Comparative effects of combined omeprazole and dose-escalated striatin therapy versus monotherapy on VEGF, ITF, gastric ulcer area, epithelial thickness, and inflammatory response in rats with aspirin-induced gastric injury.

INTRODUCTION: Aspirin-induced gastritis results from reduced prostaglandin synthesis and increased oxidative stress, compromising gastric mucosal integrity. While omeprazole effectively suppresses gastric acid secretion, it does not directly promote mucosal regeneration. Striatin, a bioactive fraction of Channa striata, has anti-inflammatory and regenerative properties and may enhance mucosal healing. PURPOSE: This study aimed to evaluate whether adjunctive striatin at escalating doses enhances the therapeutic effect of omeprazole in aspirin-induced gastric injury. METHODS: A controlled expe

Isodeoxyelephantopin mitigates DSS-induced ulcerative colitis by suppressing IL-1β-driven inflammation via the TXNIP/NLRP3 axis.

BACKGROUND: Isodeoxyelephantopin (IDET) is a sesquiterpene lactone isolated from traditional herb Elephantopus scaber, which is known for its anti-inflammatory activities. While our previous study demonstrated that IDET inhibits NLRP3 expression in an acute peritonitis model, its therapeutic potential in chronic inflammatory diseases such as ulcerative colitis (UC), as well as the underlying mechanisms involving inflammasome signaling, have not yet been fully elucidated. PURPOSE: This research was designed to explain the protective capacity of IDET in UC and to clarify how IDET modulates IL-1

Protocadherin 20 Is a POU Class 2 Homeobox 3 Target Gene Required for Proper Tuft Cell Microvillus Organization.

BACKGROUND & AIMS: Tuft cells play protective roles in infection, inflammation, and tumorigenesis through the secretion of cytokines and eicosanoids. Tuft cells are known for their tall, blunt microvilli, thought to be analogous to mechanosensory hair cell stereocilia; however, a functional role for the microvillar apparatus has not been identified. POU Class 2 Homeobox 3 (POU2F3) is the master regulator transcription factor for tuft cells, yet how POU2F3 drives formation of this unique structure is unknown. Here, we aimed to identify POU2F3 target genes and commonalities between tuft and hair

Δ9-Tetrahydrocannabinol exposure shifts eosinophil and macrophage transcriptional programs towards an anti-inflammatory phenotype in helminth infection.

Cannabis use is increasing globally, yet the immunological effects of Δ9-tetrahydrocannabinol (THC), the main intoxicating component of cannabis, remain incompletely understood. Given prior evidence that endocannabinoid signaling influences helminth immunity and type 2 inflammation, we investigated how sustained THC exposure alters immune responses to the helminth Nippostrongylus brasiliensis (Nb), which infects the lung and small intestine of mice. C57BL/6J mice were treated with THC (5 mg/kg/day) or vehicle for 14 days prior to helminth infection and assessed for parasite burden, innate immu

The Metabolic Revolution in Dermatology: Obesity, Insulin Resistance, and Ultra-Processed Diets as Drivers of Skin Inflammation.

Inflammatory dermatoses are increasingly linked to systemic metabolic factors. Obesity and insulin resistance create a pro-inflammatory milieu that affects the skin. Adipose tissue functions as an endocrine organ secreting adipokines and cytokines that drive chronic inflammation with notable skewing toward T-helper 1 (Th1)/Th17 signaling. Hyperinsulinemia, elevated insulin-like growth factor-1 promote keratinocyte proliferation, sebum production, and autoinflammation, contributing to a multitude of skin diseases including hidradenitis suppurativa, acne, psoriasis, atopic dermatitis (AD), acant

The gut microbiota in Salmonella Typhi infection and translocation: Mechanisms of colonization resistance, pathogen subversion, and prospects for microecological intervention.

The systemic establishment of the strictly human-adapted pathogen Salmonella Typhi critically depends on functional interplay between its virulence effectors and the host gut microbiome. Organized within a "colonization-toxicity-immunity- microecological intervention" framework, this review synthesizes recent molecular and multi-omics evidence to delineate key host-pathogen-microbiota dynamics. During colonization, a healthy gut microbiota exerts multi-layered colonization resistance through nutrient and niche competition, as well as via microbial metabolites such as short-chain fatty acids an

Bitter Taste Receptors in Bacterial Infections and Innate Immunity.

BACKGROUND: Bitter taste receptors (TAS2Rs), originally identified for their role in gustation, are now recognized for their functions in extraoral tissues, particularly in innate immune responses. TAS2Rs detect bacterial quorum sensing molecules (QSMs) and other metabolites, enabling the host to sense and respond to pathogenic threats across mucosal surfaces. OBJECTIVE: This review synthesizes current knowledge of TAS2Rs in the context of bacterial infection, emphasizing their mechanisms of immune modulation, genetic polymorphisms, tissue-specific expression, and therapeutic potential. Relationship between gastric mucosal atrophy by endoscopy and non-ampullary duodenal epithelial tumors.

BACKGROUND: The pathogenesis of non-ampullary duodenal epithelial tumors (NADETs) is not fully understood. NADETs that express gastric-type mucin phenotypes (G-NADETs) are noteworthy because of their high malignancy. Gastric foveolar metaplasia, from which G-NADETs originate, protects the duodenal mucosa from gastric acidity. As gastric acid secretion is affected by endoscopic gastric mucosal atrophy (EGMA), we hypothesized that EGMA would be associated with G-NADETs. AIM: To evaluate the association between EGMA and the occurrence of G-NADETs. METHODS: This cross-sectional retrospective stu

Exploring the immunomodulatory mechanism of total glucosides of paeony on Sjögren's syndrome dry eye disease based on the "gut-eye axis" pathway.

PURPOSE: Sjögren's syndrome-related dry eye disease (SS-DED) is an autoimmune disorder characterized by ocular surface inflammation and lacrimal gland dysfunction, with limited clinical treatment options currently available. METHODS: Based on the traditional Chinese medicine theory of "the liver opens into the eyes" and the modern "gut-eye axis" hypothesis. This study investigated the therapeutic effects and immunomodulatory mechanisms of total glucosides of paeony (TGP) using a NOD mouse model. RESULTS: The results showed that TGP significantly increased tear secretion in SS mice, alleviate

Loss of sialic acid side-chain O-acetylation exacerbates colitis.

Sialic acids (Sias) are a diverse family of nine-carbon backbone monosaccharides occupying terminal positions on cell surface and secreted glycans and are abundant at mucosal surfaces. Sias can be modified with O-acetyl esters on the side chain (C7 to C9) hydroxyls. Structural analysis and functional studies of these modifications are challenging due to chemical lability and variable resistance to sialidases. For in-depth analysis of the expression and functions of O-acetyl Sia modifications, we used a unique set of sialoglycan-recognizing probes, HPLC analysis of DMB-derivatized Sias and mice

Telotristat ethyl affects tumour-fibroblast crosstalk in small intestinal neuroendocrine tumours.

Small intestinal neuroendocrine tumours (SI-NETs) are associated with mesenteric fibrosis, which causes significant morbidity and mortality. Telotristat ethyl was developed to treat carcinoid syndrome in SI-NET patients. Recent studies indicated telotristat ethyl could have anti-tumour activity; however, the mechanism remains unclear. This study aimed to investigate the effects of telotristat ethyl on SI-NET-fibroblast crosstalk in tumour progression and mesenteric fibrosis. A co-culture paracrine model with GOT1 (tumour) cells and LX2 (stromal) cells was optimized. Cells were treated with con

An oral norovirus vaccine tablet was safe and elicited mucosal immunity in older adults in a phase 1b clinical trial.

Norovirus is a leading cause of acute gastroenteritis globally, with infections in older adults associated with heightened severity and increased risk of mortality. Currently, no licensed vaccines are available to prevent norovirus infection. We developed an orally administered vaccine tablet (VXA-G1.1-NN) that delivers a nonreplicating adenoviral vector expressing norovirus GI.1 major capsid protein VP1 to the small intestine. Here, we report safety and immunogenicity results of a randomized, double-blind, placebo-controlled clinical trial (NCT04854746) that investigated the oral administrati

A recombinant adenovirus-vectored PEDV vaccine co-expressing S1 and N proteins enhances mucosal immunity and confers protection in piglets.

Porcine epidemic diarrhea virus (PEDV) is a highly contagious enteric coronavirus that causes severe diarrhea and high mortality in neonatal piglets. Effective control requires systemic, mucosal, and T cell-mediated immune responses, highlighting the need for a safe, effective, and versatile vaccine platform. In this study, a recombinant adenovirus type 5-based vaccine co-expressing the PEDV S1 and N proteins (rAd5-S1-N) was developed, and its immunogenicity and protective efficacy were evaluated in mice and piglets. Intranasal immunization of BALB/c mice with rAd5-S1-N induced strong and sust

Core microbiome-associated proteins associated with ulcerative colitis interact with cytokines for synergistic or antagonistic effects on gut bacteria.

Inflammatory bowel disease (IBD), including Crohn's disease (CD) and ulcerative colitis (UC), is associated with a loss or an imbalance of host-microorganism interactions. However, such interactions at protein levels remain largely unknown. Here, we applied a depletion-assisted metaproteomics approach to obtain in-depth host-microbiome association networks of IBD, where the core host proteins shifted from those maintaining mucosal homeostasis in controls to those involved in inflammation, proteolysis, and intestinal barrier in IBD. Microbial nodes such as short-chain fatty-acid producer-relate

Novel insight into the feed conversion ratio in laying hens and construction of its prediction model.

Feed efficiency (FE) is an important economic factor in poultry production, and feed conversion ratio (FCR) is one of the most widely used measures of FE. Factors associated with FCR include genetics, the environment, and other factors. However, the mechanisms responsible for FCR in chickens are still less well appreciated. In this study, we examined the pattern changes of FCR, then delved into understanding the mechanisms behind these variations from both genetic and environmental perspectives. Most interestingly, the FCR at the front section of henhouse exhibited the lowest value. Further in

Peripheral GABAA receptors - Physiological relevance and therapeutic implications.

The role of γ- aminobutyric acid (GABA) and GABAA receptors is not only essential for neurotransmission in the central nervous system (CNS), but they are also involved in communication in various peripheral tissues such as the pancreas, liver, kidney, gastrointestinal tract, trachea, immune cells and blood vessels. GABAA receptors located outside the CNS ("peripheral GABAA receptors") enable both neuronal and non-neuronal GABA-ergic signaling in various physiological processes and are generally thought to have similar properties to the extrasynaptic receptors in the CNS. By activating these pe
