Microbial thiocyanate utilization under highly alkaline conditions.

Three kinds of alkaliphilic bacteria able to utilize thiocyanate (CNS-) at pH 10 were found in highly alkaline soda lake sediments and soda soils. The first group included obligate heterotrophs that utilized thiocyanate as a nitrogen source while growing at pH 10 with acetate as carbon and energy sources. Most of the heterotrophic strains were able to oxidize sulfide and thiosulfate to tetrathionate. The second group included obligately autotrophic sulfur-oxidizing alkaliphiles which utilized thiocyanate nitrogen during growth with thiosulfate as the energy source. Genetic analysis demonstrate

Acral gangrene as a presentation of non-uremic calciphylaxis.

We are describing a case of 55-year-old obese female with significant history of uncontrolled rheumatoid arthritis, who recently had decreased her immune-suppression medications. She presented with extensive acral gangrene involving multiple fingers and toes. Clinical picture and laboratory findings were suggestive of vasculitis; however, skin biopsy established diagnosis of calciphylaxis, in settings of normal kidney function. Patient was treated with sodium thiosulfate with gradual improvement in her skin lesions.

Radioiodination of mouse anti-III beta-tubulin antibodies and their evaluation with respect to their use as diagnostic agents for peripheral neuropathies.

The monoclonal antibody TU-20 and its scFv fragment were radiolabeled with 125 I in order to develop new imaging agents against the specific neuronal marker III beta-tubulin. The reaction via chloramine-T using thiosulfate as a stopping reductant was determined as the most convenient way for radioiodination. The preserved immunological properties of radioiodinated species were estimated by ELISA, electrophoresis, and immunohistochemistry with autoradiography. Biodistribution studies revealed a different behavior of radioiodinated TU-20 and its scFv.

[Disorders of microcirculation in food poisoning].

Thirty-seven patients with alimentary toxicoinfections were examined. The values of central and peripheral circulation and the fluid spaces were determined by one-stage dilution of two indicators, T-1824 and sodium thiosulfate. The capillary blood flow was studied by means of bulbar microscopy in reflected light. A deficit of interstitial fluid prior to treatment and disorder of microhemo- and lymphocirculation were revealed. Infusion of a crystalloid blood substitute with an optimum volume and rate increased capillary filtration and corrected the transinterstitial metabolism.

Modeling and Feasibility Assessment of Mineral Carbonation Based on Biological pH Swing for Atmospheric CO2 Removal.

Mitigating climate change requires both the reduction of greenhouse gas emissions and the removal of CO2 from the atmosphere. This study investigates a novel biological pH swing strategy for mineral carbonation at ambient conditions as a potential option for atmospheric CO2 removal. Through mathematical modeling, we evaluated a mineral carbonation system that utilized Desulfovibrio vulgaris and Acidithiobacillus thiooxidans to achieve alternating sulfur reduction and oxidation, respectively, with the cyclic process to effect pH swing for promoting the dissolution of a silicate mineral and the

Arcobacteraceae are ubiquitous mixotrophic bacteria playing important roles in carbon, nitrogen, and sulfur cycling in global oceans.

Mixotrophy is an important trophic strategy for bacterial survival in the ocean. However, the global relevance and identity of the major mixotrophic taxa remain largely elusive. Here, we combined phylogenetic, metagenomic, and metatranscriptomic analyses to characterize ubiquitous Arcobacteraceae based on our deep-sea in situ incubations and the global data. The phylogenomic tree of Arcobacteraceae is divided into three large clades, among which members of clades A and B are almost all from terrestrial environments, while those of clade C are widely distributed in various marine habitats in ad

Therapy-resistant dermatomyositis with extensive 'lumbar belt' calcinosis.

Calcinosis cutis (CC) is the umbrella term for calcium salt deposition on skin and subcutaneous tissue. We present a unique case of CC associated with anti-Mi2-positive dermatomyositis, having a distinctive distribution of subcutaneous calcifications appearing as a 'lumbar belt'. Treatment of CC remains challenging for clinicians due to a lack of high-quality evidence. Corticosteroids, methotrexate, bisphosphonates, intravenous immunoglobulin replacement, rituximab and sodium thiosulfate failed to halt calcinosis progression in this case. Newer therapies, such as Janus kinase inhibitors, shoul

Penile and generalised calciphylaxis in peritoneal dialysis.

This is a rare case of penile and generalised calciphylaxis. We describe the case of a patient admitted to our hospital for septic shock and necrotic skin findings, end-stage renal disease on peritoneal dialysis. Skin findings turned out to be calciphyactic lesions. The patient was taken to the operating room for penile debridement and started on antibiotics. He was treated with sodium thiosulfate and switched to haemodialysis. Calciphylaxis is a rare disease in which the treatment is basically supportive. Further studies are needed to identify the risk factors, mechanisms of disease and treat

Vitamin C-silver: an environmentally benign choice for DNA visualization on polyacrylamide gels.

A practical, sensitive and environment-benign protocol for the detection of DNA on polyacrylamide gels was described. In this method, the most commonly used formaldehyde-based developer in DNA silver stain, which poses potential hazards to the health of operators, is firstly replaced by vitamin C (Vc) in sodium thiosulfate solution. This allows user-friendly and efficient visualization of DNA that takes about 20 min to complete all the procedures, and provides comparable sensitivity (8 pg of single band) to the most sensitive formaldehyde-based silver staining method developed before.

Cotoneaster sp. poisoning in a llama (Lama glama).

A case of poisoning with a Cotoneaster sp. is described in a llama (Lama glama). The clinical signs were dyspnea, with congested mucous membranes, chewing, recumbency, and opisthotonos. Several hours after ingestion of the plant, the llama died, despite treatment with sodium thiosulfate and nitrite. Necropsy revealed approximately 676 g of leaves and fruit of Cotoneaster in the first compartment of the stomach. The blood was light red and did not clot. Because Cotoneaster sp. contains low concentrations of cyanogenic glycosides, toxicosis occurs only following massive ingestion of plant materi

Arteriolization of venous blood gases: a clue to the diagnosis of cyanide poisoning.

Prompt diagnosis and treatment of cyanide poisoning is essential for a successful therapeutic outcome. We present a patient with acute cyanide poisoning in whom venous blood gases disclosed an abnormally high pO2. Prompt treatment of the patient with the cyanide antidote, sodium nitrite plus sodium thiosulfate, rapidly reversed the cardiovascular and central nervous system manifestations of cyanide toxicity. The pathogenesis of cyanide-induced blood gas abnormalities and their potential value in the recognition of cyanide intoxication are discussed. Current treatment recommendations for cyanid

Effect of antidotes of the acute toxicity of methacrylonitrile.

When rats were exposed for 30 min to methacrylonitrile at concentrations between 3180 and 5700 ppm, the clinical symptoms observed suggested a toxic activity of metabolically formed cyanide. This is in contrast to the signs of toxicity observed in the same species after inhalation of acrylonitrile where metabolic cyanide formation plays only a minor role. The acute toxicity of methacrylonitrile could be antagonized with cyanide antidotes (4-dimethylaminophenol plus sodium thiosulfate) as well as with N-acetyl-cysteine which directly reacts with alpha,beta-unsaturated nitriles.

Non-chronic kidney disease-induced calciphylaxis: a rare case report.

Calciphylaxis is a disorder causing ischemic skin necrosis, typically associated with end-stage renal disease or those receiving dialysis. Occurrence is rare in those without end-stage renal disease, and treatment options are limited. This case report describes a patient with calciphylaxis without end-stage renal disease or history of dialysis. Treatment with sodium thiosulfate, a first line option, had to be stopped due to metabolic derangements, limiting the healing process. Diagnosis and treatment of this rare disorder are important to prevent further complications that may result.

[Efficacy of sodium thiosulphate in a case of calciphylaxis in a chronic nephropathic ninety-year-old patient on conservative therapy].

Calcific uremic arteriolopathy, also known as calciphylaxis (CUA), is a rare and potentially fatal condition that occurs in 1-4% of the population with chronic renal failure, most often on dialysis treatment. The pathogenesis is not yet clear although several hypotheses have been advanced, most importantly the alteration of the calcium phosphorus metabolism. Administration of sodium thiosulfate (STS) is the emerging therapy. Below, we report the case of an elderly chronic kidney patient on conservative therapy suffering from CUA who has been successfully treated with STS.

Combined antidotal and hemodialysis treatments for nitroprusside-induced cyanide toxicity.

A case report is presented for a patient with nitroprusside-induced cyanide intoxication. The patient received approximately 1 g sodium nitroprusside through an intravenous infusion of the drug over 6 d. The toxicity was treated with antidotes, sodium nitrite and sodium thiosulfate, followed by hemodialysis. Hemodialysis significantly removed thiocyanate but not cyanide from the patient's extracorporeal circulation. The removal of thiocyanate by hemodialysis enhanced the liberation of cyanide from the cytochrome oxidase. Hemodialysis is a significant addition to the therapy of cyanide toxicity

Sodium thiosulphate induced immobilized bacterial disintegration of sludge: An energy efficient and cost effective platform for sludge management and biomethanation.

The present study aimed to gain better insights into profitable biomethanation through sodium thiosulphate induced immobilized protease secreting bacterial disintegration (STS-IPBD) of sludge. STS disperse the flocs at 0.08 g/g SS of dosage and assists the subsequent bacterial disintegration. Immobilization of bacteria increases the hydrolytic activity of cells towards effective liquefaction of sludge. A higher liquefaction of 22% was accomplished for STS-IPBD when compared to immobilized protease secreting bacterial disintegration (IPBD alone). The kinetic parameters of Line Weaver Burk plot

Sodium thiosulphate: a new way of treatment for calciphylaxis?

Sodium thiosulphate has been used in medicine for almost 100 years. There are some very important areas of application such as cyanide intoxications or the treatment of cisplatin toxicity. Till this day, this compound is almost unknown to most physicians. This is astonishing, especially because there are some promising reports in the literature describing a possibly wider area of application in different domains of medicine such as urology, nephrology or dermatology. Perhaps one of the most exciting and recent domains of sodium thiosulphate use is the treatment of calciphylaxis, providing new
