Caylobolide B: Structure Revision, Total Synthesis, Biological Characterization, and Discovery of New Analogues.

The unique potential of marine polyhydroxylated macrolides in chemical biology and drug discovery has long been constrained by their structural complexity and limited material availability, frustrating efforts in stereochemical assignment, synthesis, and mechanism-of-action elucidation. Here, we establish an integrated workflow, combining chemogenomic profiling, ultra-high-resolution NMR, and modular total synthesis, for the comprehensive functional and structural interrogation of this challenging natural product class. Applying this approach to caylobolides, natural products isolated from sca

Acridine Derivatives as Antifungal and Antivirulence Agents Against Candida albicans.

Candida albicans is a clinically important fungal pathogen capable of causing both superficial and systemic infections, particularly in immunocompromised individuals. A key factor contributing to its pathogenicity is its ability to form biofilms, structured microbial communities that confer significant resistance to conventional antifungal therapies. Addressing this challenge, we explored the antivirulence potential of acridine derivatives, a class of heterocyclic aromatic compounds known for their diverse biological activities, including antimicrobial, antitumor, and antiparasitic properties.

Activity of cloxacillin against Cutibacterium acnes: MIC evaluation and utility of gradient test.

BACKGROUND: Cutibacterium acnes is a cause of implant-associated infections, often co-occurring with methicillin-susceptible Staphylococcus aureus (MSSA), especially in orthopedic settings. In Scandinavia, cloxacillin may be used as initial empiric intravenous treatment if MSSA implant-associated infection is suspected. Clinical breakpoints for C. acnes susceptibility to cloxacillin or other isoxazolyl-penicillins are lacking to guide therapy if C. acnes is subsequently cultured after empiric cloxacillin therapy has been initiated. OBJECTIVE: We aimed to compare MIC distributions of C.acnes t

Membrane-Targeting Poly(Amino Acids) with a Phage-Like Action Process for Antifungal Therapy.

Invasive fungal infections and the rising prevalence of drug resistance highlight the urgent need for alternative antifungal strategies. In this study, a guanidine-functionalized poly(amino acid) (PArg20) with a phage-like action process was synthesized, exhibiting superior antifungal activity and favorable biocompatibility. Emulating the three-step infection process of bacteriophages, PArg20 exhibits a phage-like "adsorption-penetration-disruption" action process. Initially, electrostatic interactions contribute to membrane adsorption for targeting, followed by membrane penetration driven by

In vitro evidence to support amphotericin B and flucytosine combination therapy for talaromycosis.

BACKGROUND: Talaromyces marneffei causes talaromycosis, a life-threatening fungal disease with limited treatment options. The standard treatment of amphotericin B (AmB) induction followed by itraconazole consolidation still results in 15% to 30% mortality. This study aimed to investigate the potential of AmB and flucytosine (5FC) combination therapy to enhance antifungal activity. METHODS: The in vitro antifungal activity of AmB and 5FC alone and in combination against 60 T. marneffei clinical isolates was evaluated using a validated colorimetric antifungal susceptibility assay and the checke

Antibacterial Peptides-Produced Cell Hydrogel Eliminates Intracellular Pathogens and Remodels Immune Microenvironment for Osteomyelitis Therapy.

Featuring bacterial invasion and colonization within cells, chronic infection-induced immune suppression, and inflammatory cell infiltration, osteomyelitis is currently an intractable and recurrent bone disease. In this study, an injectable hydrogel that gels in situ and is loaded with engineered antimicrobial cells (LL37-MSC@OCAHM) is developed. This anti-inflammatory hydrogel not only maintains cell activity in the inflammatory environment but also releases magnesium ions (Mg2+) to promote the differentiation of MSCs into bone-forming cells, contributing to bone mass formation, enhancing bon

Antibacterial potential of Thymus vulgaris (Thyme) oil against Escherichia coli strains isolated from food.

The search for meat-based foods, even if it allows to increase the shelf life of commercialized foods, can lead to contamination by pathogenic microorganisms. In this sense, the present study aimed to test the potential antibacterial effect of Thymus vulgaris (thyme) essential oil against food-borne strains of the bacterium Escherichia coli. Therefore, the Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) analyses were performed on microdilution plates. In addition, the study of the association of the product with synthetic antimicrobials, performed by the dis

Cobalt(II) Complexes of 4'-Bromo-Fenamic Acid: Antioxidant Properties, Antibacterial Activity, and Interaction with DNA and Albumins.

The reaction of 4'-bromo-fenamic acid, a bromo-derivative of fenamic acid (the scaffold of the fenamate non-steroidal anti-inflammatory drugs), with Co(II) in the absence or presence of various nitrogen-donor ligands yielded nine novel, neutral mononuclear Co(II) complexes. These complexes were characterized by physicochemical and spectroscopic techniques and single-crystal X-ray crystallography. The biological evaluation of the compounds focused on their antioxidant and antimicrobial efficacy, as well as their interaction with calf-thymus DNA, pBR322 plasmid DNA (in the absence or presence of

Identification of a Novel Antibacterial Function of Mammalian Calreticulin.

Calreticulin is a highly conserved and multifunctional molecular chaperone ubiquitously expressed in humans and animals. Beyond its well-established roles in calcium homeostasis, protein folding, and immune regulation, recent studies in aquatic species have suggested a previously unrecognized antimicrobial function of calreticulin. These findings raise the question of whether calreticulin also exerts antibacterial activity in terrestrial mammals, which has not been systematically investigated to date. To address this knowledge gap, we successfully constructed and expressed recombinant goat cal

Beyond the usual suspects: A case report of multi-etiological and multidrug-resistant Candida onychomycosis.

This is a rare case of Candida onychomycosis caused by three Candida species that were resistant to several antifungal drugs. A 56-year-old Iranian woman with no previous medical or injury history experienced discoloration and lifting of her left thumbnail for six months. Laboratory tests identified yeast and pseudohyphae consistent with Candida onychomycosis. Conventional and molecular techniques confirmed that onychomycosis was caused by three different Candida species. All strains were resistant to various antifungal drugs. Therapeutic failure, in addition to multi-drug resistance, may be d

Octenidine Lozenges Intended for Oral Administration Display In Vitro Activity Against Oropharyngeal Pathogens and Safety Toward Intestinal Microbiota.

Pharyngitis is a leading cause of outpatient antibiotic use, despite its typically viral or self-limiting nature. Such unnecessary antibiotic therapies are not only the cause of increasing antibiotic resistance, but also significant changes in the human microbiota in the intestines and other locations, which translate into immune disorders and an increased risk of developing several chronic diseases. Orally administered octenidine-containing lozenges provide a topical alternative; however, their effects on the host microbiota of the oral cavity, throat, and intestine remain unclear. In this st

Precision Dosing of Meropenem in a Neonate on CARPEDIEM Dialysis: A Grand Round.

BACKGROUND: Optimal antibiotic dosing is challenging in critically ill neonates because of the substantial pharmacokinetic variability, which is influenced by factors such as immature renal function, body composition, and critical illness. The use of extracorporeal therapies adds complexity, making predictions difficult in many cases. Meropenem, a broad-spectrum antibiotic, is commonly used due to resistant gram-negative organisms in neonates; however, dosing guidelines for neonates on CARPEDIEM dialysis are lacking. METHODS: This is a case of a neonate with liver failure of unclear etiology,

Antibacterial therapy in central nervous system infections - breakpoints versus minimal inhibitory concentrations related to body fluid levels.

Breakpoints to define the susceptibility of pathogens to antibiotics are becoming increasingly popular as guidance for antimicrobial therapy. Some breakpoints consider divergent concentrations of antibiotics in different compartments, others list one breakpoint for infections at all sites. Compared to the determination of exact minimal inhibitory concentrations (MICs) and relation of these MICs to concentrations achievable in ventricular CSF, the exclusive use of breakpoints for central nervous system (CNS) infections is a setback. The lack of the determination of exact MICs may be a risk, par
