Advances in Exosome Research in Renal Cell Carcinoma: Mechanisms, Biomarkers, and Therapeutic Applications.

Renal cell carcinoma (RCC) is a common malignant neoplasm of the urinary system, characterized by the difficulty of early diagnosis and the frequent emergence of therapeutic resistance in advanced stages, both of which profoundly compromise patient prognosis. Exosomes, as nanoscale vesicles secreted by cells, are capable of transporting bioactive molecules such as proteins and nucleic acids, thereby mediating communication within the tumor microenvironment and offering novel avenues for the noninvasive diagnosis and treatment of RCC. This article reviews recent advances in exosome research in …

Exploiting the role of milk extracellular vesicles: a comprehensive analysis on isolation methods, characterization, surface modifications, and their therapeutic applications.

Milk EVs (MEVs) are a class of extracellular vesicles (EVs) that have attracted significant attention for their potential as natural nanocarriers in drug delivery. These nanosized vesicles (30 - 150 nm) are composed of a lipid bilayer that encapsulates proteins, nucleic acids, and lipids. The biogenesis of EVs involves a tightly regulated "endocytosis-fusion-secretion" pathway, and they are secreted by diverse cells into physiological fluids (blood, urine, and saliva). The isolation and characterization methods of MEVs are essential to achieve high purity, structural integrity, size distributi…

A review of recent advances in exosome-mediated drug delivery for regenerative therapy and immunomodulation.

Exosomes are small, nanoscale extracellular vesicles that facilitate intercellular communication through the transport of proteins, lipids, and nucleic acids. Recent advances in utilizing exosomes as promising vehicles for targeted delivery have opened up numerous opportunities for establishing next-generation exosome-based nanocarriers and therapeutics for multiple biomedical domains. Their inherent biocompatibility, low immunogenicity, nanoscale size and ability to naturally cross biological barriers make them promising alternatives to traditional synthetic drug-delivery systems such as lipo…

Knocking Down Long Noncoding RNAs Using Antisense Oligonucleotide Gapmers.

Long noncoding RNAs (lncRNAs) are transcripts of 200 nucleotides or longer that are not translated into protein. lncRNAs are highly abundant … emically modified wings, such as 2'-O-methyl, 2'-O-methoxyethyl, constrained ethyl (cEt), or locked nucleic acids (LNAs) that enhance stability and affinity. This mechanism makes them particularly effective for silencing nuclear-retained lncRNAs, a key advantage over cytoplasm-acting methods like RNAi. Because RNase H activity is enriched in nuclei, gapmer-base …

Technological advances in microfluidic electroporation for biomedical applications.

Intracellular delivery is a fundamental enabling technology in biomedicine, allowing exogenous nucleic acids, proteins, drugs and nanomaterials to access intracellular targets and regulate cellular functions. This capability supports disease mechanism studies, cell engineering, therapeutic development and diagnostic analysis. However, conventional bulk electroporation is limited by high voltages, non-uniform electric fields, Joule heating, electrochemical by-products and reduced cell viability. Microfluidic electroporation integrates microscale cell handling with spatially confined electric fi…

Artificial intelligence in molecular diagnostics for pandemic preparedness.

INTRODUCTION: Molecular diagnostics focusing on the detection and analysis of nucleic acids are indispensable tools for early pathogen identification, transmission monitoring, and genomic surveillance during pandemics. Recent technological advances have broadened the diagnostic landscape, incorporating PCR-based methods, isothermal amplification, high-CRISPR-based amplification detection, and sequencing. Despite their diagnostic potential, widespread implementation remains limited by high validation costs, time and logistical constraints, the need for specialized professional knowledge, and a …

Extracellular Vesicles: Classification, Biological Functions, Diseases, and Therapeutic Opportunities.

Extracellular vesicles (EVs) are lipid bilayer-enclosed particles released by virtually all cell types and act as mediators of intercellular communication in physiology and disease. By transporting diverse bioactive molecules, including proteins, lipids, and nucleic acids, EVs regulate gene expression, modulate immune responses, and support tissue homeostasis. Rapid advances have revealed their heterogeneity, complex biogenesis, and broad functional relevance, as well as their importance as biomarkers and therapeutic delivery systems. The molecular principles governing selective cargo loading …

Extracellular Vesicles from Lactic Acid Bacteria: Emerging Postbiotic Effectors in Host-Microbe Crosstalk.

This article addresses extracellular vesicles derived from lactic acid bacteria (LAB-EVs), which have become fundamental mediators of host-microbe communication, yielding new insights about the molecular mechanisms underlying probiotic and postbiotic activity. The review integrates current evidence on the biogenesis, structural characteristics, and functional roles of LAB-EVs, stressing their capacity to transport bioactive cargos - including proteins, nucleic acids, and metabolites - to host cells. We highlight how LAB-EVs interact with the intestinal epithelial barrier, modulate innate and a…

Efforts Toward XNA PCR: Optimization and Characterization of PCR-Amplification of Chimeric 2'F XNA/DNA.

Xeno Nucleic Acids (XNA) are chemically modified versions of DNA. These modifications can bypass natural immune responses and impart novel properties in complex biological systems. Previous efforts have identified mutant proteins capable of robust XNA synthesis; one mutant, SFM4-3, has been shown to be able to amplify partially substituted XNA in the polymerase chain reaction (PCR) which is necessary for many applications of XNA. However, SFM4-3 mediated PCR is inefficient and has required up to 22 h of reaction time to create amplicons of ∼75 nucleotides (nt); optimizing the PCR amplification…

Optimizing macrophage-targeted intracellular delivery systems for safe and effective immunotherapies.

Recent studies increasingly highlight the crucial role of macrophages in tissue development, homeostasis and inflammation. Consequently, macrophages have become an attractive therapeutic target in the rapidly evolving field of immunotherapy, ranging from alleviating autoimmune diseases to modulating the tumor microenvironment. However, enhancing the functions of these cells typically requires ex or in vivo intracellular delivery of cell-impermeable functional molecules, such as nucleic acids. Traditionally, this is achieved using viral and non-viral carriers or methods involving membrane-disru…

Deciphering the Specificity of Reversible DNA-Phosphate ADP-Ribosylation via the Precise Synthesis and Enzymatic Profiling of Nucleotide-Phospho-ADP-Ribosyl Probes.

ADP-ribosylation (ADPr), long recognized as a canonical protein post-translational modification, has recently expanded to include targeting nucleic acids, uncovering a diverse landscape of noncanonical biological functions. Emerging evidence suggests that ADPr at the 5'-phosphate terminus of DNA is implicated in the DNA damage response, yet understanding its precise molecular function has been hampered by the lack of structurally defined chemical probes. Here, we report the stereoselective synthesis of deoxynucleotide-phospho-ADPr (dN-P-ADPr) probes, representing native fragments of terminal D…

Relationship Between Filament-Polymerizing Muscle Myosin and Droplets Generated by Liquid-Liquid Phase Separation.

From individual living cells to the tissues and organs of multicellular organisms, the internal fluids contain numerous macromolecules such as nucleic acids and proteins. Liquid-liquid phase separation (LLPS) is proving to be useful in understanding macromolecular-mediated phenomena that occur within cells, which have been hard to explain through conventional interactions between biological factors. This LLPS is applicable not only to phenomena at scales equivalent to cells but also to those at much larger scales. We studied the association between the behaviors of myosin, a representative mol…
