The systematic assembly of supramolecular arrangements is a persistent challenge in modern coordination chemistry, especially where further aspects of complexity are concerned, as in the case of large molecular mixed-metal arrangements. A convenient method for the preparation of fluorous aryl stannanes was developed as a means of expanding the general utility of the fluorous labeling strategy (FLS). Following the synthesis of a novel fluorous distannane, a palladium-catalyzed cross-coupling reaction was used to prepare the target compounds from aryl halides. A novel approach for the determination of the stannous content in cold kits for labelling with 99mTc is described. The method is based on differential pulse polarography on the hanging mercury drop electrode in a methanol/water/perchloric acid mixture and is easy to perform. Single dose treatment with stannic chloride (4 micrograms/ml) on human lymphocytes in vitro revealed a significant increase of chromosomal aberrations. These showed a distinct relationship with the donor's age. Single and isochromatid breaks including gaps, premature chromosome condensation, irregular staining, stretching of the centromere and interchange, i.e. quadriradial exchange of chromosome arms, and also aneuploidy, were observed.

Using liposomes radiolabelled by the 99mTechnetium-stannous chloride technique we have investigated the effect of surface charge on the stability of the isotope in vitro and in vivo. Dialysis of 99mTc-labelled positive, negative and neutral liposomes, which had been incubated in either saline or normal rat serum showed no significant loss of the isotope from the liposome surface with only 2 per cent of the isotope dialysed. A comparison of gel chromatography with dialysis confirmed that most of the isotope remained attached to the liposome surface, but it did reveal greater loss of the isotope

Stannous ion (Sn2+, 30 microM) increased the amplitude of endplate potential (EEP) in the frog sartorius muscle, although the amplitude of miniature endplate potential (MEPP) or acetylcholine potential evoked by iontophoretic application of acetylcholine was unchanged. Sn2+ (10-100 microM) dose-dependently increased the quantal content of the EPP. MEPP frequency was not altered by 30 microM Sn2+. These findings indicate that Sn2+ may increase the EPP as a result of acceleration of the transmitter release evoked by nerve impulses.

An in vitro Tc-99m labeling method is described, which utilizes stable stock solutions of stannous chloride and disodium edetate (EDTA). The kit procedure requires as little as 1 ml of patient blood, can be performed in only 15 min, and gives labeling yields in excess of 98%. By using EDTA, the binding capacity of RBCs for technetium is sufficient to produce Tc-99m RBC doses with specific concentration greater than 50 mCi/ml for first-pass cardiac studies. There is evidence that undernutrition may contribute to the reduction in plasma fibronectin concentration and the depression of the reticuloendothelial (RE) system associated with severe sepsis. We have investigated the effects of fasting, surgical trauma and sepsis on plasma fibronectin concentrations and RE function. Technetium-99m-labelled colloids covering the entire spectrum of particle size were prepared from commercial kits (antimony colloid, sulphur colloid, tin colloid) or by chemical (excess aluminium) or thermal modification of sulphur colloid (heating time of 30 min). Blood clearance and organ extraction of a low-dose reticulo-endothelial test agent, technetium labelled tin colloid (TTC), was measured in groups of rabbits pretreated with reticulo-endothelial blocking agents. Electron microscopy and ultrastructure analysis confirmed that Kupffer cells extracted TTC. Pretreatment with silica caused reduced Kupffer cell uptake and spillover of TTC into the spleen. A facile synthesis of potential acetylcholinesterase (AChE) inhibitors, the tacrine analogues 3a-p, has been accomplished by direct cyclocondensation of 1-aryl-4-cyano-5-aminopyrazole with β-ketoesters using tin(IV) chloride as catalyst.
