Variability in Genomic and Virulent Properties of Porphyromonas gingivalis Strains Isolated From Healthy and Severe Chronic Periodontitis Individuals.

Porphyromonas gingivalis has been extensively associated with both the onset and progression of periodontitis. We previously isolated and characterized two P. gingivalis strains, one from a patient exhibiting severe chronic periodontitis (CP3) and another from a periodontally healthy individual (H3). We previously showed that CP3 and H3 exhibit differences in virulence since H3 showed a lower resistance to cationic peptides compared with CP3, and a lower ability to induce proliferation in gingival epithelial cells. Here, we aimed to determine whether differences in virulence between these two

Re-focusing on Agnathia-Otocephaly complex.

OBJECTIVES: Agnathia-otocephaly complex is a rare condition characterized by mandibular hypoplasia or agnathia, ear anomalies (melotia/synotia) and microstomia with aglossia. This severe anomaly of the first branchial arch is most often lethal. The estimated incidence is less than 1 in 70.000 births, with etiologies linked to both genetic and teratogenic factors. Most of the cases are sporadic. To date, two genes have been described in humans to be involved in this condition: OTX2 and PRRX1. Nevertheless, the overall proportion of mutated cases is unknown and a significant number of patients r

Pancreatic dysfunction causally impacts region-specific cortical thinning: a mendelian randomization study revealing pancreas-brain axis.

BACKGROUND: Alterations in cerebral cortex structure among patients with pancreatic diseases have been reported, yet the underlying causes remain unclear. This study employs Mendelian randomization (MR) to elucidate the causal effects of four pancreatic diseases (benign and malignant pancreatic cancer, acute and chronic pancreatitis) on cerebral cortex structure. METHODS: This study utilized genome-wide association study (GWAS) summary data from 437,418 participants in the Finnish consortium to identify the predictive role of genetic factors in acute pancreatitis. Similarly, data from these 4

Exploring risk factors for autoimmune diseases complicated by non-hodgkin lymphoma through regulatory T cell immune-related traits: a Mendelian randomization study.

BACKGROUND: The effect of immune cells on autoimmune diseases (ADs) complicated by non-Hodgkin lymphoma (NHL) has been widely recognized, but a causal relationship between regulatory T cell (Treg) immune traits and ADs complicated by NHL remains debated. METHODS: Aggregate data for 84 Treg-related immune traits were downloaded from the Genome-Wide Association Study (GWAS) catalog, and GWAS data for diffuse large B-cell lymphoma (DLBCL; n=315243), follicular lymphoma (FL; n=325831), sjögren's syndrome (SS; n=402090), rheumatoid arthritis (RA; n=276465), dermatopolymyositis (DM; n=311640), psor

High rates of congenital airway anomalies in neonates with single ventricle congenital heart disease.

Neonates with single ventricle congenital heart disease (SV CHD) may have noncardiac comorbidities, including congenital airway anomalies (CAA). Using a national administrative database, we analyzed neonatal admissions with ICD-10 codes for SV CHD with and without CAA from October 2020 to December 2024, excluding admissions with genetic syndromes to reduce confounding by the potential contribution of syndromes to CAA. Among 5144 neonatal admissions with SV CHD, 535 (10.4%) had CAA, compared to 23,837 (0.4%) of 5,942,621 neonatal admissions without CHD (odds ratio [OR] [95% confidence interval]

Biallelic Variants in MIMS1 Produce a Form of Spondyloepimetaphyseal Dysplasia With Tracheal Stenosis and Ectodermal Dysplasia (SEMDTSED).

Through the GeneMatcher data-sharing database, we identified five affected individuals from four unrelated families with a skeletal dysplasia who have biallelic, putatively deleterious variants in MIMS1 (formerly FAM210A). The common features included chondrodysplasia with short stature, tracheal stenosis, conical teeth and/or early tooth decay, and sparse hair suggestive of ectodermal dysplasia as a component of the phenotype. The radiographic features included platyspondyly, epiphyseal dysplasia of the long bones, short phalanges with delayed carpal ossification, lacy iliac crests, and calca

Famine, plague, and slaughters: The fleeing famine hypothesis of anorexia nervosa reconsidered.

The fleeing famine hypothesis (FFH) posits that a genetically and evolutionarily based predisposition for food restriction and hyperactivity is activated among susceptible humans and nonhumans upon experiencing a prolonged energy deficit. Such circumstances manifest in the counterintuitive responses to starvation that are characteristic of anorexia nervosa: continued dietary restriction, denial of declining physical state, and, in at least a subset of those with anorexia nervosa, increased motor activity. We consider the nine predictions derived by Guisinger (2003) in her original formulation

Identification and pathological significance of variants in the promoter region of ACTC1 gene in congenital ventricular septal defect.

Ventricular septal defect (VSD) is one of the most common congenital heart diseases, but the role of variants at the ACTC1 gene promoter region in VSD is unclear. We investigated variants in the promoter region of ACTC1 gene in 627 subjects (309 sporadic VSD patients and 318 healthy controls) by Sanger sequencing and 6 variants including 1 novel heterozygous variant [g.5163 T > A] were identified. Of the 6 variants, 3 were found only in VSD patients. In mouse cardiomyocytes (HL-1), the transcriptional activity of the ACTC1 gene promoter was significantly changed by the variants (p < 0.05). Progress in the Study of Animal Models of Metabolic Dysfunction-Associated Steatotic Liver Disease.

Metabolic dysfunction-associated steatotic liver disease (MASLD) has recently been proposed as an alternative term to NAFLD. MASLD is a globally recognized chronic liver disease that poses significant health concerns and is frequently associated with obesity, insulin resistance, and hyperlipidemia. To better understand its pathogenesis and to develop effective treatments, it is essential to establish suitable animal models. Therefore, attempts have been made to establish modelling approaches that are highly similar to human diet, physiology, and pathology to better replicate disease progressio

Cluster analysis and visualisation of electronic health records data to identify undiagnosed patients with rare genetic diseases.

Rare genetic diseases affect 5-8% of the population but are often undiagnosed or misdiagnosed. Electronic health records (EHR) contain large amounts of data, which provide opportunities for analysing and mining. Data mining, in the form of cluster analysis and visualisation, was performed on a database containing deidentified health records of 1.28 million patients across 3 major hospitals in Singapore, in a bid to improve the diagnostic process for patients who are living with an undiagnosed rare disease, specifically focusing on Fabry Disease and Familial Hypercholesterolaemia (FH). On a bas

Molecular landscape and clinical outcome of SRSF2/TET2 Co-mutated myeloid neoplasms.

The mutations in SRSF2 and TET2 genes are frequently present in various myeloid neoplasms. The potential impact of SRSF2/TET2 co-mutations on patient survival is incompletely understood. We identified 412 patients with SRSF2/TET2 co-mutations from our NextGen sequencing database of around 8000 patients and reported likely the largest cohort study. Our study demonstrated the presence of these co-mutations in a spectrum of myeloid neoplasms, which show different genetic and molecular characteristics. Most of the patients with these co-mutations had normal karyotype. Interestingly, our study prov

Nutrition Strategy and Life Style in Polycystic Ovary Syndrome-Narrative Review.

Here we present an extensive narrative review of the broadly understood modifications to the lifestyles of women with polycystic ovary syndrome (PCOS). The PubMed database was analyzed, combining PCOS entries with causes, diseases, diet supplementation, lifestyle, physical activity, and use of herbs. The metabolic pathways leading to disturbances in lipid, carbohydrate, and hormonal metabolism in targeted patients are described. The article refers to sleep disorders, changes in mental health parameters, and causes of oxidative stress and inflammation. These conditions consistently lead to the

Detection of Babesia odocoilei in Ixodes scapularis Ticks Collected in Southern Ontario, Canada.

Tick-borne zoonotic diseases have an economic and societal impact on the well-being of people worldwide. In the present study, a high frequency of Babesia odocoilei, a red blood cell parasite, was observed in the Huronia area of Ontario, Canada. Notably, 71% (15/21) blacklegged ticks, Ixodes scapularis, collected from canine and feline hosts were infected with B. odocoilei. Consistent with U.S. studies, 12.5% (4/32) of questing I. scapularis adults collected by flagging in various parts of southwestern Ontario were positive for B. odocoilei. Our data show that all B. odocoilei strains in the p

Phexpo: a package for bidirectional enrichment analysis of phenotypes and chemicals.

Phenotypes are the result of the complex interplay between environmental and genetic factors. To better understand the interactions between chemical compounds and human phenotypes, and further exposome research we have developed "phexpo," a tool to perform and explore bidirectional chemical and phenotype interactions using enrichment analyses. Phexpo utilizes gene annotations from 2 curated public repositories, the Comparative Toxicogenomics Database and the Human Phenotype Ontology. We have applied phexpo in 3 case studies linking: (1) individual chemicals (a drug, warfarin, and an industrial

In patients with a positive family history of familial adenomatous polyposis can the condition be diagnosed from the presence of congenital hypertrophy of the retinal pigment epithelium detected via an eye examination: A systematic review.

In classic familial adenomatous polyposis (FAP) adenomas become malignant. Congenital hypertrophy of the retinal pigment epithelium (CHRPE) is a retinal pigmented lesion and is the earliest and most common potential extraintestinal manifestation of FAP. This review aims to summarize and analyse all of the published data on CHRPE in patients with classic FAP and then ascertain whether these patients should undergo a relatively cheap and non-invasive dilated fundus examination to screen for CHRPE. Adhering to Preferred Reporting Items for Systematic Reviews and Meta Analyses guidelines our datab
