Nanostructured Polyurethane-Collagen Hydrogels: Bioactive Crosslinked Networks for Enhanced Wound Healing.

Nanostructured polyurethanes (nPUs) are promising materials for biomedical applications due to their mechanical strength, controlled degradation, and bioactivity. In this study, collagen-based hydrogels were developed using nPUs synthesized from ethoxylated glycerol and either hexamethylene diisocyanate (HDI) or isophorone diisocyanate (IPDI), functionalized with L-tyrosine (T). These nPUs were incorporated at 15% and 30% by weight into porcine dermis collagen. The HDI-based nPUs (HDI-T), with particle sizes between 6 and 58 nm, achieved high crosslinking densities (> 90%) and superabsorbent c

Functionalized peptide hydrogel to generate human insulin-producing cells in vitro.

Type I diabetes is a chronic disease that affects people worldwide. When insulin administration is no longer effective, transplantation of pancreatic islets represents an alternative for diabetics. However, islet grafting carries limitations, which include poor availability of donors, surgery risks, and lifelong immunosuppressive therapy. To address this, novel approaches, such as the use of soft hydrogels as vehicles of cells are being developed for tissue grafting applications. Self-assembling peptide hydrogels (SAPHs) are biocompatible and versatile materials widely used for both, three-dim

An early rapamycin-releasing nerve wrap with dual function for nerve regeneration and adhesion prevention.

Limited ability of nerve regeneration and the formation of nerve adhesions result to poor outcomes of peripheral nerve injury (PNI) following surgical nerve repair. Currently, there is no efficient therapy for the improvement of functional recovery after surgical repair. Rapamycin (Rapa) has been found useful for promoting nerve regeneration through removing cellular debris at the early stages after PNI. However, its lower oral bioavailability and significant adverse effects limit its clinical application. In this study, we developed a biocompatible nerve wrap based on biocompatible monomethox

Bioactive glass-induced B cell depletion remodels the osteoimmunological microenvironment to enhance osteogenesis.

B cells are critical in bone homeostasis, and their dysfunction is linked to various bone disorders. Bioactive glasses (BGs) are known for their immunomodulatory properties and are increasingly utilized in bone regeneration. However, the specific effects of BGs on B cells and their subsequent impact on osteogenesis remain unclear. In this study, we investigated the immunomodulatory effects of three BGs (45S5-BG, 13-93-BG, and B3-Cu-Zn-BG) on B cells. 13-93-BG had minimal effect on the survival of activated B cells, while 45S5-BG induced a noticeable cytotoxic effect on B cells. Notably, B3-Cu-

PURPOSE: Posterior capsule opacification (PCO) is the most common postoperative complication after cataract surgery. As a pharmacological prophylaxis, we designed a drug-carrying slow-release intraocular lens (IOL) implant coated with polycaprolactone and methotrexate and characterized the release kinetics and the in-vitro effect. METHODS: Electron microscopy was used to image the coated IOL and a pharmacodynamic eye model to determine the release kinetics. FHL 124 cells were exposed to the coated IOL for 48 h and the effect was determined by migration (Boyden chamber, scratch assay), prolife

Biocompatible sinapic acid-loaded chitosan nanoparticles as a topical nanotherapy for uveitis in rats: Formulation, characterization, and anti-inflammatory efficacy.

Sinapic acid (SA), a naturally occurring polyphenol, exhibits diverse biological properties, including anti-inflammatory activity. Its potential in treating ocular inflammations remains underexplored. Ocular protective barriers and hydrophobicity of SA hinder its effective ocular delivery. To overcome these limitations, SA-loaded chitosan nanoparticles (SA-CSNPs) were developed. Characterization revealed nano-sized (234.7 nm), positively charged (+ 32.4 mV) SA-CSNPs with a narrow polydispersity index (0.304). XRD and FTIR analyses confirmed the amorphous state of encapsulated SA without advers

Antibacterial Electrophoretically Loaded Titania Nanotubes on Titanium Alloy Implants Enhance Osseointegration.

Primary hip and knee arthroplasties are common surgeries in the U.S., with periprosthetic joint infection (PJI) being the leading cause of implant revision. Systemic antibiotics often fail to achieve sufficient local concentrations, driving interest in localized drug delivery. Titanium (Ti) implants modified with titania nanotubes (TNTs) provide an increased surface area for drug loading and controlled release. Previous studies have shown that gentamicin-loaded TNTs inhibit Staphylococcus aureus growth in vitro without compromising osteoblast viability. This study investigated the effect of ge

Triple-crosslinked double-network alginate/dextran/dendrimer hydrogel with tunable mechanical and adhesive properties: A potential candidate for sutureless keratoplasty.

An ideal adhesive hydrogel must possess high adhesion to the native tissue, biocompatibility, eligible biodegradability, and good mechanical compliance with the substrate tissues. We constructed an interpenetrating double-network hydrogel containing polysaccharides (alginate and dextran) and nanosized spherical dendrimer by both physical and chemical crosslinking, thus endowing the hydrogel with a broad range of mechanical properties, adhesive properties, and biological functions. The double-network hydrogel has moderate pore sizes and swelling properties. The chelation of calcium ions signifi

Single-cell analysis of peri-implant gingival tissue to assess implant biocompatibility and immune response.

PURPOSE: The innate immune response, particularly the reaction of polymorphonuclear neutrophils (PMNs), is crucial in shaping the outcomes of chronic inflammation, fibrosis, or osseointegration following biomaterial implantation. Peri-implantitis or peri-implant mucositis, inflammatory conditions linked to dental implants, pose a significant threat to implant success. We developed a single-cell analysis approach using a murine model to assess the immune response to implant materials, offering a practical screening tool for potential dental implants. METHODS: We performed bioinformatics analys

Development and in vitro assessment of injectable, adhesive, and self-healing chitosan-based hydrogels for treatment of spinal cord injury.

Injured spinal cords have a limited ability to regenerate because of the inhibitory environment formed in situ that affects neuronal regrow. Ensuring stable contact between the injuried nerves to support neural regeneration in the lesion microenvironment remains a significant challenge. To address this challenge, we have engineered a new injectable and adhesive hydrogel to treat spinal cord injuries. This hydrogel was produced by functionalizing chitosan with catechol groups and crosslinking it with different amounts of β-glycerophosphate to obtain adhesive hydrogels with tunable mechanical pr

A crayfish chitosan-based bioactive film to treat vaginal infections: A sustainable approach.

Polymicrobial communities are seen to be a sign of health, but they can turn detrimental when an excess of pathogenic species leads to recurring vaginal infections. This microbiological imbalance may decrease women's fertility, increasing also the risk of infection by Human Papillomavirus (HPV) and/or other sexually transmitted infections (STIs). There is a worldwide need for smart/sustainable solutions to tackle these types of infections. Hereupon, we investigated, as a potential solution, the use of crayfish chitosan-based membrane as a mucoadhesive, antimicrobial, biocompatible and biodegra

Matrix Stiffness of GelMA Hydrogels Regulates Lymphatic Endothelial Cells toward Enhanced Lymphangiogenesis.

Lymphatic vessel regeneration is crucial for various tissue engineering strategies, particularly in resolving inflammation and restoring tissue homeostasis. In our study, we focused on investigating how hydrogel matrix stiffness influences lymphatic endothelial cells (LECs) in promoting lymphatic vessel regeneration. Gelatin methacrylate (GelMA) was chosen as our biomaterial due to its versatility in tissue engineering and biofabrication. We fabricated GelMA hydrogels at concentrations of 5, 7.5, and 15% (w/v) with corresponding Young's modulus values of 1.55 kPa (soft matrix), 12.02 kPa (medi

Latex-Bridged Inverse Pickering Emulsion for Durable Superhydrophobic Coatings with Dual Antibacterial Activity.

There is agreement that every colloidal structure produces its own set of unique characteristics, properties, and applications. A colloidal phenomenon of latex-bridged water in a dimethyl carbonate (DMC) Pickering emulsion stabilized by R202 hydrophobic silica was investigated for its ability to act as a superhydrophobic coating (SHC) for cellulose substrates. First, various emulsion compositions were screened for their stability and droplet size. The final composition was then cross-examined by cryogenic scanning electron microscopy and optical and fluorescent microscopy to verify the colloid
