tests with documented frontostriatal (Trail Making Test, Intra-/Extra-dimensional Shift, Stockings of Cambridge, Spatial

Effects of short-term nicotine deprivation on delay discounting among young, experienced, exclusive ENDS users: An initial study.

Delay discounting describes how rapidly delayed rewards lose value as a function of delay and serves as one measure of impulsive decision-making. Nicotine deprivation among combustible cigarette smokers can increase delay discounting. We aimed to explore changes in discounting following nicotine deprivation among electronic nicotine delivery systems (ENDS) users. Thirty young adults (aged 18-24 years) that exclusively used ENDS participated in two laboratory sessions: one with vaping as usual and another after 16 hr of nicotine deprivation (biochemically assessed). At each session, participant

Assessment of a glycine uptake inhibitor in animal models of effort-related choice behavior: implications for motivational dysfunctions.

RATIONALE: Motivated behavior can be characterized by a substantial exertion of effort, and organisms often make effort-related decisions based upon analyses of work-related response costs and reinforcement preference. Moreover, alterations in effort-based choice can be seen in people with major depression and schizophrenia. Effort-related decision making is studied using tasks offering choices between high effort options leading to highly valued reinforces vs low effort/low reward options. Gaze patterns predicting successful collision avoidance in patients with homonymous visual field defects.

Aim of the present study was to identify efficient compensatory gaze patterns applied by patients with homonymous visual field defects (HVFDs) under virtual reality (VR) conditions in a dynamic collision avoidance task. Thirty patients with HVFDs due to vascular brain lesions and 30 normal subjects performed a collision avoidance task with moving objects at an intersection under two difficulty levels. Based on their performance (i.e. the number of collisions), patients were assigned to either an "adequate" (HVFD(A)) or "inadequate" (HVFD(I)) subgroup by the median split method. Eye and head tr
