Cortical and thalamic neurones play a major role in the generation/expression of spike and wave discharges (SWDs), the main electroencephalographic (EEG) feature of absence seizures. The detailed mechanisms leading to this paroxysmal EEG activity, however, are still poorly understood. We have now made in vivo intracellular recordings from layer V cortical neurones of the facial motor cortex and from thalamocortical (TC) neurones of the ventroposteromedial and ventroposterolateral nuclei in a well established model of this disease: the Genetic Absence Epilepsy Rats from Strasbourg (GAERS). Stimulation of the thalamic nucleus ventralis intermedius (Vim) at high (130-Hz) frequency has been used over the last 8 years as a treatment in 134 patients with movement disorders (91 Parkinson's disease [PD], 23 essential tremor [ET], 21 various dyskinesias and dystonias, including four multiple sclerosis [MS]), implanted with long-term electrodes connected to a programmable stimulator. In PD patients, tremor was selectively suppressed for < or = 11 years. In ET patients, results were satisfactory, but in 35% of the cases deteriorated with time, when tremor had an action component. The present research addresses the effects of dorsomedial thalamic lesions on remembering a delayed alternation problem learned before damage. Male rats were trained on a delayed alternation task. Successful performance of the test first required learning of the general principle that Go and No Go trials alternate with each other and, second, in every trial, recalling the specific consequences of the responses in the previous trial after a relatively short or long interval. The animals which had reached the learning criteria were paired by the learning level shown during the training and assig

In 51 cases (6 cases with bilateral operations) with various kinds of tremor, stereotaxic ventralis intermedius (Vim) thalamotomies were performed using Leksell's apparatus and the results of operation evaluated. Several characteristics of the tremor, including clinical features and EMG, were correlated with the assumed location and volume of the coagulative lesion. In 54 of the 57 operations, the thalamic Vim nucleus was identified physiologically and a therapeutic lesion placed at a site that included the Vim neurons. In all these cases, except one in which the lesion was estimated to be too

Chronic experiments on conscious rabbits demonstrated that benzonal suppresses the spontaneous bioelectrical activity of the motor division of the cortex of large hemispheres while failing to affect the spontaneous biorhythms of the nonspecific nuclei of the thalamus, caudate nucleus, dorsal hippocamp and the basolateral nuclei of the tonsil. The drug decreased the electroexcitability of the motor cortex and caudate nucleus, exerted a mild depressant action on the reticular formation of the midbrain, tonsil, and hippocamp and had no effect on the excitability of nonspecific nuclei of the thala

To understand the neural basis of episodic memory it is necessary to appreciate the significance of the fornix. This pathway creates a direc … arity-based recognition (spared). This dissociation helped to show how the mammillary body-anterior thalamic nuclei axis, as well as the hippocampus, is vital for episodic memory yet is not required for familiarity-based recognition. These findings set the scene for a reformulation of temporal lobe and diencephalic amnesia. In this revised model, these two region …

The ventro-intermediate nucleus (Vim), as part of the motor thalamic nuclei, is a commonly used target in functional stereotactic neurosurgery for treatment of drug-resistant tremor. As it cannot be directly visualized on routinely used magnetic resonance imaging (MRI), its clinical targeting is performed using indirect methods. Recent literature suggests that the Vim can be directly visualized on susceptibility-weighted imaging (SWI) acquired at 7 T. Our work aims to assess the distinguishable Vim on 7 T SWI in both healthy-population and patients and, using it as a reference, to compare it w…

This study was aimed at determining (i) whether or not bilateral subtotal lesions of the anterior thalamic nuclei (ATH) in rodents produced memory deficits for spatial and/or non-spatial information and of auditory fear conditioning, and (ii) if these eventual deficits resemble those produced by chronic alcohol consumption (CAC). Working memory was assessed using both spatial (spontaneous alternation) and non-spatial (temporal alternation) delayed response tasks. Results showed that ATH lesions induced delay-dependent memory impairments in both spatial and non-spatial alternation tasks, as wel…

In this review, the clinical, neuropsychological, and neuroimaging findings in the alcoholic Korsakoff syndrome and in thalamic amnesia, resulting from focal infarction, are compared. In both disorders, there is controversy over what is the critical site for anterograde amnesia to occur-damage to the anterior thalamus/mammillo-thalamic tract has most commonly been cited, but damage to the medio-dorsal nuclei has also been advocated. Both syndromes show 'core' features of an anterograde amnesic syndrome; but retrograde amnesia is generally much more extensive (going back many years or decades)

PURPOSE: The thalamus, which consists of multiple subregions, has been of particular interest in the study of minimal hepatic encephalopathy (MHE). This study aimed to identify abnormalities of the thalamic subregions in patients with MHE and their associations with cognitive performance.
METHODS: Two groups of patients-30 patients with cirrhosis with MHE (the MHE group) and 48 patients with cirrhosis without MHE (the NHE group)-and 42 healthy controls (the HC group) underwent resting-state functional and structural magnetic resonance imaging. A histologically based atlas was used to segment

Wistar rats of a strain inbred for spontaneous generalized non-convulsive seizures present EEG spike-wave discharges (SWDs), 7-10 c/sec, occurring about once/min and lasting about 15 sec. They are bilateral and synchronous and can be recorded over the entire neocortex and in the lateral thalamic nuclei. After corpus callosum section, only a few bilateral SWDs persisted. Asymmetrical SWDs were most often recorded: bilateral SWDs starting asynchronously, or unilateral SWDs, which were also seen to alternate abruptly between hemispheres. The thalamic SWDs were always synchronized with the ipsilat…

Fatal Familial Insomnia (FFI) is characterized by loss of sleep, oneiric stupor with autonomic/motor hyperactivity and somato-motor abnormalities (pyramidal signs, myoclonus, dysarthria/dysphagia, ataxia). Positon emission tomography (PET) disclosed thalamic hypometabolism and milder involvement of the cortex; neuropathology severe neuronal loss in the thalamic nuclei variably affecting the caudate, gyrus cinguli and fronto-temporal cortices. Genetic analysis disclosed a mutation in the PRNP gene and FFI was transmitted to experimental animals, thus classifying FFI within the prion diseases. R…

AIM: To describe the speech rehabilitation history of patients with thalamic lesions.
BACKGROUND: Thalamic lesions can affect speech and language according to diverse thalamic nuclei involved. Because of the strategic functional position of the thalamus within the cognitive networks, its lesion can also interfere with other cognitive processes, such as attention, memory and executive functions. Alterations of these cognitive domains contribute significantly to language deficits, leading to communicative inefficacy. This fact must be considered in the rehabilitation efforts.

Selective, 35S-labeled, oligonucleotide probes were designed from sequences of the rat beta-1 and beta-2 adrenoceptor messenger RNAs for use in situ hybridization experiments on sections of unfixed rat brain and spinal cord. After hybridized sections were exposed to film or dipped in autoradiographic emulsion, specific and selective labeling patterns characteristic for each receptor messenger RNA and region of the central nervous system were observed. For example, labeling for beta-1 messenger RNA was found in the anterior olfactory nucleus, cerebral cortex, lateral intermediate septal nucleus

INTRODUCTION: Patients often complain about sensory symptoms that appear to the doctor as harmless, and reassurances are often given. Sensory strokes may easily be ignored.
CASE PRESENTATION: A 48-year-old Caucasian woman with insulin-dependent diabetes and hyperlipidemia experienced symptoms that progressed within hours to a complete left-sided hemisensory syndrome. This was caused by a lacunar infarct in the ventral posterior tier nuclei of the right thalamus. A few days later she gradually developed an almost identical, but incomplete hemisensory syndrome on the opposite side caused by a c

We determined the subjective visual vertical (SVV), ocular torsion (OT), skew deviation, and lateral head tilt in 35 patients with acute tha … ted moderate SVV tilts that were either ipsiversive or contraversive. In these 11 cases, vestibular thalamic nuclei (nucleus ventro-oralis intermedius, nucleus ventrocaudalis externus, and nucleus dorsocaudalis) were involved; infarctions in the remaining six were more ventromedial. Anterior polar infarctions did not affect vestibular function in roll.

BACKGROUND: Brain diffusion tensor imaging (DTI) has been shown to reflect cognitive changes in early Parkinson's disease (PD) but the diffusion-based measure free water (FW) has not been previously assessed.
OBJECTIVES: To assess if FW in the thalamic nuclei primarily involved with cognition (ie, the dorsomedial [DMN] and anterior [AN] nuclei), the nucleus basalis of Meynert (nbM), and the hippocampus correlates with and is associated with longitudinal cognitive decline and distinguishes cognitive status at baseline in early PD. Also, to explore how FW compares with conventional DTI, FW-corr…
