The yellow fever virus (YFV), prototype of the genus Orthoflavivirus (family Flaviviridae), contains a positive-sense RNA genome that is translated as a single polyprotein which comprises approximately 11,000 nucleotides, encoding 3411 amino acids. In recent years, a number of molecular assays have been reported for YFV RNA detection, with different levels of sensitivity and specificity. Reverse transcription Polymerase Chain Reaction (RT-PCR) assays are the most frequent methods for YFV molecular diagnostics currently. Molecular methods of diagnostics for viral genome detection offer a fast, …

Quick and accurate detection of neutralizing antibodies (nAbs) against yellow fever is essential in serodiagnosis during outbreaks for surveillance and to evaluate vaccine efficacy in population-wide studies. All of this requires serological assays that can process a large number of samples in a highly standardized format. Albeit being laborious, time-consuming, and limited in throughput, the classical plaque reduction neutralization test (PRNT) is still considered the gold standard for the detection and quantification of nAbs due to its sensitivity and specificity. Here, we report the develop…

Yellow fever vaccine contains ovalbumin, and guidelines for vaccination of egg allergic patients vary widely. We present our experience of administering yellow fever vaccine to 11 egg-allergic children, including 3 with anaphylaxis to egg, in 2 Australian tertiary pediatric hospitals. There was variation in the vaccination protocols used; however, all patients were successfully vaccinated and no serious adverse events were reported.

BACKGROUND: The yellow fever (YF) vaccination is recommended by the WHO for people traveling or living in endemic areas at risk for yellow fever infections in Africa and South America. Although the live attenuated yellow fever vaccine is a safe and efficient vaccine, rare serious adverse events after vaccination have been reported.
CASE PRESENTATION: We present the case of a 74-year-old male with multiorgan failure after yellow fever vaccination for a trip to Brazil. The patient required admission to the intensive care unit with a prolonged stay due to severe organ dysfunction. Five days afte…

In April 2016, a yellow fever outbreak was detected in Uganda. Removal of contaminating ribosomal RNA in a clinical sample improved the sensitivity of next-generation sequencing. Molecular analyses determined the Uganda yellow fever outbreak was distinct from the concurrent yellow fever outbreak in Angola, improving our understanding of yellow fever epidemiology.

In the 19th century, yellow fever thrived in the tropical, urban trade centers along the American Gulf Coast. Industrializing and populated, New Orleans and Memphis made excellent habitats for the yellow fever-carrying Aedes aegypti mosquitoes and the virulence they imparted on their victims. Known for its jaundice and black, blood-filled vomit, the malady terrorized the region for decades, sometimes claiming tens of thousands of lives during the near annual summertime outbreaks. In response to the failing medical community, a small, pronounced population of sick and healthy laypeople openly c…

BACKGROUND: Yellow fever, a mosquito-borne viral haemorrhagic fever, is one of the most lethal viral diseases. At present, an efficient vaccine for prevention is available, but may cause serious adverse events.
METHODS: The authors review the up-to-date knowledge for serious adverse events of the yellow fever vaccine (YFSAE): hypersensitivity reactions, neurotropic and viscerotropic syndromes.
RESULTS: The incidence of YFSAE has been associated with increasing age and thymus disorders, but still a number of cases with any risk factor remain unexplained, which suggests that other factors, fro…

Any person travelling in countries where yellow fever (YF) is endemic and without presenting contra-indication for the vaccination against YF may be vaccinated. This vaccination can very rarely induce a potentially lethal neurotropic or viscerotropic disease. In severely immunodeficient patients, the vaccination is contra-indicated because postvaccinal encephalitis may occur after the vaccination, due to vaccine strain pathogenecity. It is important to evaluate the general health status in elderly individuals before vaccinating because of the increased risk of viscerotropic disease in people o…

In recent years, a growing number of serious adverse events (including deaths) associated with the yellow fever (YF) vaccine has been reported. If YF vaccination were incorporated in routine programs, administered to children, the risk of deaths from this vaccine would be minimized provided that mortality of children vaccinated below 1 year were negligible. However, in affected areas the vaccine is administered to all age groups. This poses a dilemma to public health authorities - what proportion of a population subject to low risk of YF outbreaks should be vaccinated in order to minimize the …
