BACKGROUND: Leishmaniasis is caused by Leishmania parasites and is transmitted to humans through the bite of infected sand flies. Development of Leishmania to infective metacyclic promastigotes occurs within the sand fly gut where the gut microbiota influences development of the parasite. Paratransgenesis is a new control method in which symbiotic bacteria are isolated, transformed and reintroduced into the gut through their diet to express anti-parasitic molecules. In the present study, the midgut microbiota of three sand fly species from a steppe and a mountainous region of northern Iran, wh

Microglia are innate immune system cells which reside in the central nervous system (CNS). Resting microglia regulate the homeostasis of the CNS via phagocytic activity to clear pathogens and cell debris. Sometimes, however, to protect neurons and fight invading pathogens, resting microglia transform to an activated-form, producing inflammatory mediators, such as cytokines, chemokines, iNOS/NO and cyclooxygenase-2 (COX-2). Excessive inflammation, however, leads to damaged neurons and neurodegenerative diseases (NDs), such as Parkinson's disease (PD), Alzheimer's disease (AD), Huntington's dise

Herein, high quality DNA-CuInS2 QDs are facilely synthesized through a one-pot hydrothermal method with fluorescence quantum yield as high as 23.4%, and the strongly fluorescent DNA-CuInS2 QDs have been utilized as a novel fluorescent biosensor for label-free and ultrasensitive detection of anthrax lethal factor DNA. L-Cysteine (L-Cys) and a specific-sequence DNA are used as co-ligands to stabilize the CuInS2 QDs. The specific-sequence DNA consists of two domains: phosphorothiolates domain (sulfur-containing variants of the usual phosphodiester backbone) controls the nanocrystal passivation an

Genetic code expansion (GCE) enables the site-specific incorporation of noncanonical amino acids into proteins, providing a powerful platform to investigate and engineer biological functions with molecular precision. While GCE has facilitated transformative studies in Escherichia coli, its utility in bacteria other than E. coli has been hindered by the lack of universal, high-efficiency methods for its implementation and optimization. To address this limitation, we introduce a modular, extensible platform for the generalizable implementation and optimization of GCE in phylogenetically diverse

Histiocytic sarcoma (HS) is an aggressive hematological malignancy whose transformed cells exhibit morphological and immunophenotypic characteristics similar to macrophages, and arises de novo or as part of a clonal 'evolution' of other pre-existing hematological neoplasms. This study investigates the potential use of the J774A.1 cell line (a cell line derived from murine tumor cells, commonly used in macrophage research) as a research model to study the role of polydisperse extracellular vesicles (PEVs) secreted by the HS cells, considering that bacterial infections are common in patients wit

Antibiotics are a medical wonder, but an increasing frequency of resistance among most human pathogens is rendering them ineffective. If this trend continues, the consequences for public health and for the general community could be catastrophic. The current clinical pipeline, however, is very limited and is dominated by derivatives of established classes, the "me too" compounds. Here, we have exploited our recent identification of a bacterial toxin to transform it into antibiotics active on multidrug-resistant (MDR) gram-positive and -negative bacterial pathogens. We generated a new family of

Chronic infectious keratitis represents a distinct clinical entity characterized by indolent progression, diagnostic difficulty, and prolonged therapeutic courses. Unlike acute bacterial keratitis, chronic infections often present with subtle inflammatory signs, recurrent episodes, or poor response to empirical antimicrobial therapy. Among the most important causes are herpetic keratitis, acanthamoeba keratitis, Microsporidial stromal keratitis, and atypical (non-tuberculous) mycobacterial keratitis. Herpes simplex virus keratitis may manifest as recurrent epithelial disease or immune-mediated

The remediation of polycyclic aromatic hydrocarbon and lead co-contaminated soils in karst regions remains challenging because of strong soil buffering capacity, complex metal speciation, and the distinct transformation mechanisms of organic pollutants and heavy metals. Although urea and biochar are widely used as soil amendments, their interactive effects on plant-assisted remediation and the associated rhizosphere microbial and metabolic mechanisms remain unclear. In this study, urea and biochar were co-applied in a ryegrass pot experiment to evaluate their synergistic effects on the remedia

Antimicrobial resistance threatens both human health and economic stability, urgently demanding antibiotic-free infection strategies. This global crisis brings to the fore a central challenge in antimicrobial material design lying in reconciling potency with biocompatibility, a dilemma acutely epitomized by cationic amphiphilic materials, despite their longstanding therapeutic significance. To address this, we present a glyco-cloaked charge-masking strategy that converts an intrinsically toxic cationic scaffold into a biocompatible and multifunctional platform. By employing a deep-red aggregat

This study explores the synthesis and antibacterial properties of silver nanoparticles (AgNPs) as a safer, eco-friendly alternative to traditional chemical treatments for bacterial infections. AgNPs were synthesized using aqueous extracts of marine microalgae, Isochrysis galbana and Chaetoceros calcitrans, via conventional and microwave-assisted methods, with the latter accelerating nanoparticle production. Extracts in ethanol, hexane, and acetone were tested, with the ethanolic extract of I. galbana showing the strongest antibacterial effects. The AgNPs exhibited broad-spectrum antibacterial

BACKGROUND: Meningitic Escherichia coli (E. coli) is the major etiological agent of bacterial meningitis, a life-threatening infectious disease with severe neurological sequelae and high mortality. The major cause of central nervous system (CNS) damage and sequelae is the bacterial-induced inflammatory storm, where the immune response of the blood-brain barrier (BBB) is crucial.
METHODS: Western blot, real-time PCR, enzyme-linked immunosorbent assay, immunofluorescence, and dual-luciferase reporter assay were used to investigate the suppressor role of transforming growth factor beta 1 (TGFβ1)

Composting has become an alternative for the treatment of organic effluents, due to its low cost, easy handling, and a great capacity for treating swine manure. As it is a biological process, many microorganisms are involved during the composting process and act in the degradation of organic matter and nutrients and also have the ability to degrade contaminants and accelerate the transformations during composting. The objective of this work was to identify microorganisms present in the swine effluent composting system, under the contamination by most used veterinary drugs in Brazil. The compos

Neisseria meningitidis, the meningococcus, is naturally competent for transformation throughout its growth cycle. The uptake of exogenous DNA into the meningococcus cell during transformation is a multi-step process. Beyond the requirement for type IV pilus expression for efficient transformation, little is known about the neisserial proteins involved in DNA binding, uptake and genome integration. This study aimed to identify and characterize neisserial DNA binding proteins in order to further elucidate the multi-factorial transformation machinery. The meningococcus inner membrane and soluble

Sound for the human voice is produced via flow-induced vocal fold vibration. The vocal folds consist of several layers of tissue, each with differing material properties. Normal voice production relies on healthy tissue and vocal folds, and occurs as a result of complex coupling between aerodynamic, structural dynamic, and acoustic physical phenomena. Voice disorders affect up to 7.5 million annually in the United States alone and often result in significant financial, social, and other quality-of-life difficulties. Understanding the physics of voice production has the potential to significant

Two regimens for transformation have been devised for the phage group 2 strains UT0002-19 and UT0017 of Staphylococcus aureus. Strain UT0002-19 produces exfoliative toxin, which is responsible for the clinical manifestations of the staphylococcal scalded skin syndrome, whereas strain UT0017 does not produce exfoliative toxin. A large pool of antibiotic-resistant and auxotrophic mutants from strains UT0002-19 and UT0017 were isolated by using (i) antibiotic gradient plates, (ii) trimethoprim selection, and (iii) nitrosoguanidine mutagenesis, which sometimes was coupled by enrichment with penici

Non-typeable Haemophilus influenzae (NTHi) is a common acute otitis media pathogen, with an incidence that is increased by previous antibiotic treatment. NTHi is also an emerging causative agent of other chronic infections in humans, some linked to morbidity, and all of which impose substantial treatment costs. In this study we explore the possibility that antibiotic exposure may stimulate biofilm formation by NTHi bacteria. We discovered that sub-inhibitory concentrations of beta-lactam antibiotic (i.e., amounts that partially inhibit bacterial growth) stimulated the biofilm-forming ability o

The cereolysin AB genes from Bacillus cereus VKM-B164 have been expressed in Bacillus anthracis strains: virulent H-7 (PXO1, PXO2), vaccine STI-1 (PXO1), 221 (without its own plasmids). Expression was achieved by cloning the genes in a high copy number plasmid pE194. This construct was integrated with host genomes in amplified form. Gold hamsters were vaccinated with parental and recombinant B. anthracis STI-1 and 221 strains and challenged with virulent ones subcutaneously. Gold hamsters vaccinated with 221 strains showed, absence of protection. STI-1 immunisation protected against the H-7 st

The escalating prevalence of multidrug-resistant (MDR) bacteria presents a critical global health challenge, necessitating the urgent development of alternative antimicrobial strategies. Silver nanoparticles (AgNPs) have emerged as promising antimicrobial agents due to their broad-spectrum activity, unique physicochemical properties, and multiple mechanisms of bacterial inhibition. Their nanoscale size, high surface area-to-volume ratio, and ability to generate reactive oxygen species (ROS) make them highly effective against both Gram-positive and Gram-negative bacteria. AgNPs exert their anti
