Chemical constituents of Callistemon citrinus from Egypt and their antiausterity activity against PANC-1 human pancreatic cancer cell line.

Human pancreatic cancer is resistant to almost all conventional chemotherapeutic agents. It is known to proliferate aggressively within hypovascular tumor microenvironment by exhibiting remarkable tolerance to nutrition starvation,  a phenomenon termed as "austerity". Search for the new agents that eliminate the tolerance of cancer cells to nutrition starvation is a promising strategy in anticancer drug discovery. In this study, two new meroterpenoids named callistrilones O and P (1 and 2) together with eight known triterpenes (3-10) were isolated from the active dichloromethane extract of Cal…

Phenolic constituents from Alisma plantago-aquatica Linnaeus and their anti-chronic prostatitis activity.

BACKGROUND: The plant Alisma plantago-aquatica Linnaeus, which is widely distributed in southwest of China, is the main material of traditional Chinese medicine "Zexie". It was used as folk medicine for immune-modulation, anti-tumor, anti-inflammatory and antibacterial. Previous chemical studies on A. plantago-aquatica reported the identification of triterpenes, diterpenes, sesquiterpenes, steroids, alkaloids and phenolic acid. Terpenes and phenolic acid were regard as major secondary metabolites from this medicine plant. RESULTS: A new phenolic acid, plantain A (1), along with four known com…

Role of Nitric Oxide and Hydrogen Sulfide in the Vasodilator Effect of Ursolic Acid and Uvaol from Black Cherry Prunus serotina Fruits.

The present research aimed to isolate the non-polar secondary metabolites that produce the vasodilator effects induced by the dichloromethan … ginine methyl ester (L-NAME), DL-propargylglycine (PAG) and glibenclamide (Gli). Additionally, both triterpenes increased NO and H2S production in aortic tissue. Molecular docking studies showed that ursolic acid and uvaol are able to bind to endothelial NOS and CSE with high affinity for residues that form the oligomeric interface of both enzymes. These resu …

From this fraction, we isolated an anacardic acids mixture and three known triterpenes: masticadienonic acid; 3alpha-hydroxymasticadienonic acid; 3-epi-oleanolic; as well as the sterol beta-sitosterol. Only the anacardic acids mixture exhibits a potent dose-dependent antibacterial activity (MIC=10 microg/ml in broth cultures). It is e …

Geographical Variation in Centella asiatica (L.) Urban: Morphological Adaptations Associated With Distinct Phytochemical Signals and Bioactivity of Extracts.

Centella asiatica (L.) Urban is a medicinal plant whose extract quality can vary with environmental context and associated differences in plant morphology and secondary metabolism. This study examined how geographical variation across three Indonesian agroecological regions (Kediri, Kulon Progo, and Tawangmangu) is associated with morphological variation, qualitative phytochemical profiles, and measured bioactivity of C. asiatica extracts. Comparative analyses revealed distinct region-specific morphological traits that covaried with differences in extraction yield, qualitative phytochemical si

Rethinking the Cucurbitane-Type Triterpenoid Profile of Bitter Gourd (Momordica charantia L.): The Chemical Transformation of Momordicine I in Solution.

Momordica charantia L., commonly known as bitter gourd, is a popular vegetable and medical herb in many countries. The most characteristic phytochemical profile of bitter gourd is its high content of cucurbitane-type triterpenoids, which are responsible for its bitter taste and therapeutic potential. More than 260 cucurbitane-type triterpenoids, including at least 11 5,19-hemiacetals, 40 5,19-methyl acetals, 46 methyl ethers, 6 5,19-ethyl acetals, and 7 ethyl ethers, have been identified from bitter gourd. In this work, by studying the chemical transformation of momordicine I, one of the most

[Isolation and quantification of a 17-epi-dammarane triterpenoid rhuslactone from roots of Rhus chinensis and its preventive effects on coronary heart disease and thrombosis in zebrafish].

Based on mass spectrometry(MS)-guided separation strategy, compound 1 was obtained from the roots of Rhus chinensis. By comprehensive analysis of high resolution-electrospray ionization-mass spectrometry(HR-ESI-MS), nuclear magnetic resonance(NMR) data, and quantum chemical calculation of NMR(qcc-NMR) parameters, compound 1 was elucidated as rhuslactone, a 17-epi-dammarane triterpenoid with a rare 17α-side chain. An HPLC-ELSD method for its quantification in R. chinensis was established and adopted for the quantification of rhuslactone in different batches of R. chinensis. Rhuslactone displaye

Comparison of Bioactive Secondary Metabolites and Cytotoxicity of Extracts from Inonotus obliquus Isolates from Different Host Species.

Inonotus obliquus, a wood-decaying mushroom, has been used as a health-promoting supplement and nutraceutical for centuries. It is a source of bioactive compounds accumulated in both the conks (pseudosclerotia/sclerotia) and the biomass obtained in vitro. This study aimed to qualitatively and quantitatively analyze the bioelements and selected metabolites produced in mycelial cultures obtained from different host species. The mycochemical potential of mycelial cultures isolated from pseudosclerotia grown in Betula pendula, Alnus glutinosa, and Carpinus betulus was compared. Parent cultures wer

Amoebicidal Activity of Caffeine and Maslinic Acid by the Induction of Programmed Cell Death in Acanthamoeba.

Free-living amoebae of the genus Acanthamoeba are the causal agents of a sight-threatening ulceration of the cornea called Acanthamoeba keratitis, as well as the rare but usually fatal disease granulomatous amoebic encephalitis. Although there are many therapeutic options for the treatment of Acanthamoeba infections, they are generally lengthy and/or have limited efficacy. For the best clinical outcome, treatments should target both the trophozoite and the cyst stages, as cysts are known to confer resistance to treatment. In this study, we document the activities of caffeine and maslinic acid

Phytochemical and antibacterial studies of the ethanolic stem barks extract of Anthocleista vogelii planch, and the in silico study of its secondary metabolites on two bacteria strains Staphylococcus aureus and Salmonella typhi.

This study investigates the phytochemical composition, antibacterial efficacy, and molecular interactions of Anthocleista vogelii stem barks extracts. Phytochemical screening revealed the presence of flavonoids, alkaloids, triterpenes, phenols, saponins, and other bioactive compounds in 95 % ethanol and hydro-ethanol extracts, while coumarins, steroids, and anthraquinones were absent in aqueous extracts. The 95 % ethanol extract demonstrated significant antibacterial activity against Esherichia coli (MIC: 64 μg/mL) and Klebsiella pneumoniae (MIC: 8 μg/mL), with moderate effects against Staphyl…

Synthesis and anti-inflammatory activity of saponin derivatives of δ-oleanolic acid.

Pentacyclic triterpenes (PTs) are the active ingredients of many medicinal herbs and pharmaceutical formulations, and are well-known for their anti-inflammatory activity. On the other hand, anti-inflammatory effects of AMP-activated protein kinase (AMPK) have recently drawn much attention. In this study, we found that a variety of naturally occurring PTs sapogenins and saponins could stimulate the phosphorylation of AMPK, and identified δ-oleanolic acid (10) as a potent AMPK activator. Based on these findings, 23 saponin derivatives of δ-oleanolic acid were synthesized in order to find more po…
