Liver sinusoidal endothelial cells (LSECs) are highly specialized endothelial cells forming the hepatic sinusoidal wall. Besides their high endocytic potential, LSECs have been demonstrated to markedly contribute to liver homeostasis and immunity, and may partially explain unexpected hepatotoxicity of drug candidates. However, their use for in vitro investigations is compromised by poor cell yields and a limited proliferation capacity. Here, we report the transient expansion of primary human LSECs from three donors by lentiviral transduction. Transduced ("upcyte®") LSECs were able to undergo a

Macroautophagy/autophagy, a mechanism of degradation of intracellular material required to sustain cellular homeostasis, is exacerbated under stress conditions like nutrient deprivation, protein aggregation, organelle senescence, pathogen invasion, and hypoxia, among others. Detailed in vivo description of autophagic responses triggered by hypoxia is limited. We have characterized the autophagic response induced by hypoxia in Drosophila melanogaster. We found that this process is essential for Drosophila adaptation and survival because larvae with impaired autophagy are hypersensitive to low o

BACKGROUND: ADNP is essential for embryonic development. As such, de novo ADNP mutations lead to an intractable autism/intellectual disabili … nces (e.g., cell cycle), accentuated in females (with significant effects on antigen processing and cellular senescence) and corrected by NAP. Differentially expressed, NAP-correctable transcripts, including the autophagy and microbiome resilience-linked FOXO3, were also deregulated in human patient-derived ADNP-mutated lymphoblastoid cells. There were also Tyr sex-s …

Peroxiredoxin 3 (PRX3), a typical 2-Cys peroxiredoxin located exclusively in the mitochondrial matrix, is the principal peroxidase responsible for metabolizing mitochondrial hydrogen peroxide, a byproduct of cellular respiration originating from the mitochondrial electron transport chain. Mitochondrial oxidants are produced in excess in cancer cells due to oncogenic transformation and metabolic reorganization, and signals through FOXM1 and other redox-responsive factors to support a hyper-proliferative state. Over-expression of PRX3 in cancer cells has been shown to counteract oncogene-induced

BACKGROUND: Fibroblast behaviour is a key determinant of outcomes in interstitial lung diseases (ILDs), yet mechanisms governing the switch between reversible repair and progressive fibrosis remain unclear. How disease-specific cellular niches shape fibroblast fate across ILD phenotypes has not been compared in situ.
METHODS: We profiled peripheral lung tissues from controls, organising pneumonia (OP), connective tissue disease-associated ILD (CTD-ILD), and idiopathic pulmonary fibrosis (IPF) using High-Definition Visium spatial transcriptomics. A matched single-cell RNA-seq atlas was integra

BACKGROUND: During sepsis, serve vascular dysfunctions lead to life-threatening multiple organ failure, due to vascular smooth muscle cells (VSMC) impairments, resulting in vasoplegia, hypotension and hypoperfusion. In addition, septic patients have an altered cell metabolism that leads to lactic acidosis. Septic patients suffering from lactic acidosis have a high risk of mortality. In addition, septic survivors are at risk of secondary vascular disease. The underlying mechanisms of whether and how lactic acidosis leads to the changes in VSMCs is not well understood. The aim of this study was

Cyclin-dependent kinase 4/6 inhibitors (CDK4/6i) are standard therapies for estrogen receptor-positive breast cancer, but their efficacy is limited by drug resistance. While dysregulation of alternative splicing has been implicated in cancer progression, the underlying mechanisms remain unclear. Here, we uncover a splicing-mediated mechanism whereby exon 2 inclusion in SUV39H2 drives its overexpression in CDK4/6i-resistant breast cancer cells. Clinically, elevated exon 2 inclusion in SUV39H2 correlates with poor prognosis in patients with breast cancer. Targeting SUV39H2 with gapmer antisense

BACKGROUND: Cancer-associated fibroblasts (CAFs) are critical components of the tumor microenvironment in endometrial cancer (EC). Primary isolation and culture of CAFs serve as the standard method for investigating their biological roles. However, it remains unclear whether prolonged in vitro culture alters the intrinsic properties of CAFs, potentially affecting experimental reproducibility. This study aims to evaluate the phenotypic and transcriptomic evolution of primary EC-derived CAFs across different passages.
METHODS: CAFs were isolated from human EC tissues using enzymatic digestion a

BACKGROUND: Liver fibrosis represents a frequent pathological outcome of chronic hepatic insults. As a bioactive constituent of ginger, 6-shogaol has shown hepatoprotective potential. Hepatic stellate cell (HSC) activation is widely regarded as an important driver in the occurrence and advancement of liver fibrosis. However, whether 6-shogaol can regulate HSC activation remains to be dissected.
PURPOSE: To assess the anti-fibrotic effects of 6-shogaol and elucidate the molecular mechanisms, we focused on its impact on the fate of HSCs and the subsequent alleviation of liver fibrosis.

RATIONALE: Chemoresistance severely limits therapeutic options for advanced bladder cancer. Histone deacetylases (HDACs) have been implicated in tumour progression and treatment resistance, yet their role in acquired chemoresistance remains incompletely defined. We investigated whether epigenetic modulation could restore chemotherapy sensitivity in drug-refractory bladder cancer.
METHODS: Expression of HDAC1-3 was analysed in tumour tissues and correlated with clinical outcomes and chemoresistance-associated transcriptional programmes. A co-delivery therapeutic strategy combining the pan-HDAC

Osteoarthritis (OA) is a degenerative joint disease characterized by cartilage degradation and chondrocyte senescence. Emerging evidence implicates NADPH oxidase 4 (Nox4) in OA pathogenesis. This study investigated the role of Nox4 in regulating extracellular matrix (ECM) homeostasis and chondrocyte senescence through the p38 mitogen-activated protein kinase (p38MAPK) signaling pathway. We found that Nox4 expression was significantly upregulated in OA chondrocytes and cartilage. In a post-traumatic OA mouse model (Anterior cruciate ligament transection, ACLT), Nox4 knockout attenuated cartilag

In multiple myeloma (MM), resistance to B-cell maturation antigen (BCMA)-targeted chimeric antigen receptor T-cell (CAR-T) treatment continues to limit therapeutic benefit, whereas the tumor-intrinsic factors governing sensitivity to CAR-T remain poorly understood. Here, integrating public single-cell RNA-seq data with transcriptomic profiling of MM cells before and after CAR-T exposure, we identified the mitochondrial protein PPTC7 as a key modulator linking tumor mitophagy to innate immune activation and CAR-T efficacy. Mechanistically, PPTC7 acted as a negative regulator of tumor-intrinsic

Intervertebral disc degeneration (IDD), a condition characterized by extracellular matrix (ECM) degradation and nucleus pulposus (NP) cell senescence, compromises disc function. The Akt signaling pathway is essential for NP cell viability and ECM homeostasis. This study identifies the serine/threonine kinase PIM3 as significantly downregulated in degenerated human NP tissues. Notably, PIM3 mRNA expression levels were found to be negatively correlated with the clinical severity of IDD (Pfirrmann grade). The functional role of PIM3 was investigated through knockdown and overexpression experiment

Age-related macular degeneration (AMD) is the most prevalent form of irreversible blindness worldwide in the elderly population. In our previous studies, we found that deficiencies in the nuclear factor, erythroid 2 like 2 (NFE2L2) and peroxisome proliferator-activated receptor gamma coactivator 1-α (PGC-1α) genes caused AMD-like pathological phenotypes in mice. In the present work, we show hijacked epithelial-mesenchymal transition (EMT) due to the common loss of PGC-1α and NFE2L2 (double knock-out, dKO) genes in aged animals. The implanted area was assessed by histology, immunohistochemistry

BACKGROUND: Aging is characterized by adipose tissue senescence, inflammation, and fibrosis, with trunk fat accumulation. Aging HIV-infected patients have a higher risk of trunk fat accumulation than uninfected individuals-suggesting that viral infection has a role in adipose tissue aging. We previously demonstrated that HIV/SIV infection and the Tat and Nef viral proteins were responsible for adipose tissue fibrosis and impaired adipogenesis. We hypothesized that SIV/HIV infection and viral proteins could induce adipose tissue senescence and thus lead to adipocyte dysfunctions.

The tumor suppressor p53 acts as a transcription factor that regulates the expression of a number of genes responsible for DNA repair, cell cycle arrest, metabolism, cell migration, angiogenesis, ferroptosis, senescence, and apoptosis. It is the most commonly silenced or mutated gene in cancer, as approximately 50% of all types of human cancers harbor TP53 mutations. Activation of p53 is detrimental to normal cells, thus it is tightly regulated via multiple mechanisms. One of the recently identified regulators of p53 is RNA-binding motif protein 10 (RBM10). RBM10 is an RNA-binding protein freq

Canities (or hair greying) is an age-linked loss of the natural pigment called melanin from hair. While the specific cause(s) underlying the loss of melanogenically-active melanocytes from the anagen hair bulbs of affected human scalp remains unclear, oxidative stress sensing appears to be a key factor involved. In this study, we examined the follicular melanin unit in variably pigmented follicles from the aging human scalp of healthy individuals (22-70 years). Over 20 markers were selected within the following categories: melanocyte-specific, apoptosis, cell cycle, DNA repair/damage, senescen

Cancer is driven by genetic change, and the advent of massively parallel sequencing has enabled systematic documentation of this variation at the whole-genome scale1-3. Here we report the integrative analysis of 2,658 whole-cancer genomes and their matching normal tissues across 38 tumour types from the Pan-Cancer Analysis of Whole Genomes (PCAWG) Consortium of the International Cancer Genome Consortium (ICGC) and The Cancer Genome Atlas (TCGA). We describe the generation of the PCAWG resource, facilitated by international data sharing using compute clouds. On average, cancer genomes contained

It has been previously demonstrated that hydrogen sulfide (H2S) prevents formaldehyde (FA)-induced neurotoxicity. However, the exact mechanisms underlying this protection remain to be fully elucidated. Neuronal senescence is involved in FA-induced neurotoxicity. Leptin signaling has anti-aging function. The present work was to investigate the protection of H2S against FA-induced neuronal senescence and the mediatory role of leptin signaling. FA-exposed HT-22 cells were used as the vitro model of FA-induced neuronal senescence. The senescence-associated β-galactosidase (SA-β-Gal) positive cell
