Isolated cleft lip and/or palate (CL/CLP) is a complex congenital anomaly with many contributing factors. There are several genes involved in the aetiology of CL/CLP, they are different in selected populations. In a previous study, the mitochondrial haplotypes of Latvian subjects with CL/CLP were characterized. Latvian subjects with CL/CLP have mostly mitochondrial haplogroups U4/U5 compared with the ethnic population of Latvia. The aim of this study was to stratify the results of genotyping based on European mitochondrial DNA (mtDNA) haplotypes. DNA samples from 108 patients with CL/CLP and f

BACKGROUND: Rett syndrome is an uncommon neurodevelopmental disorder with an incidence of 1:9,000 live female births. The principal genetic cause was first reported in 1999 when the association with mutations in the methyl-CpG-binding protein 2 (or MECP2) gene was identified. This study uses data from a large international database, InterRett, to examine genotype-phenotype relationships and compares these with previous findings in a population-based cohort.
METHOD: The data set for these analyses was derived from a subset of InterRett cases with subject information collected from the family,

The study of protein-protein interactions is essential to define the molecular networks that contribute to maintain homeostasis of an organism's body functions. Disruptions in protein interaction networks have been shown to result in diseases in both humans and animals. Monogenic diseases disrupting biochemical pathways such as hereditary coagulopathies (e.g. hemophilia), provided a deep insight in the biochemical pathways of acquired coagulopathies of complex diseases. Indeed, a variety of complex liver diseases can lead to decreased synthesis of the same set of coagulation factors as in hemo

BACKGROUND: Rett syndrome is a neurodevelopmental disorder mostly affecting females and caused by mutations in the MECP2 gene. Originally the syndrome was characterised as having a normal prenatal and perinatal period with later regression. Previous work has speculated that the girl with Rett syndrome may not be normal at birth.
AIMS: to examine whether early development between birth and ten months varies by genotype in Rett syndrome.
METHODS: cases were sourced from two databases, the Australian Rett Syndrome Database (est. 1993) and the newly formed InterRett - IRSA Rett Phenotype Databas

Osteoarthritis (OA) is a degenerative disease affecting all joints in the body, impacting over 500 million people globally and imposing a significant healthcare burden. Tris(1-chloro-2-propyl) phosphate (TCPP), one of the widely used organophosphate flame retardants (OPFRs), exhibits notable environmental and biological toxicity. However, the role and molecular mechanisms of TCPP exposure in OA remain unclear. This study combined network toxicology, machine learning, and molecular docking techniques to identify key targets and pathways associated with TCPP exposure and OA. A nomogram model was

BACKGROUND: Parkinson's disease (PD) is the second most common neurodegenerative disorder worldwide, and no curative treatment is currently available. Although inflammation has been increasingly implicated in PD, the causal roles of circulating inflammatory cytokines remain largely unclear, making them potential therapeutic targets. Traditional Chinese Medicine (TCM), characterized by systemic and multi-target regulation, may offer complementary strategies for PD.
PURPOSE: This study aimed to investigate the therapeutic potential of Cistanche in PD by modulating circulating inflammatory cytok

Network pharmacology has become a central paradigm in modern drug discovery, replacing the reductionist "one drug, one target" view with a systems-level understanding of how compounds engage networks of proteins that are linked to disease. Despite its impact, a typical network-pharmacology study remains fragmented and technically demanding: researchers must query several independent databases, install and reconcile multiple standalone tools for target collection, network construction, hub-gene ranking and pathway enrichment, and then manually bridge the results into structure-based follow-up s

MOTIVATION: Proteomics has experienced substantial growth in methods and data analysis approaches, with the development of new data-dependent (DDA) and data-independent acquisition (DIA) workflow and several search engine algorithms and software packages. Each of these workflows has its unique data analysis package that performs data reduction, missing value imputation, statistical testing, and visualization. Often, these tools are designed for expert users.
RESULTS: We present Quantitative Proteomics Made Simple (QProMS), a user-friendly, search engine-agnostic data analysis and visualizatio

BACKGROUND AND OBJECTIVE: Kirsten rat sarcoma viral oncogene homolog (KRAS)-G12D is a common oncogenic mutation in non-small cell lung cancer (NSCLC) and has posed significant challenges in the development of treatment approaches that specifically target this alteration. KRAS-G12D differs from KRAS-G12C in its biochemical and signal transduction features, which limits the use of drugs targeting the KRAS-G12C mutation, creating substantial demands in clinical contexts. This review sought to summarize the major developments in treatment for KRAS-G12D-mutant NSCLC, with a focus on the recent key

Pancreatic cancer is a highly lethal digestive malignancy characterized by insidious onset and rapid progression. Approximately 80% of patients are diagnosed at an advanced stage with metastasis, missing the opportunity for radical surgery, and chemotherapy remains the main palliative treatment. Its incidence is rising annually, highlighting the urgent need for novel therapeutic strategies. MLN4924 (Pevonedistat), a first-in-class NEDD8-activating enzyme inhibitor, exhibits promising antitumor activity in various cancers, yet its role and mechanism in pancreatic cancer remain largely undefined
