Effect of chondroitin sulfate on intraocular pressure in rats.

PURPOSE: To study the effect of intracameral injections of chondroitin sulfate (CS) on intraocular pressure (IOP), retinal function, and histology in rats. METHODS: Acute or chronic injections of CS were performed unilaterally in the rat anterior chamber, whereas the contralateral eye was injected with vehicle. IOP was daily or weekly assessed by a tonometer. Retinal function was assessed by scotopic electroretinography (ERG) and the visual pathway by flash visual evoked potentials (VEPs), whereas the retinal and optic nerve head structure were examined by histologic analysis. RESULTS: A sin

Retinal ganglion cells downregulate gene expression and lose their axons within the optic nerve head in a mouse glaucoma model.

Little is known about molecular changes occurring within retinal ganglion cells (RGCs) before their death in glaucoma. Taking advantage of the fact that gamma-synuclein (Sncg) mRNA is expressed specifically and highly in adult mouse RGCs, we show in the DBA/2J mouse model of glaucoma that there is not only a loss of cells expressing this gene, but also a downregulation of gene expression of Sncg and many other genes within large numbers of RGCs. This downregulation of gene expression within RGCs occurs together with reductions in FluoroGold (FG) retrograde transport. Surprisingly, there are al

Hypoxia-inducible factor 1alpha in the glaucomatous retina and optic nerve head.

OBJECTIVE: To examine tissue hypoxia in the retina and optic nerve head of glaucomatous eyes by the assessment of a transcription factor, hypoxia-inducible factor 1alpha (HIF-1alpha), which is tightly regulated by the cellular oxygen concentration. METHODS: Using immunohistochemical analysis, the cellular localization of HIF-1alpha was studied in the retina and optic nerve head of 28 human donor eyes with glaucoma compared with 20 control eyes from healthy donors matched for several characteristics. The relationship between the retinal regions that exhibited immunostaining for HIF-1alpha and

Perfusion of the optic nerve head and visual field damage in glaucomatous patients.

PURPOSE: To examine the relation between perfusion of the optic nerve head and visual field defects in glaucomatous patients. METHODS: A study was performed on 94 patients affected with primary open-angle glaucoma. The optic nerve head blood flow was measured by means of a scanning laser Doppler flowmeter (Heidelberg Retina Flowmeter). Blood volume, flow and velocity were analysed in two areas of the rim and in one area of the lamina cribrosa. The visual field was tested with the 30-2 full threshold white-on-white program and mean deviation (MD) and corrected pattern standard deviation (CPSD)

Two cases of primary open angle glaucoma with serum autoantibody against retinal ganglion cells.

PURPOSE: To present two cases of primary open angle glaucoma (POAG) with serum autoantibody against retinal 25 kDa or 50 kDa antigen. METHODS: Ocular examinations were performed on a 60-year-old man (case 1) and a 74-year-old woman (case 2). We also performed serological and immunological examinations including Western blot analysis and immunocytochemical analysis. RESULTS: In Western blot analysis, they were found to have serum autoantibody against retinal 25 kDa or 50 kDa antigen, and immunocytochemical analysis revealed that retinal ganglion cell layers were specifically labeled by each p

Increased intraocular pressure and visual field defects in high resistance wind instrument players.

OBJECTIVE: In this twofold study, part 1 aimed to determine whether the playing of high resistance wind instruments elevates intraocular pressure (IOP) and if so, to investigate the mechanism of IOP elevation and whether its magnitude differs while playing high resistance versus low resistance instruments. The purpose of part 2 was to evaluate whether high resistance players have a greater incidence of glaucomatous changes than other musicians. DESIGN: Three case reports and a cross-sectional study. PARTICIPANTS: Two players of high resistance instruments and one player of high and low resis

Optic disc hemorrhages in a population with and without signs of glaucoma.

OBJECTIVE: This study aimed to determine the prevalence and associations of optic disc hemorrhage in a well-defined older Australian population. DESIGN: The study design was a population-based, cross-sectional study. PARTICIPANTS: A total of 3654 persons 49 years of age or older, representing 88% of permanent residents from an area west of Sydney, participated in the study. MAIN OUTCOME MEASURES: Participants underwent a detailed eye examination. The diagnosis of optic disc hemorrhage was made from masked photographic grading; disc hemorrhages were subclassified as flame or blot in shape. O

Aggregate Effects of Intraocular Pressure and Cup-to-Disc Ratio Genetic Variants on Glaucoma in a Multiethnic Asian Population.

PURPOSE: High intraocular pressure (IOP) and large vertical cup-to-disc ratio (VCDR) are important risk factors of glaucoma. Recent genome-wide association studies have discovered several genetic variants associated with IOP and VCDR. In this study, we examined the aggregate effects of these IOP-, VCDR-associated variants on glaucoma. DESIGN: Case-control genetic association study. PARTICIPANTS: A total of 6881 participants, including 194 glaucoma and 158 primary open-angle glaucoma (POAG) cases in the Singapore Epidemiology of Eye Diseases Study. METHODS: We first identified IOP and VCDR r

Von Hippel-Lindau gene deletion and expression of hypoxia-inducible factor and ubiquitin in optic nerve hemangioma.

PURPOSE: Von Hippel-Lindau (VHL) disease is a hereditary cancer syndrome expressed in multiple organs caused by germline alterations of the VHL gene. We have shown VHL deletion in the "stromal" cells of retinal angiomas. The VHL protein-associated complex is a primary ubiquitin ligase for the ubiquitination of hypoxia-inducible factor (HIF). This study examines VHL and ubiquitin expression in optic nerve hemangiomas and juxtapapillary angiomas. METHODS: Using microdissection and polymerase chain reaction, four optic nerve hemangiomas (one also had juxtapapillary angioma) associated with VHL d

Posterior Pole Vascular Changes Before Treatment of Retinopathy of Prematurity.

IMPORTANCE:: Some proponents of telemedicine screening for retinopathy of prematurity (ROP) believe a screening system based on posterior pole vessel characteristics is sufficient for identifying infants needing treatment. OBJECTIVE:: To evaluate the vascular characteristics present when it was determined treatment was needed, and the number of days between examinations from when the vessels of both eyes were last noted to be normal until a decision was made to treat. DESIGN:: Retrospective study; June 2013-May 2017 SETTING:: Neonatal intensive care unit; academic medical center PARTICIPAN

Alterations to vascular endothelium in the optic nerve head in patients with vascular comorbidities.

Vascular comorbidities are inherently linked to the pathogenesis of central retinal artery occlusion (CRAO) and central retinal vein occlusion (CRVO). However, the endothelial-mediated pathogenic mechanisms that precede, and therefore modulate, luminal occlusion have not been clarified. The aim of this study was to delineate the pattern of endothelial morphometric alteration in the central retinal artery and vein in patients with vascular comorbidities. Eyes with a previous history of vascular occlusion were not included in this study in order to avoid the confounding effects of post-occlusion
