Enhanced binding of calmodulin to the ryanodine receptor corrects contractile dysfunction in failing hearts.

AIMS: The channel function of the cardiac ryanodine receptor (RyR2) is modulated by calmodulin (CaM). However, the involvement of CaM in aberrant Ca(2+) release in diseased hearts remains unclear. Here, we investigated the pathogenic role of defective CaM binding to the RyR2 in the channel dysfunction associated with heart failure. METHODS AND RESULTS: The involvement of CaM in aberrant Ca(2+) release was assessed in normal and pacing-induced failing canine hearts. The apparent affinity of CaM for RyR2 was considerably lower in failing sarcoplasmic reticulum (SR) compared with normal SR. Thus

Inflammation alters regional mitochondrial Ca²+ in human airway smooth muscle cells.

Regulation of cytosolic Ca(2+) concentration ([Ca(2+)](cyt)) in airway smooth muscle (ASM) is a key aspect of airway contractility and can be modulated by inflammation. Mitochondria have tremendous potential for buffering [Ca(2+)](cyt), helping prevent Ca(2+) overload, and modulating other intracellular events. Here, compartmentalization of mitochondria to different cellular regions may subserve different roles. In the present study, we examined the role of Ca(2+) buffering by mitochondria and mitochondrial Ca(2+) transport mechanisms in the regulation of [Ca(2+)](cyt) in enzymatically dissoci

Cardiac basal metabolism: energetic cost of calcium withdrawal in the adult rat heart.

AIM: Cardiac basal metabolism upon extracellular calcium removal and its relationship with intracellular sodium and calcium homeostasis was evaluated. METHODS: A mechano-calorimetric technique was used that allowed the simultaneous and continuous measurement of both heat rate and resting pressure in arterially perfused quiescent adult rat hearts. Using pharmacological tools, the possible underlying mechanisms related to sodium and calcium movements were investigated. RESULTS: Resting heat rate (expressed in mW g(-1)(dry wt)) increased upon calcium withdrawal (+4.4 +/- 0.2). This response was

TRP channels in disease.

The transient receptor potential (TRP) channels are a large family of proteins with six main subfamilies termed the TRPC (canonical), TRPV (vanilloid), TRPM (melastatin), TRPP (polycystin), TRPML (mucolipin), and TRPA (ankyrin) groups. The sheer number of different TRPs with distinct functions supports the statement that these channels are involved in a wide range of processes ranging from sensing of thermal and chemical signals to reloading intracellular stores after responding to an extracellular stimulus. Mutations in TRPs are linked to pathophysiology and specific diseases. An understandin

Distinct Ca(2+) signalling mechanisms induced by ATP and sphingosylphosphorylcholine in porcine aortic smooth muscle cells.

1. The increase in the cytosolic Ca(2+) concentration ([Ca(2+)](i)) following repetitive stimulation with ATP or sphingosylphosphorylcholine (SPC) in single porcine aortic smooth muscle cells was investigated using the Ca(2+) indicator, fura-2. 2. The ATP-induced [Ca(2+)](i) increase resulted from both Ca(2+) release and Ca(2+) influx. The former was stimulated by phospholipase C activation, while the latter occurred predominantly via the receptor-operated Ca(2+) channels (ROC), rather than the store-operated Ca(2+) channels (SOC) or the voltage-operated Ca(2+) channel (VOC). Furthermore, the

Unveiling the molecular crosstalk between the purinergic P2X7 receptor and cannabinoid subtype 2 receptor in human microglial HMC3 cells: New insights for multitarget therapies in neuroinflammation.

BACKGROUND: Microglia-mediated neuroinflammation plays a pivotal role in the progression and possible onset of numerous neurodegenerative diseases. A promising therapeutic approach involves shifting microglial polarization from a pro-inflammatory to an anti-inflammatory phenotype. Two receptors central to this process are the purinergic P2X7 receptor, an ATP-gated ion channel that promotes pro-inflammatory signaling, and the cannabinoid receptor subtype 2, a Gi/o protein-coupled receptor associated with anti-inflammatory effects. METHODS: Here, we investigated the therapeutic potential of pha

Human Dorsal Root Ganglia Neuronal Cell Line to Study Nociceptive Signaling: A New Pipeline for Pain Therapy.

Nociceptive afferent neurons within the dorsal root ganglion (DRG) detect and relay painful peripheral stimuli, and the malfunctioning of this process leads to sustained pain states. Animal model studies have been invaluable for demonstrating the importance of the DRG nociceptor in pain sensation and the development of related analgesic targets. However, a human in vitro model of nociception is essential to confirming the relevance of preclinical findings for therapeutic drug development. We characterized the nociceptive properties of differentiated cells from the human DRG-derived immortalize

RIPK1-driven calcium overload and lysosomal-mitochondrial dysfunction induce testicular necroptosis following DEHP exposure.

Di-(2-ethylhexyl) phthalate (DEHP) is a plasticizer widely used to enhance the flexibility and durability of plastic products. As an environmental endocrine disruptor, DEHP impairs male reproductive function. Its metabolite, mono-(2-ethylhexyl) phthalate (MEHP), mediates many toxic effects, but the mechanisms remain unclear. We hypothesized that DEHP induces testicular necroptosis through MEHP-mediated calcium overload and the RIPK1-regulated lysosomal-mitochondrial axis. Here, we reveal this novel mechanism. This study investigated DEHP-induced testicular damage, focusing on necroptosis and c

Nitrogen gas-derived cold atmospheric plasma suppresses pruritus through modulation of peripheral sensory neurons and keratinocytes.

Chronic pruritus is a highly prevalent symptom that markedly impairs the quality of life in both humans and animals. Although pharmacological therapies, including Janus kinase (JAK) inhibitors, are widely used for itch management, long-term treatment is associated with adverse effects and increased patient burden. Therefore, the development of non-pharmacological antipruritic approaches is highly desired. In this study, we investigated the antipruritic effects of nitrogen gas-derived cold atmospheric plasma (N2-CAP) and explored its underlying mechanisms using behavioral and calcium imaging an

Palmatine enhances osteogenesis in hADMSCs via antioxidant enzyme upregulation and early RUNX2 activation.

Palmatine, a protoberberine alkaloid with recognized anti-inflammatory and anti-resorptive activities, has recently attracted interest as a potential modulator of bone homeostasis; however, its direct osteo-inductive effects on human adipose-derived mesenchymal stem cells (hADMSCs) remain undefined. Given the accessibility, proliferative efficiency, and clinical relevance of hADMSCs, identifying natural small molecules capable of enhancing their osteogenic differentiation is of considerable therapeutic significance. This study investigated the osteogenic potential of Palmatine using a multi-le

Combined treatment targeting Ca2+ store mediated Ca2+ release and store-operated calcium entry reduces secondary axonal degeneration and improves functional outcome after SCI.

Store-operated calcium entry (SOCE) is crucial for cellular processes, including cellular calcium homeostasis and signaling. However, uncontrolled activation of SOCE is implicated in neurological disorders and CNS trauma, but underlying mechanisms remain unclear. We hypothesized that inhibiting SOCE enhances neurological recovery following contusive spinal cord injury (SCI). To investigate key SOCE effectors, stromal interaction molecules (STIM) and Orai channels on neurological recovery following spinal cord injury (SCI), we utilized male and female conditional neuronal Stim1KO mice to invest

Human iPSC-derived cerebral organoids reveal oxytocin-mediated protection against amyloid-β pathology.

INTRODUCTION: Neuroinflammation is a key contributor to the pathogenesis of Alzheimer's disease (AD), and impaired clearance of amyloid-β (Aβ) by microglia is closely associated with disease progression. Oxytocin (OXT), a hypothalamic neuropeptide, has recently been reported to exert anti-inflammatory effects on microglia; however, its therapeutic potential in the human brain remains unclear. METHODS: We generated human cerebral organoids (hCOs) from induced pluripotent stem cells (iPSCs) to model early AD-like pathology. Aβ toxicity was induced by applying 3 μM Aβ1-42 for 48 h. The protectiv

Three mammalian VDAC isoforms distinctly regulate mitochondrial function and proteome to maintain cell metabolism.

The voltage-dependent anion channel (VDAC) is the most ubiquitous protein in the mitochondrial outer membrane. This channel facilitates the flux of water-soluble metabolites and ions like calcium across the mitochondrial outer membrane. Beyond this canonical role, VDAC has been implicated, through interactions with protein partners, in several cellular processes such as apoptosis, calcium signaling, and lipid metabolism. There are three VDAC isoforms in mammalian cells, VDAC1, VDAC2, and VDAC3, with varying tissue-specific expression profiles. From a biophysical standpoint, all three isoforms …
