3D Printed Mesh Geometry Modulates Immune Response and Interface Biology in Mouse and Sheep Model: Implications for Pelvic Floor Surgery.

Pelvic organ prolapse (POP) is a highly prevalent yet neglected health burden for women. Strengthening the pelvic floor with bioactive tissue-engineered meshes is an emerging concept. This study investigates tissue regenerative design parameters, including degradability, porosity, and angulation, to develop alternative degradable melt electrowritten (MEW) constructs for surgical applications of POP. MEW constructs were fabricated in hierarchical geometries by two-way stacking of the fibers with three different inter layer angles of 90°, 45°, or 22.5°. Implants printed at 22.5° have higher tens

Induction of MASH in three-dimensional bioprinted human liver tissue.

Metabolic dysfunction-associated steatohepatitis (MASH), formerly known as nonalcoholic steatohepatitis (MASH), is a major risk factor for cirrhosis and hepatocellular carcinoma (HCC) and a leading cause of liver transplantation. MASH is caused by an accumulation of toxic fat molecules in the hepatocyte which leads to inflammation and fibrosis. Inadequate human "MASH in a dish" models have limited our advances in understanding MASH pathogenesis and in drug discovery. This study uses complex multicellular 3D bioprinting, combining hepatocytes with nonparenchymal cells in physiologically relevan

Advances in corneal regenerative medicine with iPS cells.

The cornea is a pioneering area of regenerative medicine, and Japanese researchers have led the world in this field. In Japan, 3 different epithelial sheet regenerative medicine products have been approved for corneal epithelial stem cell deficiency, and the first-in-human studies of cultured corneal endothelial cell suspension transplants, induced pluripotent stem cell (iPS cell)-derived corneal epithelial sheet transplants, and iPS cell-derived corneal endothelial substitute cell transplants were all conducted and reported globally for the first time by Japanese researchers. In the field of

GUIDED REGENERATION OF JAW BONE DEFECTS WITH COMBINATION OF OSTEOPLASTIC MATERIALS AND STEM CELLS.

Reconstruction of mandibular defects is a challenging issue in maxillofacial surgery. Success of the treatment is defined by the process of reparative regeneration of bone tissue, which is often accompanied with wound infection, failure of microcirculation and tissue hypoxia. Aim of our project was to demonstrate the advantage of osteoplastic material and bone marrow derived mesenchymal stem cells combination in regeneration of lower jaw bone defect. For this reason, mandibular bone defect was created in laboratory rats. Experimental treatment of animals was performed in three different ways:

Fiber density of collagen grafts impacts rabbit urethral regeneration.

There is a need for efficient and "off-the-shelf" grafts in urethral reconstructive surgery. Currently available surgical techniques require harvesting of grafts from autologous sites, with increased risk of surgical complications and added patient discomfort. Therefore, a cost-effective and cell-free graft with adequate regenerative potential has a great chance to be translated into clinical practice. Tubular cell-free collagen grafts were prepared by varying the collagen density and fiber distribution, thereby creating a polarized low fiber density collagen graft (LD-graft). A uniform, high

Potential Role of Induced Pluripotent Stem Cells as Regenerative Medicine in Retinal Cell Damage.

Induced pluripotent stem cells (also called iPSCs) are somatic cells reprogrammed by overexpressing four nuclear transcriptional factors containing Sox2, Klf4, c-myc and Oct4 is the one of research hotspots. Its pluripotency, self-renewal capacity and wide accessibility to donor tissues have made possible the means for modified regenerative medicine. They are considered a possible basis of healthy tissue to cure diseases, like ophthalmic diseases, degenerative diseases, age-related macular degeneration (AMD), are primarily because of the weakening capability of photoreceptor cells, retinal gan

Beagle sciatic nerve regeneration across a 30 mm defect bridged by chitosan/PGA artificial nerve grafts.

Longitudinally oriented microstructures are essential for a nerve scaffold to promote the significant regeneration of injured peripheral axons across nerve gaps. In the current study, we present a novel nerve-guiding collagen-chitosan (CCH) scaffold that facilitated the repair of 30 mm-long sciatic nerve defects in beagles. The CCH scaffolds were observed with a scanning electron microscope. Eighteen beagles were equally divided into CCH group, autograft group and non-graft group. The posture and gait of each dog was recorded at 12 and 24 weeks after surgery. Electrophysiological tests, Fluoro

Bioengineering functional human sphincteric and non-sphincteric gastrointestinal smooth muscle constructs.

Digestion and motility of luminal content through the gastrointestinal (GI) tract are achieved by cooperation between distinct cell types. Much of the 3 dimensional (3D) in vitro modeling used to study the GI physiology and disease focus solely on epithelial cells and not smooth muscle cells (SMCs). SMCs of the gut function either to propel and mix luminal contents (phasic; non-sphincteric) or to act as barriers to prevent the movement of luminal materials (tonic; sphincteric). Motility disorders including pyloric stenosis and chronic intestinal pseudoobstruction (CIPO) affect sphincteric and

A new heterologous fibrin sealant as scaffold to recombinant human bone morphogenetic protein-2 (rhBMP-2) and natural latex proteins for the repair of tibial bone defects.

Tissue engineering has special interest in bone tissue aiming at future medical applications Studies have focused on recombinant human bone morphogenetic protein-2 (rhBMP-2) and natural latex proteins due to the osteogenic properties of rhBMP-2 and the angiogenic characteristic of fraction 1 protein (P-1) extracted from the rubber tree Hevea brasiliensis. Furthermore, heterologous fibrin sealant (FS) has been shown as a promising alternative in regenerative therapies. The aim of this study was to evaluate these substances for the repair of bone defects in rats. A bone defect measuring 3mm in d…

Electrospun magnetic poly(L-lactide) (PLLA) nanofibers by incorporating PLLA-stabilized Fe3O4 nanoparticles.

Magnetic poly(L-lactide) (PLLA)/Fe3O4 composite nanofibers were prepared with the purpose to develop a substrate for bone regeneration. To increase the dispersibility of Fe3O4 nanoparticles (NPs) in the PLLA matrix, a modified chemical co-precipitation method was applied to synthesize Fe3O4 NPs in the presence of PLLA. Trifluoroethanol (TFE) was used as the co-solvent for all the reagents, including Fe(II) and Fe(III) salts, sodium hydroxide, and PLLA. The co-precipitated Fe3O4 NPs were surface-coated with PLLA and demonstrated good dispersibility in a PLLA/TFE solution. The composite nanofibe

Decellularized human tendon-bone grafts for composite flexor tendon reconstruction: a cadaveric model of initial mechanical properties.

PURPOSE: After complex hand trauma, restoration of tendon strength is challenging. Tendon insertion tears typically heal as fibrous scars after surgical reconstruction and create a weak point at the tendon-bone interface. In addition, major tendon loss may overwhelm the amount of available autograft for reconstruction. An off-the-shelf product may help address these challenges. We hypothesized that decellularized human flexor digitorum profundus and distal phalanx tendon-bone composite grafts were a feasible option for flexor tendon reconstruction after complex hand trauma. By replacing the en

[Translational Medicine and its application in stem cell research for periodontal regeneration].

Translational Medicine is an evolutional concept that encompasses the rapid translation of basic research for use in clinical disease diagnosis, prevention, treatment and finally in public health promotion. It follows the idea "from bench to bedside and back", and hence relies on cooperation between laboratory research and clinical care. Translation process is a complex process that requires both research and non-research activities. During the past ten years, there has been intense interest in the development of new clinical procedures, therapeutic molecules, and prototypes based on translati

Regression modeling to inform cell incorporation into therapies for craniosynostosis.

Designing an appropriate tissue engineering solution for craniosynostosis (CS) necessitates determination of whether CS-derived cells differ from normal (wild-type, WT) cells and what assays are appropriate to test for differences. Traditional methodologies to statistically compare cellular behavior may not accurately reflect biologically relevant differences because they poorly address variation. Here, logistic regression was used to determine which assays could identify a biological difference between WT and CS progenitor cells. Quantitative alkaline phosphatase and MTS proliferation assays …

Red/ET recombination with chimeric oligonucleotides allows rapid generation of BAC transgenes harboring full-length or truncated huntingtin cDNA.

Huntington's disease (HD) is a fatal neurodegenerative disorder that is caused by a CAG repeat expansion encoding a polyglutamine tract in the huntingtin (htt) gene. None of the existing HD mouse models recapitulate the exact disease symptoms and course as it is seen in humans and the generation of further HD disease models is challenging because of the size and complexity of the htt gene locus. Starting from a single substrate plasmid harboring human htt cDNA comprising 98 glutamine (Q) residues, we applied Red/ET recombination to generate four BDNF-BAC transgenes harboring full-length or tru

Collagen-immobilized patch for repairing small tympanic membrane perforations: in vitro and in vivo assays.

Tympanic membrane (TM) perforation is still one of the most common otology complications. New designs of biomaterials, and lately tissue-engineered composites and grafts, have thoroughly revolutionized the management of TM perforation. In this study, we examined a biologically modified collagen-immobilized polydimethyl siloxane patch to repair TM perforation. In vitro potential of the aforementioned patch as a scaffold to support fibroblast cell growth and adhesion was assessed. An in vivo assay of the patch for initiating repair of TM perforations also was investigated. In vitro assay showed

[Aseptic necrosis of the capitulum of the humerus (Panner's disease) treated by solid chondrograft implantation].

The authors present the case of aseptic necrosis of the humeral capitulum in a top female gymnast. She was referred to our department with the diagnosis of aseptic necrosis already made and after repeated arthroscopic treatment of the elbow including removal of a loose body in another institution. Exercise-related pain and swelling of the elbow became permanent and did not allow her to pursue her sports activities. After thorough examination, the treatment by implantation of autologous cultured chondrocytes, as a solid chondrograft, was chosen. At the first stage of arthroscopic surgery, a loo

Repair of a segmental long bone defect in human by implantation of a novel multiple disc graft.

Large segmental defects of the weight bearing long bones are very difficult to reconstruct. Current treatment options are afflicted with several limitations and disadvantages. We describe a novel approach to regenerate a segmental long bone defect in a patient using a multiple disc graft. Decellularized bovine trabecular bone discs were seeded with autologous bone marrow cells and cultured in a perfusion chamber for three weeks. Multiple cell-seeded discs were implanted to close a 72 mm defect of the distal tibia in a 58-year-old woman, and fixed by an intramedullary nail. Bone formation was a
