Expression and characterization of glutathione peroxidase of the liver fluke, Fasciola gigantica.

Glutathione peroxidase (GPx) is a key member of the family of antioxidant enzymes in trematode parasites including Fasciola spp. Because of its abundance and central role as an anti-oxidant that helps to protect parasites from damage by free radicals released from the host immune cells, it has both diagnostic as well as vaccine potential against fasciolosis. In this study, we have cloned, characterized, and detected the expression of the GPx protein in Fasciola gigantica (Fg). FgGPx (582 bp) was cloned by polymerase chain reaction (PCR) from complementary DNA (cDNA) from an adult fluke. Its pu

A synoptic review of Caryophyllaeus Gmelin, 1790 (Cestoda: Caryophyllidea), parasites of cyprinid fishes.

Tapeworms of the genus Caryophyllaeus Gmelin, 1790 (Caryophyllidea: Caryophyllaeidae), common parasites of cyprinid fishes, are reviewed and taxonomic status of 42 nominal taxa that have been placed in the genus during its long history is clarified. The following seven species occurring in the Palaearctic Region are recognised as valid: C. laticeps (Pallas, 1781), C. auriculatus (Kulakovskaya, 1961), C. balticus (Szidat, 1941) comb. n. (syn. Khawia baltica Szidat, 1941), C. brachycollis Janiszewska, 1953, C. fimbriceps Annenkova-Chlopina, 1919, C. syrdarjensis Skrjabin, 1913, and newly describ

Obstructive internal hernia caused by mesodiverticular bands in children: Two case reports and a review of the literature.

INTRODUCTION: The mesodiverticular band (MDB) is an embryologic remnant of the vitelline circulation, which carries the arterial supply to the Meckel diverticulum. In the event of an error of involution, a patent or nonpatent arterial band persists and extends from the mesentery to the apex of the antimesenteric diverticulum. This creates a snare-like opening through which bowel loops may herniate and become obstructed. This report describes 2 rare cases of small bowel occlusion owing to an internal hernia caused by a MDB. CASES: Case 1 was a 5-year-old boy who presented to our Emergency Depa

Blood flow patterns underlie developmental heart defects.

Although cardiac malformations at birth are typically associated with genetic anomalies, blood flow dynamics also play a crucial role in heart formation. However, the relationship between blood flow patterns in the early embryo and later cardiovascular malformation has not been determined. We used the chicken embryo model to quantify the extent to which anomalous blood flow patterns predict cardiac defects that resemble those in humans and found that restricting either the inflow to the heart or the outflow led to reproducible abnormalities with a dose-response type relationship between blood

Prosthenystera oonastica n. sp. (Digenea: Callodistomidae) from ictalurid catfishes in southeastern United States and molecular evidence differentiating species in the genus across Americas.

Prosthenhystera oonastica n. sp. is described as a cryptic species from the gall bladder of three ictalurid catfishes, Ictalurus punctatus (Rafinesque), Ictalurus furcatus (Valenciennes), and Pylodictis olivaris (Rafinesque), in rivers in the southeastern United States. The species was originally named by Wilmer A. Rogers in 1979 but never formally described. Material used for the description consists of two specimens of Roger's original material and ten new specimens. We found no significant morphological features that are useful for discriminating between the new species and its closest rela

New genus and species of fish blood fluke (Digenea: Aporocotylidae Odhner, 1912) from stoplight parrotfish, Sparisoma viride (Bonnaterre, 1788), (Labridae: Scarinae) in the Caribbean Sea.

Primisanguis caribbeanensis n. gen., n. sp. (Digenea: Aporocotylidae) infects the stoplight parrotfish, Sparisoma viride (Bonnaterre, 1788), (Labridae: Scarinae) in the Caribbean Sea off La Parguera (17°58'26" N, 67°02'47"W), Puerto Rico. It is most easily distinguished from other nominal aporocotylids by the combination of having an adult body that is approximately 5-10 times longer than wide, a sinistral posterolateral body protuberance, tegumental body spines that are straight (lacking recurved tip) and wrap dorsoventrally around the body margin, posterior ceca that are 2-5 times the anteri

Ultrastructure of the ovarian follicles in the placentotrophic Andean lizard of the genus Mabuya (Squamata: Scincidae).

We studied the ultrastructural organization of the ovarian follicles in a placentotrophic Andean lizard of the genus Mabuya. The oocyte of the primary follicle is surrounded by a single layer of follicle cells. During the previtellogenic stages, these cells become stratified and differentiated in three cell types: small, intermediate, and large globoid, non pyriform cells. Fluid-filled spaces arise among follicular cells in late previtellogenic follicles and provide evidence of cell lysis. In vitellogenic follicles, the follicular cells constitute a monolayered granulosa with large lacunar spa

Genetic interaction between Lef1 and Alx4 is required for early embryonic development.

Lymphoid Enhancer Factor-1 (Lef1) facilitates the assembly of transcriptional regulatory complexes and mediates nuclear responses to Wnt signals. We determined previously that the mesenchymally restricted, paired-like homeodomain protein Aristaless-like 4 (Alx4) interacts with Lef1 and together alters promoter activity of candidate genes. In order to define their overlapping functions, mice deficient for both Lef1 and Alx4 activity (Lef1-/-/Alx4lstD/lstD) were produced. Whereas embryos lacking either Lef1 or Alx4 activity remain viable up to or after birth, early embryonic lethality results wh

Altered hemodynamics in chick embryos after extraembryonic venous obstruction.

OBJECTIVE: To obtain insight into hemodynamics during abnormal cardiac development, a chick model was developed recently in which a spectrum of conotruncal anomalies, in combination with abnormal semilunar valves and/or pharyngeal arch artery malformations, was induced through extraembryonic venous obstruction (venous clip) at stage 17 (70-h incubation). METHODS: In chick embryos of stage 24 and stage 34 control (n = 8; n = 21) and with venous clip (n = 11; n = 18), we simultaneously measured dorsal flow velocities with a 20-MHz pulsed Doppler velocity meter and dorsal aortic (stage 24) and v

Ultrastructural adaptations for viviparity in the female reproductive system of gyrodactylid monogeneans.

The female reproductive system of viviparous monogeneans (Gyrodactylus and Macrogyrodactylus) has been examined using fluorescence microscopy and transmission electron microscopy. The female system is tubular, made up of a thin-walled proximal seminal receptacle/ootype and a distal uterus, separated by a complex cellular region. Both parts have a continuous syncytial cytoplasmic lining. Maturing oocytes in the seminal receptacle/ootype are in intimate contact with the receptacle lining. The uterus cytoplasmic lining completely surrounds the developing embryo, and is continuous with anterior an

A study of the anatomic features of the duct of the urachus.

The urachus is an embryonic remnant resulting from involution of the allantoic duct and the ventral cloaca. Attaching the bladder dome to the umbilicus, this duct becomes progressively obliterated during fetal life. It may subsequently persist as different variants after birth, some regarded as normal, others as pathologic, due to incomplete closure. Six pediatric cases are reported here, and the literature on the embryology and anatomic basis of the duct is discussed. The urachus is present in nearly 100% of children at birth, with several possible shapes: tubular, fusiform or funnel. It grad

Morphological studies on the fetal membranes of the normal singleton foal at term.

Of 211 consecutive thoroughbred foalings, 145 satisfied a set of criteria for normal parturition and foal viability. The fetal membranes from these and from 10 pony foalings have been systematically examined morphologically and quantitatively and the findings compared to those of other authors. Five sites on the allantochorion were consistently devoid of villi. Expulsion usually occurred with the non-villous side outermost. In 24 per cent of allantochorions the non-pregant horn was of equal length or longer than the pregnant horn. These placentae tended to be shed with the villous side outermo

In silico flux balance analysis of diclofenac-induced gonadal toxicity in zebrafish (Danio rerio).

Non-steroidal anti-inflammatory drugs (NSAIDs) are cyclooxygenase (COX) inhibitors that can cause wide-ranging metabolic disruptions. We used the in silico systems biology framework of flux balance analysis (FBA) to study the effects of diclofenac (DICL) on the gonadal metabolic physiology of adult male and female zebrafish (Danio rerio). We exposed zebrafish to 1 µM DICL for 4 days, then processed their gonads for quantitative PCR or RNA-sequencing analysis. We used up- and down-regulated gene expression changes to re-constrain the min/max catalytic bounds for reactions in a zebrafish stoichi

Cellular and molecular characteristics of blastoderm adhesion of the chick embryo.

OBJECTIVE: During early avian development, the blastoderm border must adhere to the vitelline membrane (VM) in order to spread tissue evenly. Classical studies have demonstrated that blastoderm edge tissue possesses intrinsic adhesive properties to the VM; however, the spatial extent, developmental changes, and molecular characteristics of this adherence remain poorly understood. This study aimed to characterize the molecular and functional properties of blastoderm adhesion to the VM. METHODS: Explant assays were performed using chick embryos at different developmental stages. Tissues were is

Two new species of the lecanicephalidean cestode genus Hexacanalis Perrenoud, 1931 from butterfly rays in the Persian Gulf and Gulf of Oman, Iran.

The cestode order Lecanicephalidea predominately parasitizes batoid fishes. The genus Hexacanalis (family Lecanicephalidae) is defined by the presence of six osmoregulatory canals and an antero-posteriorly U-shaped ovary. This genus currently includes three recognized species distributed across the Indian Ocean and the western Pacific Ocean. Until now, no records have documented its occurrence in the Persian Gulf or the Gulf of Oman. In this study, specimens of Hexacanalis were collected and examined for the first time from the genus Gymnura in both the Gulf of Oman and the Persian Gulf. Based

A Rare Case of Gangrenous Meckel's Diverticulum and Acute Appendicitis Presenting As Closed-Loop Obstruction in a Young Adult.

Meckel's diverticulum is the most common congenital anomaly of the gastrointestinal tract, arising from incomplete obliteration of the vitelline duct. Although often asymptomatic, it can present with life-threatening complications such as inflammation, bleeding, intestinal obstruction, and perforation. Closed-loop obstruction secondary to Meckel's diverticulum is rare, and its coexistence with acute appendicitis is exceptionally uncommon, posing a diagnostic challenge. We report the case of a 22-year-old male who presented to the emergency department with a two-day history of diffuse abdominal
