Microenvironmental Cues That Regulate Cancer Cell Dormancy in Estrogen-Receptor Positive and Triple-Negative Breast Cancer Cell Lines.

The processes regulating cellular dormancy in tumor cells remain inadequately characterized. Further, there are very few models available that can recapitulate a dormancy-like phenotype in vitro. In this study, we investigated the role of microenvironmental cues in inducing a dormancy-like phenotype in estrogen receptor-positive (MCF-7) and triple-negative (MDA-MB-231) breast cancer cell lines. We characterized an in vitro model in distinct conditions defined hierarchically by the following: (a) normoxia or true hypoxia, (b) dishes coated with fibronectin or laminin or none, and (c) presence o

Growth differentiation factor-15 as a clinical biomarker of frailty, sarcopenia and functional decline: A systematic literature review.

BACKGROUND: With an aging population globally, prevention of frailty and sarcopenia will become a public health priority. Growth Differentiation Factor-15 (GDF-15), a stress-responsive cytokine of the TGF-β superfamily, has emerged as a promising biomarker linking mitochondrial dysfunction, cellular senescence, and systemic inflammation to biological and phenotypic aging. OBJECTIVES: This systematic literature review systematically synthesizes the clinical evidence on GDF-15 as a biomarker of frailty, sarcopenia, and physical function, highlighting patterns, gaps, and the biological plausibil…

The Role of Wnt Signaling in Age-Related Alveolar Bone Loss and Regeneration.

The periodontium is a uniquely dynamic tissue system requiring precise signaling for lifelong adaptation. The canonical Wnt/β-catenin pathwa … anisms of age-related decline include the upregulation of Wnt antagonists (e.g., sclerostin, DKK1), cellular senescence, altered FoxO-Wnt crosstalk under oxidative stress, and impaired mechanosensing. These changes converge to disrupt regenerative capacity, tipping the balance toward net alveolar bone loss. Therapeutically, sclerostin inhibition demonstrates robust p …

T-cell-mediated immunity to influenza A (H2N3): implications for caccine efficacy and cross-subtype protection.

Influenza A viruses pose a significant pandemic threat, with H2N3 in particular representing a subtype of particular concern due to limited population immunity and potential for zoonotic transmission. Traditional antibody-centric vaccine strategies face fundamental limitations stemming from antigenic drift and shift in hemagglutinin and neuraminidase surface proteins, necessitating annual reformulation and demonstrating reduced efficacy against mismatched strains. T-cell-mediated immunity offers a complementary approach by targeting highly conserved internal viral proteins such as nucleoprotei

Biological mechanisms governing the periodontal regenerative microenvironment: cellular crosstalk, extracellular matrix remodelling, and immunomodulation.

Periodontal regeneration is no longer viewed as the simple replacement of lost alveolar bone, but as the reprogramming of a diseased regenerative niche. Predictable repair requires coordinated reconstruction of cementum, periodontal ligament (PDL), and alveolar bone within a chronically infected, inflamed, and mechanically loaded microenvironment. Unlike regeneration in bone, skin or muscle, periodontal regeneration requires reconstruction of an integrated cementum-PDL-bone unit. Success therefore depends not on repair of a single tissue compartment, but on coordinated cementogenesis, PDL fibr

The longevity gene APOE2 enhances pericyte function and reduces lipid droplets.

Pericytes are critical for maintaining blood-brain barrier (BBB) integrity and have emerged as key contributors to Alzheimer's disease (AD) pathogenesis. Although the apolipoprotein E2 (APOE2) allele is associated with reduced AD risk and increased longevity, its impact on pericyte function is unclear. We measured pericyte density in the brains of humanized APOE2, APOE3, and APOE4 knock-in mice and found that APOE2 mice revealed increased pericyte markers and enhanced BBB integrity. To uncover the underlying mechanisms, we used CRISPR/Cas9 editing to generate isogenic human iPSC-derived pericy

Chromosomal instability in cancer: sources, consequences and new therapeutic opportunities.

Chromosomal instability (CIN) refers to a high rate of chromosome segregation errors during consecutive cell divisions and is a hallmark of cancers with aggressive behavior and poor prognosis. Cancer cells exhibiting CIN may acquire both numerical (N-CIN) and structural (S-CIN) chromosome alterations, namely by chromosome mis-segregation and DNA replication/repair defects, respectively. Importantly, these processes are tightly interconnected: mis-segregating chromosomes are prone to DNA damage, while DNA replication or repair defects can, in turn, promote chromosome mis-segregation. Consequent

Targeting RNA polymerase I to boost natural killer cell anticancer activity in multiple myeloma.

Multiple myeloma (MM) remains an incurable disease despite therapeutic advancements extending survival. Relapses driven by drug resistance and minimal residual disease underscore the need for novel treatment strategies. Natural Killer (NK) cells play a key role in MM immunity, yet their function is suppressed by inhibitory cytokines and metabolites from the tumor microenvironment. Developing anticancer drugs with immunomodulatory properties, such as enhancing tumor sensitivity to NK cell recognition, remains a critical challenge. MM cells exhibit high protein synthesis rates, making them vulne

Microvesicle inhibition enhances the therapeutic effects of ATRA in acute promyelocytic leukemia cells via changes in miRNAs: the promising antileukemic potential of imipramine.

Extracellular vesicles (EVs) represent an essential role in cancer progression through intercellular communication. Therefore, the use of EV … onstrated that these genes are involved in cancer-related pathways, including MAPK, FOXO, AMPK, and cellular senescence. Given the significant efficacy of imipramine in potentiating the anti-tumor effects of chemotherapeutic drugs, it can be considered as a potential treatment agent for APL.

Design and evaluation of highly selective histone deacetylase 6 inhibitors derived from the natural product tryptoline.

The development of isoform-selective histone deacetylase (HDAC) inhibitors offers a promising approach to minimize the adverse effects of nonselective HDAC inhibitors. HDAC6, due to its unique structural and functional properties, regulates critical cellular processes like gene expression, proliferation, senescence, and apoptosis. Inspired by a tryptoline-derived natural product, callophycin A, a series of compounds were synthesized and evaluated for HDAC6 selectivity. In the HDAC enzyme assay, compound 6a stood out as the lead, demonstrating 21-fold higher potency against HDAC6 compared with

Association of phenotypic frailty and hand grip strength with telomere length in SLE.

OBJECTIVE: Frailty and objective hand grip strength (one of the components of the frailty phenotype) are both risk factors for worse health outcomes in SLE. Whether telomere length, an established cellular senescence marker, is a biologic correlate of the frailty phenotype and hand grip strength in patients with SLE is not clear. First, we aimed to evaluate differences in telomere length between frail and non-frail women with SLE and then assessed whether frailty or hand grip strength is differentially associated with telomere length after adjusting for relevant confounders. METHODS: Women ≥1…

Evaluation of respiratory exchange ratio (RER) for predicting postoperative outcomes in elderly patients undergoing oncological resection for gastrointestinal malignancies - A prospective cohort study.

BACKGROUND AND AIMS: Predicting complications after major oncosurgery is particularly daunting in the elderly subcategory of patients owing to factors like preexisting age-related immune cellular senescence and a significant imbalance of oxygen delivery (DO2) and consumption (VO2) characteristic of major oncological surgeries. The respiratory exchange ratio (RER) indicates DO2-VO2 balance and onset of anaerobic metabolism. We evaluated the ability of RER in predicting the occurrence of postoperative complications following geriatric oncosurgery. MATERIALS AND METHODS: In the study, we enrolle…

Cell clusters in intervertebral disc degeneration: an attempted repair mechanism aborted via apoptosis.

Cell clusters are a histological hallmark feature of intervertebral disc degeneration. Clusters arise from cell proliferation, are associated with replicative senescence, and remain metabolically, but their precise role in various stages of disc degeneration remain obscure. The aim of this study was therefore to investigate small, medium, and large size cell-clusters. For this purpose, human disc samples were collected from 55 subjects, aged 37-72 years, 21 patients had disc herniation, 10 had degenerated non-herniated discs, and 9 had degenerative scoliosis with spinal curvature <45°. 15 non-
