CALR-mutant myeloproliferative neoplasms: insights from next-generation sequencing.

Essential thrombocythemia and primary myelofibrosis belong to the group of BCR::ABL1-negative myeloproliferative neoplasms. The presence of mutations in the JAK2, CALR, and MPL genes is essential for the diagnosis of myeloproliferative neoplasms. These mutations are called "driver" mutations. However, not only leading mutations have been identified in patients with MPN, but also more than half of individuals with essential thrombocythemia and more than 80% of patients with myelofibrosis have additional mutations. One technique that makes it possible to find prognostic, predictive, and diagnost

Progression from acute to chronic pancreatitis in children: a systematic review and meta-analysis.

BACKGROUND: Most children recover after an initial acute pancreatitis (AP) episode; however, some progress to recurrent AP (RAP) or chronic pancreatitis (CP). PURPOSE: We aimed to quantify progression rates and identify the risk factors associated with these transitions. METHODS: PubMed/MEDLINE, Embase, and Cochrane databases were searched on December 21, 2024, for pediatric studies reporting progression to RAP or CP (PROSPERO number: CRD420251086520). All observational studies were included, while case reports and case series were excluded. To evaluate the differences in RAP rates, we condu

[Brain and Meningeal Metastases of Lung Cancer Manifested as Brain Calcifications:
A Case Report and Literature Review].

Lung cancer is still one of the most common malignant tumors in the world. With the increase of its incidence and the development of medical technology, the overall survival of lung cancer patients has significantly extended compared to before. The incidence of brain and meningeal metastases from lung cancer has also been rising year by year, but patients with brain and meningeal metastases from lung cancer have a poor prognosis and a very high mortality rate, and the diagnosis is mainly based on computed tomography (CT), magnetic resonance imaging (MRI) and other imaging examinations. However

Neuroimaging and Genetic Markers of Cerebral Small Vessel Disease and Cognitive Outcomes: A Systematic Review and Meta-Analysis (NEUROGEN-SVD Study).

Background/Objectives: Cerebral small vessel disease (CSVD) is a leading cause of cognitive decline and dementia. The comparative prognostic value of MRI-based neuroimaging markers and genetic risk factors such as the APOE ε4 allele for cognitive outcomes remains uncertain. The objectives of this study were to estimate the pooled prevalence of cognitive impairment in CSVD, evaluate the associations of key neuroimaging markers (white matter hyperintensities [WMHs], cerebral microbleeds [CMBs], lacunes) and APOE ε4 with cognitive outcomes, and assess their diagnostic performance. Methods: This s

Federated learning framework for predicting multi-drug resistant tuberculosis across regional databases.

BACKGROUND: Multi-Drug Resistant Tuberculosis (MDR-TB) is a major public health problem around the world, especially in low- and middle-income countries where quick and accurate testing is needed for effective treatment and control. Traditional ways of diagnosing, like culture-based drug resistance tests and genetic studies, have problems like taking a long time to run, being expensive, and not being easy to get to in places with few resources. While machine learning models that combine genetic and clinical data show promise in predicting MDR-TB, their progress is slowed by worries about data

New insights into the evolutionary origin of Francisella tularensis subsp. holarctica.

BACKGROUND: As a global expansion of Francisella tularensis subsp. holarctica in the northern hemisphere, its origin has been of great interest. Based on the phylogenetic analysis of the whole genome sequences (WGS) available in public databases, estimating temporal signal in accordance with random permutations of sampling dates, and searching the powerful SNPs within the B.16 clade, we intend to explore the historical origin of this subspecies and routes of dispersal across its regional distribution. METHODS: We constructed a phylogenetic tree for the four main clades based on 395 WGSs, incl

EHMT2 as a Candidate Gene for an Autosomal Recessive Neurodevelopmental Syndrome.

Neurodevelopmental disorders (NDD) comprise clinical conditions with high genetic heterogeneity and a notable enrichment of genes involved in regulating chromatin structure and function. The EHMT1/2 epigenetic complex plays a crucial role in repression of gene transcription in a highly tissue- and temporal-specific manner. Mutations resulting in heterozygous loss-of-function (LoF) of EHMT1 are implicated in Kleefstra syndrome 1 (KS1). EHMT2 is a gene acting in epigenetic regulation; however, the involvement of mutations in this gene in the etiology of NDDs has not been established thus far. A

Human visceral leishmaniasis and polymorphisms in interleukin-coding genes: a systematic review.

Visceral leishmaniasis (VL) is a neglected disease that is typical of tropical and subtropical parts of the world and is caused by the trypanosomatid Leishmania donovani complex. This disease is a multifactorial condition that involves parasitic, environmental, and immunogenetic characteristics. Genetic changes in genes encoding cytokines may be associated with changes in their expression and, consequently, with the development of clinical resistance or susceptibility to the disease. This systematic review and meta-analysis aimed to assess whether single nucleotide polymorphisms (SNPs) in inte

Psychosis in Laurence-Moon Syndrome: A Case Report.

BACKGROUND: Laurence-Moon syndrome (LMS) is a rare autosomal recessive genetic disorder characterized by a combination of neurological, visual, and endocrine abnormalities. The coexistence of psychiatric disorders in LMS patients can complicate clinical management due to the intricate interplay between neurodevelopmental and psychiatric symptoms. A review of the literature failed to identify a similar case study published in major databases, including PubMed, highlighting the significance of this case report. Understanding these complexities is crucial for improving treatment strategies and pa

Loop-mediated isothermal amplification assays for the detection of antimicrobial resistance elements in Vibrio cholera.

BACKGROUND: The bacterium Vibrio cholerae causes diarrheal illness and can acquire genetic material leading to multiple drug resistance (MDR). Rapid detection of resistance-conferring mobile genetic elements helps avoid the prescription of ineffective antibiotics for specific strains. Colorimetric loop-mediated isothermal amplification (LAMP) assays provide a rapid and cost-effective means for detection at point-of-care since they do not require specialized equipment, require limited expertise to perform, and can take less than 30 min to perform in resource limited regions. LAMP output is a co

Reproductive decision-making and the utilization of preimplantation genetic testing among individuals with inherited aortic or vascular disease.

Preimplantation genetic testing for monogenic disorders (PGT-M) is a reproductive technology used in conjunction with in-vitro fertilization (IVF) to reduce the risk of passing on a known genetic condition from parent to child. There is limited research describing the experience and emotional impact of PGT-M among individuals with inherited aortic or vascular disease (IAVD). Our qualitative study aims to explore the factors that influence reproductive decision-making and the uptake of PGT-M within this population. Individuals diagnosed with IAVD who have considered PGT-M, and/or their reproduc

Horizontal transfer of probable chicken-pathogenicity chromosomal islands between Staphylococcus aureus and Staphylococcus agnetis.

Staphylococcus agnetis is an emerging pathogen in chickens but has been most commonly isolated from sub-clinical mastitis in bovines. Previous whole-genome analyses for known virulence genes failed to identify determinants for the switch from mild ductal infections in cattle to severe infections in poultry. We now report identification of a family of 15 kbp, 17-19 gene mobile genetic elements (MGEs) specific to chicken osteomyelitis and dermatitis isolates of S. agnetis. These MGEs can be present in multiple copies per genome. The MGE has been vectored on a Staphylococcus phage that separately
