Structural insights into the activation mechanism of the human metabolite receptor HCAR1.

Hydroxycarboxylic acid receptor 1 (HCAR1) is a class A G protein-coupled receptor (GPCR) that is activated by the endogenous metabolite l-lactate and that plays an important role in various metabolic and inflammatory disorders. HCAR1 uses distinct ligand recognition and self-activation mechanisms to mediate specific pathophysiological functions through Gαi/o and β-arrestin signaling pathways. To support effective drug development targeting HCAR1, we investigated ligand recognition and activation mechanisms through cryo-electron microscopy (cryo-EM) structures of the HCAR1-Gαi1 complex in the a

Research Progress on the Role of Traditional Chinese Medicine in Regulating Ferroptosis in Cardiovascular Diseases.

Ferroptosis is an iron-dependent form of regulated cell death driven by lipid peroxidation, playing a critical role in the pathogenesis of various cardiomyopathies, including hypertrophic, dilated, diabetic, ischemic, doxorubicin-induced, and septic cardiomyopathy, as well as myocardial ischemia-reperfusion injury. This article provides a comprehensive narrative review of the molecular mechanisms of ferroptosis-centered on dysregulation of the GPX4/System Xc- axis, iron metabolism, and lipid metabolism-and its role in cardiovascular diseases, with a specific focus on the cardioprotective effec

The impact of long-term feeding with curcuminoids phospholipids enriched diet on disease progression of fALS.

Amyotrophic lateral sclerosis (ALS) is a debilitating and fatal neurodegenerative disease characterised by the progressive loss of motor functions affecting both upper and lower motor neurons. Although considered multifactorial with an unclear aetiology, it is believed that the interplay between genetic and environmental factors, with neuroinflammation playing a key role in disease progression, contributes to its development. There is currently no effective treatment for ALS. Curcumin has been recently highlighted for its potential therapeutic role in treating neurodegenerative diseases. Curcu

Whole Genome Sequencing Reveals Antibiotic Resistance Heterogeneity and Metabolic Signatures of Cutibacterium acnes in Atopic Dermatitis.

Cutibacterium acnes (C. acnes), the most abundant microbe of the skin microbiota, has been reported playing a protective role in atopic dermatitis (AD), in contrast to its proinflammatory role in acne. While antibiotic resistance in C. acnes has been extensively studied in acne, little is known about its characteristics in AD. Here, we performed an integrated genomic and functional analysis of 405 AD-derived C. acnes isolates using whole genome sequencing (WGS) and antimicrobial susceptibility testing. The result showed that antibiotic resistance genes (ARGs) were highly clustered within speci

Extracellular vesicles derived from M1 macrophages deliver miR-515-5p to suppress trophoblast migration and invasion by targeting YAP in preeclampsia.

Trophoblast migration and invasion are critical processes in the pathogenesis of preeclampsia (PE), with maternal-fetal cellular communication playing a key role in their regulation. Among the immune components at the maternal-fetal interface, M1-like macrophages are known to influence trophoblast behavior, however, the specific underlying mechanisms remain unclear. Here, we reveal a novel pathway through which M1-like macrophages regulate trophoblast function through extracellular vesicle (EV)-mediated communication. We demonstrated that M1-derived extracellular vesicles (M1-EVs) inhibit trop

Hrd1 alleviates fatty liver ischemia-reperfusion injury by regulating the phenotypic polarization of Kupffer cells.

To address the persistent shortage of donor livers for transplantation, steatotic grafts are increasingly being accepted as marginal allografts. However, fatty liver is highly susceptible to ischemia-reperfusion injury (IRI), leading to high post-transplant graft dysfunction rates, with no effective alleviation methods available to date. Our previous studies indicated that activated Kupffer cells (KCs) undergo M1 proinflammatory polarization to exacerbate fatty liver IRI, yet the underlying molecular circuitry remains largely undefined. In this study, the IRI in fatty liver mice was found to b

Reprogramming Mammal Müller Glial Cells for Retinal Neuroregeneration.

Müller glia (MG) are the predominant nonneuronal cells of the vertebrate retina, playing an integral role in retinal homeostasis and photopic signal transmission. In lower vertebrates, MG retain robust regenerative potential, acting as a source of neuronal progenitors following injury. In contrast, mammalian MG primarily respond to damage with gliosis, a process that provides initial neuroprotection but ultimately restricts their neurogenic capacity. This limitation is especially consequential in humans, where genetic disorders and age-related diseases, such as diabetic retinopathy, glaucoma,

Three carboxylesterase genes are involved in spinosad resistance in Frankliniella occidentalis (Pergande).

Carboxylesterases (CarEs) are key enzymes in insect detoxification and metabolism, playing a crucial role in resistance development to various insecticides. In this study, CarE activity was significantly higher in spinosad-resistant strains of Frankliniella occidentalis (NIL-R) than in susceptible strains (Ivf03), suggesting a potential role of CarEs in spinosad resistance. RNA-seq and quantitative PCR (qPCR) validation confirmed that four CarE genes were significantly upregulated in NIL-R. RNA interference (RNAi) validated that three of these genes FoB1, FoB1L, and FoFE4 are directly involved

Expression of TAM receptors and their ligands in unexplained recurrent pregnancy loss.

Recurrent pregnancy loss (RPL) is clinically defined as two or more pregnancy failures before 20-24 weeks of gestation. Over 50% of RPL cases have no identified etiology and are classified as unexplained recurrent pregnancy loss (URPL). The TAM receptor tyrosine kinase (RTK) family, including TYRO3, AXL, MERTK, GAS6, and PROS1, plays critical roles in suppressing excessive inflammation and apoptosis. Although increased serum levels of these molecules have been observed in pregnancy complications such as preeclampsia, their role in URPL remains unexplored. We hypothesized that TAM receptors con

The Role of Working Memory in Deaf Adults' Visual Word Recognition: Evidence From Dual-Task Paradigm.

PURPOSE: Although working memory (WM) is crucial for reading comprehension, evidence regarding its role during reading in deaf individuals is ambiguous and is primarily based on correlational findings. This study aimed to investigate whether WM influences the real-time lexical processing of deaf adults and, if so, whether this influence varies across different WM domains (e.g., verbal vs. visuospatial). METHOD: The present study employed a dual-task paradigm to examine the role of WM in word recognition among individuals with hearing loss. Experiment 1 manipulated verbal WM load during visual

Action on commercial gambling-a local government case study from Great Britain.

Commercial gambling is increasingly recognized as a public health issue, with local government playing a critical role in mitigating its harms. The challenges that public health teams in local government face when there are applications for new gambling premises licences include no mandatory regulatory role; lack of evidence to draw upon and an 'aim to permit' principal entrenched in legislation. This case study examines a city council's refusal to grant a gambling premises licence, and the subsequent legal appeal, which was dismissed in February 2025. Despite the Gambling Act 2005's 'aim to p

Impact of the brainstem in normal and impaired human movement control.

The corticospinal tract represents a highly developed neural system that dominates motor control in humans. This allows the performance of skilled finger movements, such as playing the piano. In contrast, according to preclinical studies, automatically performed complex movements, such as locomotion or cooperative forelimb movements, appear to be controlled mainly by brainstem motor centres. Only in recent years, with the advancement of new methodologies, has evidence emerged that limb coordination in humans is also based on neural limb coupling mediated by brainstem motor centres. This neural

A Novel Loss-of-Function Variant in TGFBI Associated with Non-Syndromic Orofacial Clefts in a Chinese Pedigree.

Background/Objectives: Non-syndromic orofacial clefts (NSOFCs) are among the most common congenital craniofacial anomalies, with genetic factors playing a major role in their etiology. Transforming growth factor-β-induced (TGFBI) protein is an extracellular matrix protein implicated in cell adhesion and apoptosis. Although experimental studies have implicated TGFBI in palatal fusion, its contribution to human NSOFCs remains unclear. This study investigated the genetic basis of hereditary NSOFCs in a Chinese family. The functional consequences of the identified TGFBI variant were subsequently e

Could excessive zinc supplementation during pregnancy cause menkes disease? A hypothesis worth investigating.

BACKGROUND: Copper is an essential micronutrient critical for fetal neurodevelopment, haematopoiesis, angiogenesis, and immune function, with maternal transfer-particularly in the third trimester-playing a key role in establishing fetal copper stores. Disruption of this process, due to genetic defects or micronutrient imbalance, can lead to significant neonatal complications. OBJECTIVE: This review examines the potential role of excessive maternal zinc supplementation as an underrecognized environmental modifier in Menkes disease (MD), an X-linked disorder caused by mutations in the ATP7A cop

Host SNARE Proteins Mediate Lysosome and PVM Fusion to Support Plasmodium Liver Infection.

Malaria, caused by Plasmodium parasites, remains a global health crisis, necessitating novel therapeutic strategies targeting host-parasite interactions. During liver-stage infection, parasites exploit host vesicular trafficking machinery, particularly SNARE (soluble N-ethylmaleimide-sensitive factor attachment protein receptor) proteins that mediate membrane fusion. Using a CRISPR/Cas9 knockout system in HeLa cells combined with advanced microscopy of Plasmodium berghei-infected HeLa cells, we identified specific endolysosomal SNAREs including Vesicle-Associated Membrane Protein 7 (VAMP7), Ve

β-globin mediates antimicrobial defense and inflammatory regulation in common carp (Cyprinus carpio).

β-globin is a core subunit of hemoglobin, playing an indispensable role in oxygen transport, but its immunological functions have been less extensively characterized. In this study, we identified and characterized the β-globin gene from Cyprinus carpio (designated Ccβ-globin), with a specific focus on its transcriptional response to pathogenic stimuli and antimicrobial activity against pathogens. The open reading frame (ORF) of Ccβ-globin consists of a 447 bp nucleotide sequence, encoding a 148 amino acids polypeptide. Quantitative analysis revealed that Ccβ-globin is constitutively expressed

Structural and Biophysical Characterization of the Yersinia Type Three Secretion System ATPase YscN.

Yersinia pestis was responsible for the Black Plague, one of the worst epidemiological disasters in recorded history. Today, Y. pestis , Y. enterocolitica , and Y. pseudotuberculosis remain clinically relevant human pathogens. Each of these pathogenic Yersinia species relies on a Type Three Secretion System (T3SS) for virulence, with the ATPase YscN playing a critical role in T3SS function. T3SS ATPases are responsible for powering apparatus formation and effector protein secretion through ATP hydrolysis. This study provides an extensive enzymatic characterization of recombinant YscN under sev

miR-424(322)~503 impairs colon cancer progression driven by PTEN deficiency.

Colorectal cancer (CRC) is a leading cause of cancer-related morbidity and mortality worldwide, with molecular subtypes and signaling pathways playing critical roles in its progression. The miR-424(322)~503 cluster, comprising miR-424 and miR-503, has been implicated in various malignancies, exhibiting dual roles as tumor suppressors or oncogenes depending on the context. However, its function in CRC remains poorly understood. This study investigates the role of the miR-424(322)~503 cluster in CRC driven by PTEN deficiency using genetically modified mouse models. Our findings reveal that the l

Self-Determination Theory in the Social Acceptance of Sustainable Energies and Technologies: A Comprehensive Review.

The transition toward sustainable energy systems is not only a technological and economic challenge but also a social one, with public acceptance playing a decisive role in the success of sustainability transitions. While traditional technology acceptance models primarily emphasize cognitive and utilitarian determinants, increasing attention has been directed toward underlying motivational processes. Self-Determination Theory (SDT) provides a valuable framework for understanding how autonomous and controlled motivation shape attitudes and behaviours toward sustainable energy technologies. This
