KDAC6 alters cell morphology and motility through positive and negative modulation of F-actin distribution.

Lysine deacetylase 6 (KDAC6 or HDAC6) has been associated with cell motility and deacetylation of cytoskeleton-related proteins. KDAC6 is the only human KDAC having three identified major domains, including two catalytic domains (CD). Defining the roles of KDAC6 in cell motility has been constrained by limited understanding of domain-specific contributions. Live and fixed cell imaging was used to characterize the effects of KDAC6 on F-actin distribution, cell morphology, and cell motility of genetically modified HT1080 cells containing KDAC6 with an inactivated CD or knockout. CD1 inactivation…

O-GlcNAcylation Mediates BMP2-Induced Osteogenesis/Cementogenesis via NMIIA.

The precise cellular origin and regulatory mechanisms underlying cementum development remain poorly understood, hindering progress toward id … entum formation and delaying alveolar socket healing. Mechanistically, we further revealed that the cytoskeleton, especially MYH9 (nonmuscle myosin IIA, NMIIA), is O-GlcNAcylated and is essential for BMP2-induced osteogenic/cementogenic differentiation. These findings demonstrate that O-GlcNAcylation is essential for cellular cementum formation by modulating t …

Colchicine Suppresses Adipogenic Differentiation of Mesenchymal Stem Cells: Implications for Bone Adiposity Control.

Background: Gout is an inflammatory arthritis associated with increased bone anabolism and a higher risk of ectopic bone formation. Conclusions: These findings indicate that colchicine can suppress adipogenic differentiation while favouring osteoblast commitment in mesenchymal stem cells. Podocyte-specific deletion of Il12rb1 attenuated podocyte senescence, ameliorated glomerular injury, and reduced proteinuria in both mice who were lupus prone and normal mice injected with a nephrotoxic serum. Loss of α-catenin in germ cells impairs spermatogenesis and flagellar function associated with teratozoospermia.

Infertility affects approximately 10%-20% of couples worldwide, with male factors accounting for roughly half of all cases. Teratozoospermia … In adherens junctions (AJs), α-catenin ( CTNNA1) anchors the cadherin-catenin complex to the actin cytoskeleton and regulates actin organization, but its role in germ cell development and sperm function remains unknown. We generated a germ cell-specific Ctnna1 conditional knockout mouse model ( Ctnna1 cKO). Homozygous Ctnna1-deficient mice exhibited male infe …

A molecular and spatial resource defining tubulin isotype organization during corneal development.

Microtubules are essential components of the cytoskeleton that support epithelial organization, polarity, and tissue morphogenesis. They are composed of α- and β-tubulin heterodimers, each encoded by distinct genes that generate closely related but functionally distinct isotypes. Although several tubulin isotypes have been implicated in ocular development and disease, how isotype diversity is organized during corneal morphogenesis remains poorly defined. Herein, we use the developing chick embryo as a model system to investigate the conservation and spatiotemporal localization of tubulin isoty…

Mechanistic study of ARHGAP27 promoting the progression of aortic dissection by regulating the RhoA/ROCK/YAP pathway.

BACKGROUND: Aortic dissection (AD) is a life-threatening cardiovascular disease. ARHGAP27 can regulate cytoskeleton and cellular functions, which may be related to the phenotypic switching of vascular smooth muscle cells (VSMCs) in AD. However, the role of ARHGAP27 in AD has not been reported yet. METHODS: The AD-related data sets were downloaded from GEO database to screen differential genes. HE and IHC staining was used to detect the pathological changes and the expression level of ARHGAP27 in AD tissue. AD cell models were constructed in vitro by inducing HAVSMCs with PDGF-BB to evaluate t…

Regulation of axon outgrowth during mushroom body development by acetylated α-tubulin lysine 394 and Tau.

Disruptions in the microtubule cytoskeleton play a role in various neurological diseases that afflict a large fraction of the population. Microtubule function is regulated by post-translational modifications like acetylation, and one consistently identified acetylation site in mammals and Drosophila melanogaster is α-tubulin lysine 394 (K394). Our previous research demonstrated that an acetylation-blocking point mutation-K394R-causes a decrease in microtubule stability in axon terminals at the developing neuromuscular junction. Here, we asked whether K394 acetylation regulates the development …

Inactivation of cofilin-1 in Mcpt5-Cre-nf-Cfl1fl/fl mice prevents the formation of connective tissue mast cells without affecting basophils: a new tool to investigate the specific role of CTMCs in disease.

Actin-binding proteins play a critical role in regulating the dynamic rearrangement of the actin cytoskeleton, which is essential for maintaining cellular homeostasis and facilitating various processes in eukaryotic cells. Cofilin-1 (Cfl1), an actin-binding protein, promotes the severing and depolymerization of actin filaments. To investigate the function of Cfl1 in mast cells, we generated Mcpt5-Cre-nf-Cfl1fl/fl knock-in mice, expressing a non-functional form of Cfl1 (nf-Cfl1) instead of wildtype Cfl1 under the control of the connective tissue mast cell (CTMC)-specific promoter mast cell prot…

Utrophin requires α-Syntrophin to maintain neuromuscular junction integrity in mdx mice.

Dystrophin links the actin cytoskeleton to the extracellular matrix through the dystrophin-glycoprotein complex (DGC), providing structural stability to muscle fibers. In mdx mice, which lack dystrophin, neuromuscular junctions (NMJs) remain largely structurally and functionally intact despite extensive muscle pathology. Here, using single- and double-mutant mice deficient in dystrophin and α-syntrophin (α-syn), we investigated how utrophin upregulation contributes to NMJ maintenance in dystrophic muscle. During early postnatal development, when dystrophic muscles are transiently resistant to …
