Recurrent insulin-induced hypoglycemia causes site-specific patterns of habituation or amplification of CNS neuronal genomic activation.

Antecedent hypoglycemia is a primary factor in hypoglycemia-associated autonomic failure, a pathophysiological condition characterized by impaired glucose counterregulatory function. Conventional therapeutic strategies involving administration of intermediate dosage-release formulations of insulin in the management of insulin-dependent diabetes mellitus result in frequent iatrogenic hypoglycemia. This study investigated the neuroanatomical location, direction, and magnitude of CNS neuronal genomic activation by singular versus repeated induction of hypoglycemic bouts of greater than 6 h durati

Diminished sympathoinhibition during the onset of low-intensity leg cycling in young Black males.

Previous studies have reported that non-Hispanic Black (BL) males exhibit augmented blood pressure (BP) responses to dynamic exercise relative to White males; however, the underlying mechanism(s) remain unclear. To our knowledge, no studies have examined the muscle sympathetic nerve activity (MSNA) response to dynamic exercise in BL adults. The purpose of this study was to examine potential racial differences in the sympathoinhibition that occurs at the onset of dynamic exercise. We hypothesized that cardiopulmonary baroreflex-mediated inhibition of sympathetic vasomotor outflow during the ons

Sympathetic neural and cardiovascular responses during static handgrip exercise in women with a history of hypertensive pregnancy.

PURPOSE: Women with a history of hypertensive pregnancy are at greater risk for future cardiovascular events; however, the mechanisms for this increased risk are unknown. Evidence suggests that an exercise stimulus unmasks latent hypertensive tendencies, identifying individuals at the greatest risk for developing cardiovascular disease. The current study examined the hypothesis that women with a hypertensive pregnancy history exhibit an augmented exercise pressor response. METHODS: Normotensive women with a history of healthy pregnancy (CON; n = 9) and hypertensive pregnancy (HP+; n = 12) wer

(In)activity-related neuroplasticity in brainstem control of sympathetic outflow: unraveling underlying molecular, cellular, and anatomical mechanisms.

More people die as a result of physical inactivity than any other preventable risk factor including smoking, high cholesterol, and obesity. Cardiovascular disease, the number one cause of death in the United States, tops the list of inactivity-related diseases. Nevertheless, the vast majority of Americans continue to make lifestyle choices that are creating a rapidly growing burden of epidemic size and impact on the United States healthcare system. It is imperative that we improve our understanding of the mechanisms by which physical inactivity increases the incidence of cardiovascular disease

Reversible reduction of cardiac sympathetic innervation after coronary artery bypass graft surgery: an observational study using serial iodine 123-labeled meta-iodobenzyl-guanidine (MIBG) imaging.

OBJECTIVES: Various types of surgical and interventional procedures have been reported to cause cardiac sympathetic denervation. We aimed at evaluating the effects of coronary artery bypass grafting (CABG) in cardiac sympathetic innervation through meta-iodobenzyl-guanidine (MIBG) imaging. METHODS: MIBG imaging was performed in 21 patients with coronary artery disease (CAD) 1 day before and 1 week and 6 months after CABG with concomitant measurements of corrected QT interval. In each study we evaluated MIBG defect score in a 16-segment left ventricular model, MIBG-defect size (percent) from g

The neurohormonal natural history of essential hypertension: towards primary or tertiary aldosteronism?

Use of the aldosterone-to-renin ratio has controversially suggested that approximately 10% of hypertensives have primary aldosteronism, and most of these individuals are thought to have idiopathic hyperaldosteronism. The usual renin-angiotensin system control is intact in these individuals and is similar to that in low renin and essential hypertensives, differing only in the degree of sensitivity. There is recent evidence suggesting that hyperaldosteronism relates to aldosterone synthase genetic polymorphism, and also that increased angiotensin II stimulation of the adrenal glands appears to p

Characterization of midline medulla role in the trigeminal depressor response.

The purpose of the present investigation was to determine the role of the midline medulla in mediating the trigeminal depressor response. Previously we found that lesions of the midline medulla abolished the decrease in blood pressure resulting from electrical stimulation of the spinal trigeminal complex. Electrical stimulation (5 Hz) of the spinal trigeminal tract elicited a decrease in arterial blood pressure that was associated with an inhibition of sympathetic nerve activity recorded from the inferior cardiac nerve of anesthetized cats. The effect of single shocks applied to the trigeminal

Cardiovascular dopamine receptors: physiological, pharmacological and therapeutic implications.

Dopamine receptor activation can lead to pronounced changes in cardiovascular function. The myriad of effects produced by dopamine receptor agonists results from the activation of dopamine receptors located at different anatomical sites in the cardiovascular system. Further basic research is required to better characterize these dopamine receptors so as to allow the development of more specific dopamine receptor agonists. Endogenous dopamine may be involved in the physiological control of fluid and electrolyte balance and continuing research efforts in this area should provide for a better und

The innervation of sheep mesenteric veins.

1. An isolated preparation of sheep mesenteric vein is described from which contractions of the longitudinal smooth muscle of the adventitia have been recorded in response to stimulation of intramural and periarterial nerves.2. The preparation did not respond to single stimuli but the relation between amplitude of response and frequency of stimulation was characteristic of that described for other smooth muscles innervated by sympathetic nerves.3. Responses were blocked by tetrodotoxin, guanethidine, and by a number of alpha-receptor blocking drugs. Responses to peri-arterial stimulation were

Physician burnout and type D personality relate to pericoronary fat phenotype and role of sympathetic activation: a cross-sectional study.

Biological pathways linking psychosocial stress to coronary pathology remain elusive. Burnout and type D personality (TDP), defined by negative affectivity and social inhibition, are prevalent among physicians and associated with cardiovascular risk. We investigated whether burnout and TDP relate to pericoronary adipose tissue (PCAT) attenuation, a noninvasive imaging marker of fat composition and coronary inflammatory signaling. We further investigated whether sympathetic activation is implicated as an indirect pathway in this association. Systemic inflammation was examined as a downstream co

Sympathetic Nerve Activation Drives Cardiac Fibrosis and Arrhythmia in Lean Metabolic Dysfunction-Associated Steatohepatitis Mice Concurrently Exposed to Ethanol and a High-Fat/High-Cholesterol Diet.

BACKGROUND: Lean metabolic dysfunction-associated steatohepatitis (MASH) is increasingly recognised in non-obese individuals and is associated with cardiovascular complications, including arrhythmias. Although sympathetic activation and inflammation have been implicated in disease progression, their mechanistic roles in cardiac fibrosis and arrhythmogenesis in lean MASH remain unclear. OBJECTIVE: To investigate the contribution of sympathetic nerve activation to myocardial fibrosis and arrhythmia in a lean MASH mouse model. METHODS AND RESULTS: Male C57BL/6J mice were fed a high-fat/high-cho

Ccl12 coordinates immune-neural crosstalk to promote adipose sympathetic remodeling after burn trauma.

Burn injury triggers sustained white adipose tissue (WAT) remodeling and hypermetabolism. While systemic catecholamines are considered primary drivers of this response, local mechanisms remain poorly defined. Here, we identify an intra-fat immune-neural axis that drives sympathetic remodeling and thermogenic reprogramming of WAT after burn. Using a murine scald model, we show that burn injury induces localized sympathetic activation, norepinephrine release, and WAT browning, all abolished by chemical or genetic denervation. Mechanistically, macrophage-derived Ccl12 recruits CCR2+ T cells that

The effects of limb suction during arterial occlusion on arterial baroreflex in humans.

Peripheral venous distension evokes a reflex sympathetic activation [venous distension reflex (VDR)]. Previous study showed that venous distension in a forearm with a volume infusion raised sympathetic baroreflex sensitivity (SBRS). However, the effects of venous distension on baroreflex sensitivity (BRS) in the lower limb have not been examined. We hypothesized that the venous distension in an upper or lower limb by the negative pressure would alter BRS. In 20 healthy subjects, one arm or one leg of the subjects was placed into an airtight pressure tank, although a cuff was placed proximally

Age- and sex-specific differences in myocardial sympathetic tone and left ventricular remodeling following myocardial injury.

BACKGROUND: Presentations and outcomes of acute myocardial infarction (MI) differ between women and men, with the worst outcomes being reported in younger women. Mental stress induced ischemia and sympathetic activation have been suggested to play a prominent role in the pathogenesis of MI in younger women, however, the impact of sex hormones on these parameters remains unknown. METHODS: The effect of sex hormones and age on myocardial infarct size and myocardial sympathetic activity (MSA) was assessed in male and female, as well as young (4-6 months) and aged (20-22 months) FVB/N mice (n = 1

Sympathetic Biomarker Dynamics Post-Myocardial Infarction: TH, PGP9.5, and SYN Expression Discordance in Murine Hearts.

Myocardial infarction (MI) and its sequelae continue to be the leading cause of mortality globally. Following MI, a series of structural pathophysiological changes occur in the myocardium, including sympathetic remodeling. Tyrosine hydroxylase (TH), protein gene product 9.5 (PGP9.5), and synaptophysin (SYN) are recognized as key markers of sympathetic nerves. However, the expression patterns of these biomarkers during sympathetic remodeling, particularly their temporal profiles, remain insufficiently characterized. A cohort of 60 healthy adult male C57BL/6 mice was randomly divided into a cont

Effect of regional crosstalk between sympathetic nerves and sensory nerves on temporomandibular joint osteoarthritic pain.

Temporomandibular joint osteoarthritis (TMJ-OA) is a common disease often accompanied by pain, seriously affecting physical and mental health of patients. Abnormal innervation at the osteochondral junction has been considered as a predominant origin of arthralgia, while the specific mechanism mediating pain remains unclear. To investigate the underlying mechanism of TMJ-OA pain, an abnormal joint loading model was used to induce TMJ-OA pain. We found that during the development of TMJ-OA, the increased innervation of sympathetic nerve of subchondral bone precedes that of sensory nerves. Furthe

Acute Optogenetic Activation of the Subfornical Organ Produces Sympathetically Mediated Increases in Blood Pressure.

INTRODUCTION: The subfornical organ (SFO) is a vital blood pressure-controlling region that elicits blood pressure changes likely via an excitatory (or glutamatergic) projection to the paraventricular nucleus of the hypothalamus (PVN). However, the role of this SFO-PVN pathway in blood pressure control has been poorly defined in the literature. As such, the present study aimed to examine the functional connectivity between the SFO neurons and the PVN and how they intersect to control blood pressure. METHODS: In Lewis rats (n = 10), glutamatergic SFO neurons (SFOglut) were transduced with chan
