BACKGROUND: Currently, psoriasis is thought to be an inflammatory response to an antigenic stimulation, in which angiogenesis plays a fundamental role. Very late antigen-1 (VLA-1) is a beta(1) integrin collagen receptor that is up-regulated in many angiogenic processes. Data on its role in psoriasis are sparse.
OBJECTIVE: In a prospective study, we evaluated the staining of VLA-1 in lesional skin from patients with psoriasis and atopic dermatitis.
MATERIAL AND METHODS: Frozen sections from skin biopsies of patients with chronic plaque-type psoriasis (n = 18) and chronic atopic dermatitis (n

Cellular diversity, which is a hallmark of malignancy, can be generated by both genetic and nongenetic mechanisms. We describe here variability in the adhesive and migratory behavior of malignant plasma cell populations, including multiple myeloma-derived lines and primary patient samples. Examination of the plasma cell lines ARH-77, CAG, and AKR revealed two distinct subpopulations of cells, one displaying highly adhesive properties (type A) and the other consisting of poorly adhesive, floating cells (type F). In the ARH-77 cell line, type A cells attach better to fibronectin and to human bon

Prion pathogenesis following oral exposure is thought to involve gut-associated lymphatic tissue, which includes Peyer's patches (PPs) and M cells. Recruitment of activated B lymphocytes to PPs requires alpha(4)beta(7) integrin; PPs of beta 7(-/-) mice are normal in number but are atrophic and almost entirely devoid of B cells. Here we report that minimal infectious dose and disease incubation after oral exposure to logarithmic dilutions of prion inoculum were similar in beta 7(-/-) and wild-type mice, and PPs of both beta 7(-/-) and wild-type mice contained 3-4 log LD(50)/g prion infectivity

OBJECTIVE: In cohorts of infants with proven neonatal alloimmune thrombocytopenia (NAIT) an unusual high rate of preterm infants has been reported, raising the question of whether NAIT contributes to the high rate of intracranial hemorrhage in preterm infants.
METHODS: We genotyped the HPA-1-allele in a large cohort of term (n = 205) and very low birth weight infants (VLBW-infants, n = 299) with polymerase-chain-reaction and restriction enzyme digestion.
RESULTS: HPA-1a/b is the only fetal HPA-1-genotype in which alloimmunization and NAIT could occur. Genotype distribution did not differ bet

The measurement of leukocyte rheology in vascular disease is a recent development with a wide range of new opportunities. The International Society of Clinical Hemorheology has asked an expert panel to propose guidelines for the investigation of leukocyte rheology in clinical situations. This article first discusses the mechanical, adhesive and related functional properties of leukocytes (especially neutrophils) which influence their circulation, and establishes the rationale for clinically-related measurements of parameters which describe them. It is concluded that quantitation of leukocyte a

We report two patients where the finding of isolated fetal hydrocephalus led to the detection of severe fetal thrombocytopenia, using fetal blood sampling. Serological investigation led to the diagnosis of fetomaternal alloimmune thrombocytopenia (FMAIT) due to anti-HPA-1a. Both women had had previous unsuccessful pregnancies probably due to FMAIT; one had had four miscarriages at 17-18 weeks' gestation. The other had had one previous pregnancy complicated by severe fetal anaemia, and eventually hydrocephalus developed and the fetus died without the diagnosis of FMAIT being considered. Subsequ

Genetic defects of the blood platelet membrane glycoproteins, GPIIb-IIIa (alpha IIb/beta 3; CD41/CD61) and GPIb-V-IX (CD42) are the origin of several rare bleeding disorders, the best known of which are Glanzmann's thrombasthenia, Bernard-Soulier syndrome, and platelet-type von Willebrand's disease. In Glanzmann's thrombasthenia, GPIIb-IIIa are missing or defective and platelet aggregation is lacking or reduced. Either gene can be affected and mutations leading to lack of expression or to expression of poorly functional forms have been described. In Bernard-Soulier syndrome, GPIb-V-IX are miss

As rates of type 2 diabetes mellitus (T2DM) have risen, comorbidities of T2DM and cardiovascular disease (CVD) have similarly increased. The adoption of sodium-glucose cotransporter (SGLT) 2 inhibitors as a treatment for T2DM has provided an effective means of decreasing renal glucose reabsorption. In recent clinical trials, sotagliflozin, the first FDA-approved inhibitor of both isoforms of SGLT (SGLT1 and SGLT2), demonstrated a reduction in myocardial infarction and stroke in patients with T2DM at high cardiovascular risk. The cardiovascular protective effect of sotagliflozin compared to the

Pregnancy imposes profound hemodynamic, metabolic, and biomechanical stresses on the heart, requiring coordinated structural and molecular adaptations to preserve cardiac function. To compensate, the cardiovascular system undergoes significant reversible adaptations, such as increased blood volume and elevated cardiac output. Such alterations induce physiological left ventricular hypertrophy that is regulated, in part, by mechanotransduction, a process by which cells convert mechanical stimuli such as stretch and pressure into biochemical pathways. Integrin-mediated complexes link cardiomyocyt

Our previous genome-wide screening has linked THSD7A (thrombospondin type 1 domain containing 7A) gene to human coronary artery disease (CAD). How THSD7A contributes to atherosclerosis, however, remains unclear. In this study, we show THSD7A is increased in human carotid arteries and mouse atherosclerotic plaques. In apolipoprotein E knockout (ApoE-/-) mice, the overexpression of THSD7A increases monocyte-endothelial adhesion and macrophage infiltration, exacerbating atherosclerotic lesions, whereas Thsd7A knockout (Thsd7A-/-) attenuates these phenotypic changes. Pancreatic ductal adenocarcinoma (PDAC) is a highly chemoresistant and immunoresistant tumor with an overall five-year survival rate of less than 10 %. Current treatments for PDAC are rather limited, highlighting the importance of finding novel strategies. In this study, we investigated a strategy for the tumor-specific targeting of PAPTP, a small molecule that reduces PDAC growth by inhibiting the mitochondrial potassium channel mtKv1.3, thereby inducing mitochondrial dysfunction and killing cancer cells. PAPTP was reversibly conjugated to three tumor-penetrating peptides: iRGD, VH434, and a

INTRODUCTION: Chronic cerebral hypoperfusion (CCH) is a major contributor to cognitive impairment; however, its underlying mechanisms remain poorly understood.
METHODS: We investigated CCH-induced glymphatic dysfunction and neurodegeneration in amyloid precursor protein (APP)/presenilin 1 (PS1) and wild-type mice. Glymphatic transport was assessed using contrast-enhanced magnetic resonance imaging (MRI) and real-time femoral vein imaging. Aquaporin-4 (AQP4) polarization and amyloid beta (Aβ)/phosphorylated tau 217 (p-tau217) accumulation were examined by immunofluorescence staining. Single-ce

Phosphatidylinositol-5-phosphate 4-kinases (PI5P4Ks) have emerged as candidate drug targets in cancer, neurological, inflammatory, and infectious diseases. Although their canonical function is to phosphorylate PI(5)P to generate PI(4,5)P2, growing evidence points to additional catalytic-independent roles, but how these functions are organized within protein interaction networks remains unclear. Here, we use proximity-dependent biotin identification (BioID) in HeLa cells to map isoform-resolved interactomes of human PI5P4Kα, PI5P4Kβ, and PI5P4Kγ. This approach captures PI5P4K-proximal proteins

The malignant progression of renal cell carcinoma (RCC) is closely associated with the establishment of an immunosuppressive tumor microenvironment (TME). This study aims to elucidate the oncogenic functions of DNA polymerase ε subunit POLE2 in RCC and its mechanism in modulating the tumor immune microenvironment. We found that POLE2 was significantly upregulated in clear cell RCC (ccRCC) tissues and cell lines. Gain-of-function and loss-of-function experiments demonstrated that POLE2 markedly enhanced tumor cell proliferation, migration, and tumorigenicity both in vitro and in vivo. Mechanist

Precise extracellular matrix (ECM) coordination is essential for cardiac septation, yet the molecular etiology of human isolated perimembranous ventricular septal defects (VSD) remains elusive. Here, we constructed a high-resolution single-cell transcriptomic atlas of human VSD, benchmarking the pathological state against a 7-22 week developmental trajectory. Integrating these single-cell profiles with a reference-based spatial transcriptomic map, we deconvolved the distinct molecular signatures of membranous and muscular septal regions. We found that aberrant cardiomyocyte hypertrophy operate

Cisplatin-based chemotherapy combined with immunotherapy is the standard first-line treatment for advanced nasopharyngeal carcinoma (NPC), but its efficacy is limited by off-target toxicity, platinum resistance, and the immunosuppressive tumor microenvironment (TME). Herein, we report NP2, a reactive oxygen species (ROS)-sensitive nanoparticle co-loaded with a platinum (IV) prodrug (CisPt-IV) and a CDK1 inhibitor (RO-3306), for targeted delivery to NPC through surface-functionalization with RGD peptide. Following intravenous administration, NP2 homes to NPC tissues overexpressing integrin rece

Zika virus (ZIKV) can traverse the placental barrier, leading to fetal microcephaly and congenital zika syndrome (CZS). The viral E protein mediates host-cell interactions and infection. Here, we demonstrated that cord blood natural killer cell-derived extracellular vesicles (CBNK-EVs) potently inhibit ZIKV infection in vitro without compromising cellular viability. Mechanistically, CBNK-EVs engage ZIKV through ITGB2, a surface-enriched integrin that interacts with the viral E protein, facilitating nanoparticle-virion contact or membrane fusion. This interaction triggers antiviral activity via

The Achilles tendon, the largest and strongest tendon in the human body, exhibits dynamic adaptive changes in its structure and function through mechanobiological regulation. This review synthesizes the dual regulatory effects of mechanical loading on Achilles tendon homeostasis and pathology: Moderate mechanical stimulation activates integrin-mediated signaling pathways (including PI3K/Akt and MAPK/ERK cascades), promoting tenocyte proliferation/differentiation, collagen biosynthesis, and orderly remodeling of extracellular matrix (ECM), thereby enhancing tendon stiffness, elastic modulus, an
