Triple-helical ligands for collagen-binding proteins improve cartilage extracellular matrix production in nasal chondrocytes.

The use of articular chondrocytes for cartilage repair is constrained by donor-site morbidity, poor proliferation, and difficulties to maintain a stable phenotype. As a result, nasal chondrocytes (NCs) have recently emerged as an alternative cell source. To better establish the potential of NCs in tissue engineering, we have explored their response to the activation of collagen-binding proteins (integrins, discoidin domain receptors (DDRs) andthe secreted protein acidic cysteine-rich (SPARC)) which are essential to cartilage homeostasis. Ligands for these proteins were synthesized as triple-he…

Susceptibility to quantum dot induced lung inflammation differs widely among the Collaborative Cross founder mouse strains.

Quantum dots (QDs) are engineered semiconductor nanoparticles with unique physicochemical properties that make them potentially useful in clinical, research and industrial settings. However, a growing body of evidence indicates that like other engineered nanomaterials, QDs have the potential to be respiratory hazards, especially in the context of the manufacture of QDs and products containing them, as well as exposures to consumers using these products. The overall goal of this study was to investigate the role of mouse strain in determining susceptibility to QD-induced pulmonary inflammation

Cells as vehicles for therapeutic genes to treat liver diseases.

Gene therapy involves the transfer of genetic sequences to tissues to obtain a curative effect. Effective gene transfer can be achieved by introducing the therapeutic gene into virus-like particles that facilitate the penetration of the transgene into the cells. However, direct injection of viral vectors may activate innate immunity leading to toxic effects. On the other hand, viral vectors frequently induce neutralizing antibodies, which limit the efficacy of repeated vector administration. Moreover, targeting of the transgene to the desired tissue is a goal that not always can be attained wi

Healing potential of transplanted allogeneic chondrocytes of three different sources in lesions of the avascular zone of the meniscus: a pilot study.

UNLABELLED: Successful treatment of tears to the avascular region of the meniscus remains a challenge. Current repair techniques, such as sutures and anchors, are effective in stabilizing the peripheral, vascularized regions of the meniscus, but are not adequate for promoting healing in the avascular region. The purpose of this study was to demonstrate the healing ability of a tissue-engineered repair technique using allogenic chondrocytes from three different sources for the avascular zone of the meniscus. MATERIAL AND METHODS: Articular, auricular, and costal chondrocytes were harvested fro

CD40-CD40L interactions in systemic autoimmune diseases.

Systemic autoimmune diseases arise from a heterogeneous interplay of genetic, epigenetic, and environmental factors that culminates in loss of tolerance, chronic inflammation, and tissue injury. Among the pathways linking susceptibility to effector disease, CD40-CD40L is a central regulator of antigen presentation, T-B cell collaboration, and germinal centre responses, while also contributing to extrafollicular autoantibody production. In this review, we first introduce the disease-specific rationale for targeting CD40-CD40L interactions, with a particular focus on systemic lupus erythematosus

Emerging Strategies for Bioprinting Organized Musculoskeletal Tissues with Biomimetic Collagen Architecture.

Collagen fiber organization is critical to the biomechanical function of musculoskeletal tissues. This has motivated increased interest in tissue engineering strategies aiming to recapitulate native collagen architecture in order to generate more biomimetic grafts. In particular, three-dimensional (3D) bioprinting has emerged as a powerful biofabrication platform capable of generating patient-specific constructs containing cells, supporting biomaterials, and diverse regulatory factors (e.g., genes, proteins, and small molecules). This paper will review how emerging bioprinting strategies can a…

Nonuniform Curvature and Bending Rigidity of Adsorbed Collagen Molecules in Aqueous Solution.

Collagen, the most abundant protein in mammals, plays a key role in tissue formation and mechanics due to its triple-helix structure. We used atomic force microscopy to study individual type-I and type-III human collagen molecules adsorbed on smooth mica surfaces from low-salt, near-neutral aqueous solutions. Statistical analysis of their two-dimensional contours revealed nonuniform curvature in both collagen types, which persisted after surface drying and molecular dehydration, owing to robust collagen-mica adsorption. In addition, the angle between tangent vectors at the ends of molecular se

Plant-Derived Nanovesicles as a Cross-Kingdom Therapeutic Approach for T2DM and Related Metabolic Disorders: Current Evidence and Future Perspectives.

Type 2 diabetes mellitus (T2DM) is increasingly viewed as a systemic metabolic disorder rather than as an isolated glycemic defect. It is closely linked to obesity and metabolic dysfunction-associated steatotic liver disease (MASLD), sharing mechanisms such as insulin resistance, chronic inflammation, and interorgan crosstalk among the gut, liver, adipose tissue, pancreas, and central nervous system. Plant-derived nanovesicles (PDNVs) are emerging cross-kingdom nanoplatforms with the potential to modulate metabolic and inflammatory signaling. Evidence from T2DM, obesity, and MASLD models sugge

Nanostarch-Stimulated Cell Adhesion in 3D Bioprinted Hydrogel Scaffolds for Cell Cultured Meat.

Three-dimensional (3D) bioprinting has great potential in the applications of tissue engineering, including cell culturing meat, because of its versatility and bioimitability. However, existing bio-inks used as edible scaffold materials lack high biocompatibility and mechanical strength to enable cell growth inside. Here, we added starch nanoparticles (SNPs) in a gelatin/sodium alginate (Gel/SA) hydrogel to enhance printing and supporting properties and created a microenvironment for adherent proliferation of piscine satellite cells (PSCs). We demonstrated the biocompatibility of SNPs for cell…
