Current techniques for aortic valve replacement in the child carry multiple disadvantages. Longer-term follow-up of the Ross procedure has documented disappointing late results for an increasing proportion of patients. Many challenges continue to face the development of a tissue-engineered valve with growth potential. In this setting, aortic valve repair is a useful temporizing procedure that allows a child to have an excellent quality of life, free from the need for anticoagulation and the risk of thromboembolism. Repair techniques are primarily based on the use of autologous pericardium to e

UNLABELLED: Developmental absence of teeth (hypodontia) is associated with characteristic morphological changes in the teeth, alveolar volume deficiencies, and skeletal jaw mal-relationships. Management may be complex, involving several dental specialties, ideally working as a close-knit team. Improved diagnostic and treatment technologies continue to evolve, ever widening the management opportunities for these patients. This paper reviews current treatment methods and looks to what the future may hold.
CLINICAL RELEVANCE: Specialist teams with access to traditional and evolving diagnostic an

Tissue remodeling follows the initial phase of wound healing and stops inflammatory and scar-forming processes, then restores the normal tissue morphology. The human peptide Gly-(L-His)-(L-Lys) or GHK, has a copper 2+ (Cu(2+)) affinity similar to the copper transport site on albumin and forms GHK-Cu, a complex with Cu(2+). These two molecules activate a plethora of remodeling related processes: (1) chemoattraction of repair cells such as macrophages, mast cells, capillary cells; (2) anti-inflammatory actions (suppression of free radicals, thromboxane formation, release of oxidizing iron, trans

This study is aimed at investigating the potentials of ex vivo expansion and pluri-differentiation of cryopreservation of adult human bone marrow mesenchymal stem cells (hMSCs) into chondrocytes, adipocytes and neurocytes. Cryopreserved hMSCs were resuscitated and cultured for 15 passages, and then induced into chondrocytes, adipocytes and neurocytes with corresponding induction medium. The induced cells were observed for morphological properties and detected for expressions of type II collagen, triglyceride or neuron-specific enolase and nestin. The result showed that the resuscitated cells c

Genetic mutations affecting the capacity of basal keratinocytes to adhere firmly to the underneath derma lead to severe, often lethal, blistering disorders of the skin known as Epidermolysis Bullosa (EB). About 400,000-500,000 people worldwide are affected and no definitive treatments have yet been developed. Gene therapy might represent an alternative therapeutic approach for these devastating inherited disorders. In the last 10 years pre-clinical studies have shown that human epidermal stem cells can be stably transduced using integrating vectors allowing long-term genetic correction of the

A promising strategy for the regeneration of degenerated cartilage tissue structure in osteoarthritic joints is the use of mesenchymal precursor cells. These cells can be triggered to undergo differentiation into functional active chondrocytes resulting in newly synthesized cartilage. Because chondrogenic differentiation is initiated by the step of mesenchymal condensation in vitro, it is of great interest to fully characterize the first lineage specific step in vitro. Therefore, a modified culture system was developed which mimics the process in vitro and may finally help to identify the key

The most common causes of disability and death are diseases of the heart, lungs, liver, kidneys, and pancreas, many of which are potentially treated by organ transplantation. The effect of organ dysfunction and failure will likely grow over time, and patients will increasingly expect "safer" transplants, in particular in cases of "preemptive transplantation." New technologies are being developed in part because of the limited availability of organs, and include transplantation with stem cells, tissue engineering, cloning, and xenotransplantation, which some researchers believe promise ready so…

Aplasia cutis congenita (ACC) is an uncommon congenital malformation. It is characterized by defects of the skin that occur most frequently on the scalp along the midline, but can also be localized on the trunk, face and limbs, usually with a symmetrical distribution. When it is localized in the skull it can extend to the dura mater, leaving only the thin pia mater to protect the brain. The most common complications related to this disorder are infection, hemorrhage, and, in defects localized on the vertex, meningitis and bleeding from the sagittal sinus can occur with dramatic consequences. Apligraf, a manufactured living human skin equivalent, was the first true composite tissue analog to become commercially available. It has been approved by the FDA for the treatment of venous leg ulcers and diabetic foot ulcers. The tissue engineered bi-layered skin equivalent has both an epidermis and a dermis, which consist of keratinocytes and fibroblasts derived from neonatal foreskin and bovine collagen. The skin equivalent produces a great number of cytokines and growth factors and is immunologically well tolerated. The product is easy to handle in clinical use and can be applied in an o

Despite progress in mouse models of bacterial pathogens, studies are often limited by evaluating infections in an individual organ or tissue or at a given time. Here we present a technique to engineer the pathogen, e.g., Brucella melitensis, with a bioluminescent marker permitting analysis of living bacteria in real time during the infectious process from acute to chronic infection. Using this bioluminescent approach, tissue preference, differences between virulent and mutant bacteria, as well as the response of the bacteria to host metabolites can provide extraordinary data enhancing our unde

Osteoarthritis (OA), as a degenerative disease, leads to high socioeconomic burdens and disability rates. The knee joint is typically the most affected and is characterized by progressive destruction of articular cartilage, subchondral bone remodeling, osteophyte formation and synovial inflammation. The current management of OA mainly focuses on symptomatic relief and does not help to slow down the advancement of disease. Recently, mesenchymal stem cells (MSCs) and their exosomes have garnered significant attention in regenerative therapy and tissue engineering areas. Preclinical studies have …

Osteochondral defects remain a major clinical challenge mainly due to the combined damage to the articular cartilage and the underlying bone, and the interface between the two tissues having very different properties. Current treatment modalities have several limitations and drawbacks, with limited capacity of restoration; however, tissue engineering shows promise in improving the clinical outcomes of osteochondral defects. In this study, a novel gradient scaffold has been fabricated, implementing a gradient structure in the design to mimic the anatomical, biological and physicochemical proper…

Tissue engineering holds potential in the treatment of intervertebral disc degeneration (IDD). However, implantation of tissue engineered constructs may cause foreign body reaction and aggravate the inflammatory and oxidative microenvironment of the degenerative intervertebral disc (IVD). In order to ameliorate the adverse microenvironment of IDD, in this study, we prepared a biocompatible poly (ether carbonate urethane) urea (PECUU) nanofibrous scaffold loaded with fucoidan, a natural marine bioactive polysaccharide which has great anti-inflammatory and antioxidative functions. Compared with …

Despite antiretroviral therapy's success in human immunodeficiency virus (HIV) management, no cure or preventive vaccine exists; three-dimensional (3D) human tissue models-emerging from biomedical research, tissue engineering, and microfluidics-offer new potential, yet a scientometric analysis of their progress remains lacking. We reviewed the current status of three in vitro 3D models for HIV research: organoids, organ-on-a-chip, and 3D bioprinting. We conducted a bibliometric comparative analysis of 3D human tissue models in HIV research. A total of 852 documents published between 2014 and 2…
