Reduction of Proinflammatory Effector Functions Through Remodeling of Fatty Acid Metabolism in CD8+ T Cells From Rheumatoid Arthritis Patients.

OBJECTIVE: Rheumatoid arthritis (RA) CD8+ T cells maintain their effector proinflammatory phenotype by changing their metabolism toward aerobic glycolysis. However, their massive energy and biosynthesis needs may require additional substrates other than glucose. Since systemic alterations in lipid metabolism have been reported in RA patients, we explored the role of fatty acid (FA) metabolism in CD8+ T cells to identify potential targets to curb their proinflammatory potential. METHODS: The expression of FA metabolism-related genes was analyzed for total CD8+ T cells and CD8+ T cell subsets i

Ferulic acid improves growth performance and lipid metabolism and bile acid level partially by activating the PPAR and CYP7A1 pathways in broilers.

BACKGROUND: High-energy and high-protein diets promote excessive fat deposition in broilers, increasing their susceptibility to metabolic disorders. Ferulic acid (FA) is a phenolic acid with multiple biological functions and has potential application value. The present study evaluated the effects of dietary FA on the growth performance, lipid metabolism, bile acid synthesis and gut microbiota in broilers. In total, 640 male broilers were allocated to five groups at 1 day of age, and fed with a basal diet supplemented with 0 (control group), 50, 100, 150 or 200 mg kg-1 FA for 42 days. RESULTS:

Involvement of receptor-interacting protein 140 in estrogen-mediated osteoclasts differentiation, apoptosis, and bone resorption.

Estrogen withdrawal following menopause results in an increase of osteoclasts formation and bone resorption, which is one of the most important mechanisms of postmenopausal osteoporosis. Recently, growing evidence has suggested that receptor-interacting protein 140 was implicated in estrogen-regulated metabolic disease, including fat metabolism and lipid metabolism. However, little is known regarding the role of receptor-interacting protein 140 in the regulation of bone metabolic by estrogen. In the present study, Western blotting disclosed that estrogen brings a significant increasing express

Novel insights into FAS defects underlying autoimmune lymphoproliferative syndrome revealed by studies in consanguineous patients.

Autoimmune lymphoproliferative syndrome (ALPS) is a primary immunodeficiency disease due to impaired Fas-Fas ligand apoptotic pathway. It is characterized by chronic nonmalignant, noninfectious lymphadenopathy and/or splenomegaly associated with autoimmune manifestations primarily directed against blood cells. Herein, we review the heterogeneous ALPS molecular bases and discuss recent findings revealed by the study of consanguineous patients. Indeed, this peculiar genetic background favored the identification of a novel form of AR ALPS-FAS associated with normal or residual protein expression,

Autoimmune lymphoproliferative syndrome (ALPS).

In patients with ALPS, defective homeostasis of lymphocytes is reflected in abnormal accumulation of lymphocytes, leading to lymphadenopathy, (hepato)splenomegaly and hypersplenism, autoimmunity due to a failure to remove autoreactive lymphocytes, and inappropriate survival of lymphocytes associated with an increased occurrence of lymphoma. Several of the laboratory findings are unique for ALPS and reflect defective Fas-mediated apoptosis and abnormal immune regulation. Much has been learned about the molecular mechanisms that underlie defective Fas-mediated apoptosis and the complex relations

[Apoptosis in the cryptorchidism. Role of the FAS protein in it's modulation].

The cellular degeneration, present at the criptorchidism leads to a malfunction in the spermatogenosis and it's due to cellular apoptosis (programed cellular death). This process is turned on and modulated by different ways. One of then is mediated by Ag-Ac reactions and a protein called FAS seems to have an important role in it's modulation. We studied the relation between FAS an the increased apoptosis in undescended testics. Testicular biopsies were done in criptorchid patients during orchidopexy and FAS was measured by immune-techniques. Our results seem to dismiss FAS as a modulation apop

Pathogenesis of primary biliary cirrhosis: CD95-induced apoptosis at last?

Apoptosis is a basic mechanism involved both in maintaining tissue homeostasis by elimination of senescent or potentially harmful cells and in the regulation of immune responses. If not properly regulated, however, it may lead to serious tissue damage. CD95(Fas/APO-1) is a surface receptor that mediates apoptosis upon oligomerization by its ligand, CD95L. The CD95/CD95L-system is one of the major effectors of cytotoxicity in inflammation with implications for both the prevention and the pathogenesis of autoimmune diseases. In primary biliary cirrhosis (PBC), an autoimmune disease, apoptosis ha

Fas-mediated pathway and apoptosis in normal, hyperplastic, and neoplastic endometrium.

OBJECTIVES: Abnormalities in the control of cell proliferation and apoptosis have been suggested to contribute to the development and progression of neoplasia. There are at least two pathways that activate apoptosis. The first is a mitochondria-dependent route governed by bcl-2 family proteins. The second is a parallel mechanism which involves the activation of a group of tumor necrosis factor (TNF) receptors, such as Fas. METHODS: The aim of this study was to examine the distribution and interrelation between the expression patterns of apoptosis-related proteins such as Fas, caspase-3 (CPP32

Therapeutic Nanoplatform Integrating Photothermal Effect and Silver Nanoparticles for the Treatment of Intracellular Mycobacterium tuberculosis.

The global challenge of tuberculosis (TB), particularly in the context of rising drug resistance and intracellular bacterial persistence, highlights the need for innovative therapeutic strategies. In this study, we develop a multifunctional nanoplatform (CuS-Ag@FA) that integrates near-infrared (NIR)-responsive photothermal CuS, antimicrobial Ag nanoparticles (Ag NPs), and folate-receptor-mediated macrophage targeting for enhanced intracellular Mycobacterium tuberculosis (M.tb) clearance. Our work represents the first FA-functionalized CuS-Ag nanoplatform designed for dual-targeting of M.tb-in

Control of grip force during restraint of an object held between finger and thumb: responses of cutaneous afferents from the digits.

Unexpected pulling and pushing loads exerted by an object held with a precision grip evoke automatic and graded increases in the grip force (normal to the grip surfaces) that prevent escape of the object; unloading elicits a decrease in grip force. Anesthesia of the digital nerves has shown that these grip reactions depend on sensory signals from the digits. In the present study we assessed the capacity of tactile afferents from the digits to trigger and scale the evoked grip responses. Using tungsten microelectrodes inserted percutaneously into the median nerve of awake human subjects, unitar

The expression level of VEGFR2 regulates mechanotransduction, tumor growth and metastasis of high grade serous ovarian cancer cells.

Recent data shows that alterations in the expression and/or activation of the vascular endothelial growth factor receptor 2 (VEGFR2) in high grade serous ovarian cancer (HGSOC) modulate tumor progression. However, controversial results have been obtained, showing that in some cases VEGFR2 inhibition can promote tumorigenesis and metastasis. Thus, it is urgent to better define the role of the VEGF/VEGFR2 system to understand/predict the effects of its inhibitors administered as anti-angiogenic in HGSOC. Here, we modulated the expression levels of VEGFR2 and analyzed the effects in two cellular

Myofibroblasts persist through immune privilege mechanisms to mediate oral submucous fibrosis: Uncovering the pathogenesis.

Immune privilege is the ability to tolerate foreign antigens without eliciting an inflammatory immune response. Several mechanisms explain a structure's immune privilege status, which is regulated by innate and adaptive immune responses. The role of myofibroblasts in perpetuating fibrosis by acquiring an immune privileged phenotype against the backdrop of oral submucous fibrosis (OSF) is evolving. Myofibroblasts persist through the Fas/FasL autocrine pathway and induce apoptosis in epithelial cells, explaining the juxtaposition of apoptotic cells in areas of fibrosis. However, increased matrix

Zinc supplementation inhibits hepatic apoptosis in mice subjected to a long-term ethanol exposure.

Hepatocyte apoptosis has been documented in both clinical and experimental alcoholic liver disease. This study was undertaken to examine the effect of dietary zinc supplementation on hepatic apoptosis in mice subjected to a long-term ethanol exposure. Male adult 129S6 mice fed an ethanol-containing liquid diet for 6 months developed hepatitis, as indicated by neutrophil infiltration and elevation of hepatic keratinocyte chemoattractant (KC) and monocyte chemoattractant protein-1 (MCP-1) levels. Apoptotic cell death was detected in ethanol-exposed mice by a terminal deoxynucleotidyl transferase

Increased osteoblast apoptosis in apert craniosynostosis: role of protein kinase C and interleukin-1.

Apert syndrome is an autosomal dominant disorder characterized by premature cranial ossification resulting from fibroblast growth factor receptor-2 (FGFR-2)-activating mutations. We have studied the effects of the prominent S252W FGFR-2 Apert mutation on apoptosis and the underlying mechanisms in human mutant osteoblasts. In vivo analysis of terminal deoxynucleotidyl transferase-mediated nick-end labeling revealed premature apoptosis of mature osteoblasts and osteocytes in the Apert suture compared to normal coronal suture. In vitro, mutant osteoblasts showed increased apoptosis, as demonstrat

Acceleration of lpr lymphoproliferative and autoimmune disease by transgenic protein kinase CK2 alpha.

MRL-lpr/lpr mice have a Fas receptor mutation that leads to abnormalities of apoptosis, lymphoproliferation, and a lupus-like autoimmune disease associated with the production of autoantibodies. Other than Fas pathway defects, little is known about molecular abnormalities that predispose to autoimmunity. Protein kinase CK2 (also termed casein kinase II), a serine-threonine protein kinase whose targets include many critical regulators of cellular growth, is highly expressed in a lymphoproliferative disease of cattle and in many human cancers. Overexpression of the CK2alpha catalytic subunit in …

Preimplantation maternal stress impairs embryo development by inducing oviductal apoptosis with activation of the Fas system.

STUDY QUESTION: What are the mechanisms by which the preimplantation restraint stress (PIRS) impairs embryo development and pregnancy outcome? SUMMARY ANSWER: PIRS impairs embryo development by triggering apoptosis in mouse oviducts and embryos, and this involves activation of the Fas system. WHAT IS KNOWN ALREADY: Although it is known that the early stages of pregnancy are more vulnerable than later stages to prenatalstress, studies on the effect of preimplantation stress on embryo developmentare limited. Furthermore, the mechanisms by which psychological stress impairs embryo development ar

Staphylococcal enterotoxin B induces potent cytotoxic activity by intraepithelial lymphocytes.

In food poisoning, Staphylococcus aureus secretes staphylococcal enterotoxin B (SEB), a superantigen that causes intense T-cell proliferation and cytotoxicity. The effects of SEB on lytic activity by human intestinal intraepithelial lymphocytes (IEL) were investigated. Jejunal IEL, from morbidly obese individuals undergoing gastric bypass operations, were tested for SEB-induced cytotoxicity against C1R B-lymphoblastoid cells, HT-29 adenocarcinoma cells, or CD1d-transfected cells using the 51Cr-release assay. Fas and Fas ligand expression were detected by immunofluorescence and flow cytometry a
