Simulation of pulsed dynamic nuclear polarization in the steady state.

In pulsed dynamic nuclear polarization (DNP), enhancement of bulk nuclear polarization requires the repeated application of a microwave pulse sequence. So far, analysis of a one-time transfer of electron spin polarization to a dipolar-coupled nuclear spin has guided the design of DNP pulse sequences. This has obvious shortcomings, such as the inability to predict the optimal repetition time. In an actual pulsed DNP experiment, a balance is reached between the polarization arriving from the unpaired electrons and nuclear relaxation. In this article, we explore three algorithms to compute this s

Optimizing Global Network Alignment With a Genetic Algorithm: Leveraging Pre-Trained Embeddings for Protein Sequences and Gene Ontology Terms.

Multiple objectives have emerged in tuning protein-protein interaction (PPI) networks, such as identifying cross-species network similarities and predicting protein complexes and functions. Despite the proliferation of tuning methodologies, challenges remain in balancing accuracy and efficiency. In this paper, we introduce GA2Vec, a novel approach for globally aligning multiple PPI networks using genetic algorithms in a many-to-many fashion. GA2Vec leverages vector embeddings of protein sequences from ProtBERT, ESM-2, and ProtT5-XL-UniRef50 to reconstruct weighted PPI networks, incorporating f

Real-World Data and Machine Learning to Predict Cardiac Amyloidosis.

(1) Background: Cardiac amyloidosis or "stiff heart syndrome" is a rare condition that occurs when amyloid deposits occupy the heart muscle. Many patients suffer from it and fail to receive a timely diagnosis mainly because the disease is a rare form of restrictive cardiomyopathy that is difficult to diagnose, often associated with a poor prognosis. This research analyses the characteristics of this pathology and proposes a statistical learning algorithm that helps to detect the disease. (2) Methods: The hospitalization clinical (medical and nursing ones) records used for this study are the ba

Differential diagnosis of ameloblastoma and odontogenic keratocyst by machine learning of panoramic radiographs.

PURPOSE: The differentiation of the ameloblastoma and odontogenic keratocyst directly affects the formulation of surgical plans, while the results of differential diagnosis by imaging alone are not satisfactory. This paper aimed to propose an algorithm based on convolutional neural networks (CNN) structure to significantly improve the classification accuracy of these two tumors. METHODS: A total of 420 digital panoramic radiographs provided by 401 patients were acquired from the Shanghai Ninth People's Hospital. Each of them was cropped to a patch as a region of interest by radiologists. Furt

A real-time data-based scan conversion method for single element ultrasound transducers.

The current work investigates the performance of a real-time scan conversion algorithm for generating a 2-D ultrasound image from a laterally scanned single-element ultrasound transducer, which has applications in point-of-care devices such as for skin imaging. The algorithm employs a fixed calibration curve to update a predefined image grid in real time. Simulations showed that the calibration curve (with a maximum of 1) is robust to changes in scatterer concentration (8.3×10-3 mean absolute error), signal to noise ratio (1.0×10-3 mean absolute error for -5 dB SNR), and can be accurately pred

Predicting neurofibromatosis type 1 risk among children with isolated café-au-lait macules.

BACKGROUND: Although isolated cafe-au-lait macules (CALMs) are a common skin finding, they are an early feature of neurofibromatosis type 1 (NF1). OBJECTIVE: We sought to develop an algorithm determining the risk of children with CALMs to have constitutional NF1. METHODS: We conducted a retrospective study of patients with isolated CALMs. Diagnosis of NF1 was based on detecting NF1 mutation in blood or fulfilling clinical criteria. RESULTS: In all, 170 of 419 (41%) and 21 of 86 (24%) children with isolated CALMs who underwent molecular testing and clinical follow-up, respectively, were give

A comparative reference study for the validation of HLA-matching algorithms in the search for allogeneic hematopoietic stem cell donors and cord blood units.

The accuracy of human leukocyte antigen (HLA)-matching algorithms is a prerequisite for the correct and efficient identification of optimal unrelated donors for patients requiring hematopoietic stem cell transplantation. The goal of this World Marrow Donor Association study was to validate established matching algorithms from different international donor registries by challenging them with simulated input data and subsequently comparing the output. This experiment addressed three specific aspects of HLA matching using different data sets for tasks of increasing complexity. The first two tasks

Development and validation of a decision-making algorithm to guide the use of plerixafor for autologous hematopoietic stem cell mobilization.

Plerixafor is an inhibitor of CXCR-4 (CXC chemokine receptor-4)/SDF (stromal cell-derived factor)-1 binding used in combination with granulocyte colony-stimulating factor (G-CSF) for mobilization of autologous peripheral blood hematopoietic stem cells (HSCs). We developed a data-generated, cost-saving decision-making algorithm that uses the CD34+ count in the peripheral blood on the fourth day of G-CSF administration (PB-CD34+), and the collection target (T-CD34+) to decide between continuing G-CSF only (G approach) or adding plerixafor to the mobilization regimen (G+P approach) aiming at the

Aspects of diagnosis of acute otitis media.

BACKGROUND: Acute otitis media is a common disease, particularly among children. The importance of a correct diagnosis is crucial, especially as unjustified prescription of antibiotics has become a major problem in clinical praxis. OBJECTIVES: Our aim was to evaluate the predictive value of different otological findings in diagnostics and treatment of acute otitis media among GPs and ear specialists and to investigate if the diagnosis could be improved by the use of an ear microscope instead of an otoscope. Furthermore, we aimed to test the value of following an algorithm, METHODS: Thirty-on

Predicting the Timing of the Metabolic Inflection Point in Type 1 Diabetes Progression Using Machine Learning and Survival Analysis Models.

UNLABELLED: An inflection point (IP) marking accelerated β-cell decline occurs ∼1-2 years before type 1 diabetes diagnosis. Precisely determining this timing could optimize clinical intervention. We developed machine learning models to predict proximity to the inferred metabolic IP using oral glucose tolerance test (OGTT) data from islet autoantibody-positive individuals in the TrialNet Pathway to Prevention study. OGTTs were retrospectively labeled by estimating time to the IP using thresholds from 1.2 to 1.6 years. Models were trained on engineered glucose and C-peptide dynamics, slopes, and

Hybrid model based on fuzzy logic and classification trees for the prediction of mortality of critical trauma patients: Pre-hospital variables from the RETRAUCI registry.

BACKGROUND: In critically injured trauma patients, tools that stratify injury severity and estimate mortality are essential. Fuzzy logic (FL) enables the creation of accurate, interpretable models but requires decision rules, which can be generated using machine learning (ML) techniques like classification trees (CT). Our objective was to develop a hybrid model combining fuzzy logic and classification trees to estimate ICU mortality risk using only prehospital variables. METHODS: We conducted a retrospective study using data from the Spanish Trauma ICU registry (RETRAUCI) from 2015 to 2022. P

Machine Learning-Based Prediction and Feature Attribution Analysis of Contrast-Associated Acute Kidney Injury in Patients with Acute Myocardial Infarction.

Background and Objectives: Contrast-associated acute kidney injury (CA-AKI) is a frequent and clinically significant complication in patients with acute myocardial infarction (AMI) undergoing coronary angiography. Early and accurate risk stratification remains challenging with conventional models that rely on linear assumptions and limited variable integration. This study aimed to evaluate and compare the predictive performance of multiple machine learning (ML) algorithms with traditional logistic regression and the Mehran risk score for CA-AKI prediction and to explore key determinants of ris

A prediction model of elderly hip fracture mortality including preoperative red cell distribution width constructed based on the random survival forest (RSF) and Cox risk ratio regression.

UNLABELLED: An independent correlation between pre-RDW and 1-year mortality after surgery in elderly hip fracture can be used to predict mortality in elderly hip fracture patients and has predictive significance in anemia patients. With further research, a treatment algorithm can be developed to potentially identify patients at high risk of preoperative mortality. INTRODUCTION: Red blood cell distribution width (RDW) is an independent predictor of various disease states in elderly individuals, but its association with the prognosis of elderly hip fracture patients is controversial. This study

Development of models for predicting the 7-month risk of venous thromboembolism and clinically relevant bleeding in ambulatory patients with cancer: analysis from the apixaban for the prevention of venous thromboembolism in high-risk ambulatory cancer patients trial.

BACKGROUND: Patients with cancer are at elevated risk of venous thromboembolism (VTE). While primary thromboprophylaxis reduces VTE incidence, it also increases bleeding risk, necessitating accurate risk stratification. Existing tools (eg, the Khorana score) have modest predictive value, and bleeding risk models have not been validated in ambulatory patients with cancer. OBJECTIVES: To develop and internally validate prediction models for VTE and clinically relevant bleeding in ambulatory patients with cancer initiating chemotherapy. METHODS: We used data from 514 participants in the apixaba
