Single-nucleus and spatial transcriptomics reveal intestinal cellular heterogeneity, differentiation, and cell communication mechanisms in SAP-induced intestinal injury.

BACKGROUND: Acute pancreatitis (AP) ranges from mild to severe, and severe disease frequently causes multi-organ damage, with intestinal injury being a major complication. The mechanisms underlying SAP-induced intestinal injury remain unclear, particularly regarding spatial cellular reorganization and functional interactions. METHODS: This study constructed a severe acute pancreatitis (SAP) rat model and employed single-nucleus RNA sequencing (snRNA-seq) and spatial transcriptome sequencing (stRNA-seq) technologies to systematically analyze the dynamic changes in intestinal cellular compositi

A case report: an unusual case of ulcerative intestinal IgG4-related disease with clinical features overlapping with probable intestinal Behcet's disease.

BACKGROUND: IgG4-related disease (IgG4-RD) is a systemic fibroinflammatory condition characterized by IgG4-positive plasma cell infiltration, typically presenting with organ enlargement or tumor-like masses. Luminal gastrointestinal involvement is rare, and ulcerative lesions are even less common. Overlap syndromes of IgG4-RD with other immune disorders have been reported, but overlap of its clinical features with those of intestinal Behcet's disease has not been documented. CASE PRESENTATION: A 62-year-old man with a history of recurrent oral ulcers presented with sudden hematochezia and hyp

Maternal intestinal and placental mitochondrial dysfunction, autophagy, and ferroptosis involving intestinal microbiota by gut microbiota transplantation from sheep to mice†.

Exposure to testosterone (T) in pregnant ewes resulted in placental dysfunction and fetal growth restriction (FGR). However, the impact of T on gut microbiota and its contribution to exacerbating intestinal and placental pathologies remains uncharacterized. Pregnant sheep received intramuscular injections of 100 mg T propionate or a control vehicle. To examine the gut microbiota' s role in T-induced FGR, gut microbiota transplantation (GMT) was conducted from T-exposed and control ewes into antibiotic-treated pregnant mice. The findings demonstrated that T exposure exacerbated mitochondrial im

Arctic Fox-Derived Lactiplantibacillus plantarum J3 Alleviates Colitis in Mice in Association with Strengthening of the Intestinal Barrier and Reshaping of the Intestinal Flora.

Probiotic interventions for inflammatory bowel disease (IBD) have become a hot research topic in this field. However, no previous studies have investigated whether fox-derived probiotics exert therapeutic effects against colitis. The present investigation evaluated the effects of Lactiplantibacillus plantarum (syn. Lactobacillus plantarum) J3, sourced from the intestines of healthy Arctic foxes, on colitis. In an in vitro LPS-stimulated Caco-2 cell model, strain J3 exhibited prominent anti-inflammatory and antioxidant capacities, indicating potential to preserve intestinal barrier integrity. F…

Deep-Infiltrating Endometriosis Causing Acute Mechanical Intestinal Obstruction Without Intestinal Invasion: A Case Report with Diagnostic and Surgical Insights.

BACKGROUND: Endometriosis is a chronic, estrogen-dependent disorder that may extend beyond the pelvis to involve the gastrointestinal tract, most commonly the rectosigmoid and, less frequently, the small bowel. Although often asymptomatic, such lesions may rarely manifest as acute bowel obstruction. CASE: We report a 42-year-old woman who presented with small bowel ileus caused by deep-infiltrating endometriosis (DIE). Imaging revealed a right ovarian endometrioma with severe adhesions resulting in a distal ileal transition point. After partial decompression with conservative treatment, lapar

Dissecting the differentiation origins of intestinal metaplasia and early intestinal-type gastric cancer in gastric antrum by single-cell RNA profiling.

Intestinal-type gastric cancer (IGC) is associated with a multi-step carcinogenic process, comprising non-atrophic gastritis, chronic atrophic gastritis, intestinal metaplasia, and gastric dysplasia. The risk of developing IGC gradually increases as the disease progresses. However, the origin of cell differentiation and its carcinogenic potential in different stages of gastric disease remains poorly understood. To address this issue, we analyzed the differentiation trajectory of epithelial cells in different disease stages from gastric antrum biopsies in patients with precancerous lesions and

Intestinal Ischemia Secondary to Ladd's Bands Obstruction Associated With Intestinal Malrotation in a One-Year-Old Pediatric Patient: A Case Report.

Intestinal malrotation is a congenital anomaly resulting from an alteration in the rotation and fixation of the midgut during embryonic development. Ladd's bands are abnormal peritoneal adhesions that can cause extrinsic compression of the bowel, obstruction, and vascular compromise, leading to intestinal ischemia. Although the diagnosis is typically made in the neonatal period, late presentations exist, posing a diagnostic and therapeutic challenge. We describe the case of a previously healthy one-year-old male patient who was admitted for feeding intolerance, accompanied by vomiting, progres

Sodium butyrate stimulates intestinal health and immunity by reshaping the intestinal microbiota and metabolite profile in big-belly seahorses (Hippocampus abdominalis).

This study examined the effects of sodium butyrate (SB) on growth, intestinal health and immunity of big-belly seahorses (Hippocampus abdominalis) suffering frozen diet-induced enteritis and stress. Total 750 seahorses were randomly assigned to five groups in triplicate, and were fed frozen mysids mixed with SB at 0, 1.25, 2.5, 5 and 10 mg/L (based on culture water volume) for 8 weeks, followed by an Edwardsiella tarda challenge. Results demonstrated that the SB1.25 group had a significantly higher specific growth rate, hepatic T-SOD and CAT activities, and lower MDA content than the SB0 group

Intestinal Microecology Dysbiosis, Cultivable Pathogen Composition, and Antimicrobial Susceptibility Profiles Associated with Intestinal Exposure and Ocular Opacity in Hybrid Porcupinefish.

Intestinal exposure and ocular opacity occur in cultured hybrid porcupinefish, but the associated pathological lesions and gut microecological changes remain poorly characterized. Hybrid porcupinefish were assigned to a healthy control group (CT-CD-JK), an intestinal exposure group (CT-CD-FB), or an ocular opacity group (CT-YQ-FB). Gut microbiota, histopathology, cultivable bacteria, and antimicrobial susceptibility were assessed using 16S rRNA sequencing, hematoxylin and eosin (H&E) staining, bacterial isolation and identification, and disk diffusion testing. Diseased fish showed evident inte

Common mechanisms of Gut microbe-based strategies for the treatment of intestine-related diseases: based on multi-target interactions with the intestinal barrier.

The concurrent occurrence and exacerbation of multiple diseases, including geriatric diseases and chronic diseases, impose a heavy burden on human health and medical expenses. Clarifying the common mechanisms of related multifarious diseases and developing preventive and therapeutic strategies with synergistic effects for multiple diseases are of great significance in alleviating the burden on the medical system and reducing patients' burden of drug metabolism. Recent studies have revealed that gut microbiota disorders and intestinal barrier damage, which consequently cause metabolic and immun

[Hypaphorine alleviates Crohn's disease-like colitis in mice by inhibiting intestinal epithelial inflammatory response and protecting intestinal barrier function].

OBJECTIVES: To investigate the effect of hypaphorine (HYP) on Crohn's disease (CD)‑like colitis in mice and its molecular mechanism. METHODS: Thirty male C57BL/6J mice were equally randomized into WT, TNBS, and HYP groups, and in the latter two groups, mouse models of CD-like colitis were established using TNBS with daily gavage of 15 mg/kg HYP or an equivalent volume of saline. The treatment efficacy was evaluated by assessing the disease activity index (DAI), body weight changes, colon length and histopathology. The effect of HYP was also tested in a LPS-stimulated Caco-2 cell model mimicki

Intestinal Neutral Ceramidase Deficiency Triggers Regulatory T Cell Response via Gd3 to Protect the Host from Intestinal Inflammation.

Sphingolipids play a crucial role in gut inflammation. Neutral ceramidase (NcDase) serves as a pivotal regulator of ceramide, the central intermediate in sphingolipid metabolism. The contribution of intestinal epithelial cells (IEC) NcDase to colitis is not well understood. Here, a protective mechanism by which IEC NcDase deficiency (Asah2ΔIEC) and its-related gangliosides prevent dextran sulfate sodium (DSS)-induced colitis in mice is described. Asah2ΔIEC mice display reduced susceptibility to DSS-induced colitis and increase regulatory T (Treg) cells compared to Asah2fl/fl littermates. Delet

Tibetan Tea Alleviates the Intestinal Dysfunction in Sleep-Deprived Mice Through Regulating Oxidative Stress and Inflammation-Related Intestinal Metabolisms.

Sleep deprivation (SD) disrupts intestinal homeostasis through excessive reactive oxygen species (ROS) accumulation. Tibetan tea is a potential dietary intervention for inflammation, it's effect on SD-induced intestinal inflammation remains unclear. This study investigates the alleviating effects of Tibetan tea water-soluble extract (TTE) on intestinal dysfunction in SD mice. After TTE supplementation, the physiological activity, inflammatory cytokines, and oxidative stress levels were assessed in SD-induced intestinal dysfunction mice. SD increased ROS levels and pro-inflammatory cytokines in
