Predictive value of tryptophan/large neutral amino acids ratio to antidepressant response.

The molar ratio of total plasma tryptophan to the sum of other large neutral amino acids (viz., valine, isoleucine, leucine, tyrosine, phenylalanine), thought to reflect brain serotonin formation, was estimated in 69 patients with major depression before and after 1 week of treatment with different serotonin reuptake inhibitors, fluvoxamine (n = 28), fluoxetine (n = 10), citalopram (n = 7), mixed serotonin-noradrenaline reuptake inhibitor amitriptyline (n = 11), clomipramine (n = 8) and the preferentially noradrenaline reuptake inhibitor, nortriptyline (n = 5). A significant difference among '…

Tryptophan loading increases daytime serum melatonin levels in intact and pinealectomized rats.

L-tryptophan (L-Trp) effects on serum and pineal melatonin were studied in fasted and non-fasted rats. Oral administration of L-Trp during the day caused serum melatonin to increase 4-fold compared to that of saline treated rats; this occurred in both fasted and non-fasted animals. In the same animals, neither NAT, HIOMT, not melatonin levels were changed. In a second study, pinealectomized rats were loaded with L-Trp. In these animals, as in those that had an intact pineal gland, L-Trp loading caused a large increase in immunoreactive serum melatonin. The results suggest that the increased me…

Demonstration of non-neural tryptophan 5-mono-oxygenase in mouse intestinal mucosa.

Tryptophan 5-mono-oxygenase was demonstrated and its activity was measured in mucosal extracts of the mouse digestive tract by means of highly sensitive h.p.l.c. detection. The intestinal enzyme was activated by anaerobic incubation with dithiothreitol, as are the enzymes from mouse mastocytoma cells and bovine pineal gland. The dithiothreitol-enhanced activity was highest at the proximal portion of colon followed by that at the duodenum, where the unenhanced activity/enhanced activity ratio was highest. The enzymic and immunochemical properties of the intestinal tryptophan 5-mono-oxygenase we…

Orchestrating Physiological Homeostasis: The Spatial and Cellular Heterogeneity of Tryptophan Transport and Metabolism.

In addition to being incorporated into proteins, tryptophan (Trp) is increasingly recognized as a critical metabolic hub orchestrating systemic physiological homeostasis through interactions among the digestive, nervous and immune systems. This review systematically delineates the latest advancements in the molecular mechanisms governing the spatial and cellular heterogeneity of Trp transport. We summarize the sophisticated transmembrane machinery of Trp transport in intestinal epithelial cells, including angiotensin-converting enzyme 2-dependent broad neutral amino acid transporter 1 complex …

Serotonin, Kynurenine, and Indole Pathways of Tryptophan Metabolism in Humans in Health and Disease.

Tryptophan (TRP) is a proteinogenic and nutritionally essential amino acid involved in the formation of numerous bioactive substances. A crucial role in the TRP molecule is played by indole, a bicyclic ring formed by benzene and pyrrole, which confers hydrophobic and antioxidant properties and the ability to act as a ligand for aryl hydrocarbon and pregnane X receptors. The first parts of the article examine sources, nutritional requirements, and three pathways of TRP catabolism. Physiologically, ~5% of dietary TRP is catabolized through the pathway forming serotonin and melatonin in the brain…

Magnetosensitivity of Model Flavin-Tryptophan Radical Pairs in a Dynamic Protein Environment.

Light-induced radical pairs in cryptochrome proteins located in the retina are thought to be the receptors at the heart of the magnetic compass sense of migratory songbirds. Reliable simulations of the performance of such sensors face several fundamental challenges. The quantum spin dynamics of large spin systems must be modeled for periods in excess of a microsecond including realistic local magnetic interactions that fluctuate on a picosecond to microsecond time scale as a result of thermal motion. Here we employ newly developed computational methods that combine explicitly time-dependent in

Tryptophan as a biomarker of pregnancy-related immune expression and modulation: an integrative review.

BACKGROUND: Degradation pathways of Tryptophan (TRP) are implicated in a spectrum of physiological adaptations and outcomes associated with pregnancy. The immunomodulatory role of TRP and its metabolites through the indoleamine 2,3-dioxygenase (IDO) pathway is particularly relevant to pregnancy due to its potential influence on maternal and fetal immune tolerance and the mother's health. METHODS: A targeted literature search was conducted via PubMed, Web of Science, and Embase, focusing on maternal serum TRP levels in pregnancy. We included original human subject research on maternal serum TR…

Leaf beetles employ tryptophan to detoxify the chemical defenses of poplar trees.

Herbivorous insects have evolved many fascinating adaptations to overcome the chemical defenses of their host plants. This study employed targeted and untargeted metabolomics, coupled with isotope labeling, to shed light on the metabolism of salicinoids-potent antiherbivore phenolic defenses present in the Salicaceae family-in the poplar-specialized leaf beetle, Chrysomela tremulae. C. tremulae was found to produce a range of metabolites from salicortin and utilize the essential amino acid tryptophan and its breakdown products, namely kynurenine, kynurenic acid, and 4-hydroxyquinoline, to form…

Evolutionary computation for reconstructing threshold networks of the tryptophan operon in Escherichia coli.

A recent Boolean model of the tryptophan operon in Escherichia coli has been developed, aiming to capture the operon's on/off states through a set of fixed points. However, when updated synchronously, the model also reveals the presence of unwanted spurious limit cycles, that have a larger basin of attraction. This study introduces an evolutionary computation framework to search for threshold network models that accurately portray the desired fixed points while eliminating occurrences of limit cycles. Employing both differential evolution and particle swarm optimization within the proposed fra…

Effect of tryptophan starvation on inclusion membrane composition and chlamydial-host interactions.

Chlamydia is an obligate intracellular bacterial pathogen that develops within a membrane-bound vacuole called an inclusion. Throughout its developmental cycle, Chlamydia modifies the inclusion membrane (IM) with type III secreted (T3S) membrane proteins, known as inclusion membrane proteins (Incs). Via the IM, Chlamydia manipulates the host cell to acquire lipids and nutrients necessary for its growth. One key nutrient is tryptophan (Trp). As a Trp auxotroph, Chlamydia is very sensitive to Trp starvation and, in response to low Trp levels induced by the immune response, enters a viable but no…

Characterization of CSF tryptophan metabolites in South African children with tuberculous meningitis.

OBJECTIVES: Tuberculous meningitis (TBM), a severe form of tuberculosis, remains difficult to diagnose and is fatal if left untreated. Children are particularly vulnerable to the disease due to their immature immune systems. Often, diagnosis is delayed by nonspecific symptoms, leading to a more advanced stage of disease at presentation. Tryptophan metabolism has been linked to disease severity in adult TBM; however, its role in children remains largely unexplored. This study investigates cerebrospinal fluid (CSF) tryptophan metabolism, both the serotonin and kynurenine pathways, in paediatric …

Copper(II) phthalocyanine as an electrocatalytic electrode for cathodic detection of urinary tryptophan.

Herein, we introduce a novel method for tryptophan detection via a reduction reaction facilitated by its interaction with a copper(II) phthalocyanine (CuPc) electrocatalytic electrode. This method addresses challenges associated with the susceptibility of the oxidation response to interference from various species when measuring tryptophan in bodily fluids. The reduction currents exhibit a linear increase with tryptophan concentrations in two ranges: 0.0013-0.10 mM and 0.10-1.20 mM, with the sensitivities of 14.7 ± 0.5 μA mM-1 and 3.5 ± 0.1 μA mM-1, respectively. The limit of detection (LOD, 3…

L-tryptophan carbon dots as a fluorescent probe for malachite green detection.

Development of a rapid and sensitive detection method for hazardous dyes attracts considerable research interest. In this work, L-Tryptophan-based Carbon dots were developed as a fluorescence sensor for the detection of Malachite green (MG). Green fluorescent L-Trp-C-dots were synthesized by a simple pyrolysis technique using L-Trp as the starting precursor. L-Trp-C-dots exhibited different quenching responses to MG, and other interfering species, consequently offering a selective strategy to detect MG. The proposed sensor shows a limit of detection (LOD) of 0.06 μM and a limit of quantificati…

The correlation of inflammation, tryptophan-kynurenine pathway, and suicide risk in adolescent depression.

Accumulating evidence suggests a role for the tryptophan-kynurenine pathway (TKP) in the psychopathology of major depressive disorder (MDD). Abnormal inflammatory profile and production of TKP neurotoxic metabolites appear more pronounced in MDD with suicidality. Progress in understanding the neurobiology of MDD in adolescents lags significantly behind that in adults due to limited empirical evidence. Aims of this study was to investigate the association between inflammation, TKP, and suicidality in adolescent depression. Seventy-three adolescents with MDD were assessed for serum levels of int…

Alzheimer's disease-specific transcriptomic and epigenomic changes in the tryptophan catabolic pathway.

BACKGROUND: Neurodegenerative disorders, including Alzheimer's disease (AD), have been linked to alterations in tryptophan (TRP) metabolism. However, no studies to date have systematically explored changes in the TRP pathway at both transcriptional and epigenetic levels. This study aimed to investigate transcriptomic, DNA methylomic (5mC) and hydroxymethylomic (5hmC) changes within genes involved in the TRP and nicotinamide adenine dinucleotide (NAD) pathways in AD, using three independent cohorts. METHODS: DNA derived from post-mortem middle temporal gyrus (MTG) tissue from AD patients (n = …
