Exploring the role of TWIST1 in malocclusion and craniofacial morphology.

OBJECTIVE: Despite increasing evidence that common genetic variation contributes to variation in jaw and cranial base morphology, the biological mechanisms underlying malocclusion remain poorly defined. This study tested the hypothesis that a noncoding variant near TWIST1 alters craniofacial development by disrupting transcriptional regulation, contributing to skeletal phenotypes associated with malocclusion. METHODS: In a cohort of 277 non-syndromic individuals with malocclusion, we performed targeted genotyping and deep sequencing of the TWIST1 locus, followed by multivariate genotype-pheno

Investigation of WNT2, DISC1, and DOCK4 polymorphisms in a Turkish autism spectrum disorder cohort: a case-control and bioinformatics analysis.

BACKGROUND: Autism Spectrum Disorder (ASD) is a complex neurodevelopmental disorder characterized by social communication deficits and repetitive behaviors. The Wnt signaling pathway is a fundamental regulator of neural development, governing critical processes such as synapse formation and axonal guidance. Despite strong biological rationale, population-specific genetic data for Wnt-related candidate genes remain limited. This study investigated the association of WNT2 (rs4727847), DISC1 (rs4366301), and DOCK4 (rs2217262) polymorphisms with ASD in a Turkish population. METHODS AND RESULTS: A

From Lived Experiences to Systemic Change: Patient Perspectives on the Diagnostic Journey of Loeys-Dietz Syndrome.

BACKGROUND: Loeys-Dietz syndrome (LDS) is a rare genetic connective tissue disorder characterized by early-onset aortic aneurysms and dissections, making early diagnosis crucial for patients' survival. Due to its rarity and overlap with other conditions, LDS is often challenging to diagnose. This study explored the diagnostic experiences of Canadians with LDS to identify perceived barriers, supports, and patient-informed gaps for improving diagnosis and care. METHODS: In-depth semi-structured interviews were conducted with 9 adults across Canada who were the first in their family to receive a

Associations between Dietary Selenium Intake and Risk of Incident Arthritis: Based on UK Biobank WebQ Assessment (2009-2012).

Although previous studies investigated the association between dietary selenium (Se) intake and arthritis risk, with inconsistent results. Our study aims to assess whether Se intake can delay arthritis onset through a large-scale, long-term population-based cohort. 133,230 - 153,247 arthritis-free participants from the UK Biobank were included. Arthritis was defined using ICD-10 codes. Dietary Se intake was evaluated through 24-hour dietary recall questionnaires. Association between Se intake and incident arthritis was evaluated using accelerated failure time model. Additionally, we conducted

Advancing mental health and well-being of Nigerian children through public health screening for Fragile X disorders (CHAMP-FX): protocol of a prospective multicentre screening study with longitudinal follow-up.

There is an increasing burden of neurodevelopmental disorders worldwide, with scarce data in low and middle-income countries, including Nigeria. Little is known about Fragile X disorders (Fragile X syndrome and Fragile X premutation-associated conditions) in developing countries and there is no national data in Nigeria. Advancing Mental Health and Well-being of Nigerian Children Through Public Health Screening for Fragile X Disorders (CHAMP-FX) is a multicentre public health screening initiative designed to estimate the prevalence of Fragile X disorders among children with neurodevelopmental d

Rare type I collagen variants in early-onset bicuspid aortic valve disease: Overlap with Ehlers-Danlos syndrome and osteogenesis imperfecta.

Bicuspid aortic valve (BAV) is the most common congenital heart lesion in adults and is often associated with thoracic aortic aneurysms and aortic stenosis. The genetic causes of most non-syndromic cases of BAV remain unknown. Pathogenic variants in COL1A1 or COL1A2, which encode type I collagen (COL1), cause osteogenesis imperfecta (OI), a rare disorder marked by bone fragility. Although aortic valve phenotypes, including BAV, have been described in OI, COL1 genes have not been implicated in sporadic BAV. We report rare COL1 variants in individuals with early-onset BAV (EBAV) complications. W

Decoding Primary Open-Angle Glaucoma: A Multi-Omics Approach to Identify Druggable Effector Genes.

PURPOSE: Genomewide association studies (GWAS) have identified numerous primary open angle glaucoma (POAG) risk loci, yet most reside in non-coding regions with unclear function. Mapping these loci to effector genes can elucidate disease mechanisms, identify functionally conserved variants, improve cross-ancestry risk prediction by reducing population-specific noise, and uncover shared therapeutic targets. METHODS: Here, we integrate European POAG GWAS with six types of multi-omics molecular Quantitative Trait Locis (xQTLs) using multi-trait colocalization to identify candidate effector varia

Sickle cell disease and maternity care: A qualitative synthesis of women's and health providers experiences.

INTRODUCTION: Sickle Cell Disease (SCD) is one of the most prevalent genetic diseases in the world. For women with SCD or those with Sickle Cell Trait (SCT) reproductive choices not only require consideration of genetic inheritance but also the exacerbation of pregnancy induced SCD symptoms and potential birth complications. To explore these issues a qualitative synthesis was conducted incorporating the views of women with SCD or SCT as well as healthcare professionals working with these women in maternity settings. METHODS: Database searches were performed in MEDLINE, Embase, MIDIRS, CINAHL

Comparative whole-genome sequencing of seven invasive Streptococcus pneumoniae isolates from Malaysia reveals genetic diversity, recombination events, and global lineage linkages.

BACKGROUND: Streptococcus pneumoniae remains a major global health threat, causing diseases ranging from mild respiratory infections to severe conditions like pneumonia, sepsis, and meningitis. Although pneumococcal conjugate vaccines (PCVs), including PCV7, PCV10, and PCV13 have significantly reduced disease burden, especially in children, S. pneumoniae continues to exhibit high serotype and genetic diversity. Whole-genome sequencing (WGS) analysis offers high-resolution insights into clonal lineages and multidrug-resistant strains. However, genomic data on Malaysian isolates remain limited.

Draft Genome Assembly and Annotation of a Male Mezayen Dromedary Camel of the Majaheem Type (Camelus dromedarius).

The Arabian camel (Camelus dromedarius), notably the show camels of the Mezayen tradition, holds significant cultural and socio-economic importance in the Middle East. Mezayen is a camel beauty contest tradition highlighting the remarkable aesthetic qualities of these animals. Despite the importance of these show camels, their genetic characteristics have been minimally explored. Currently, a reference genome assembly for Mezayen Arabian camels is lacking, creating a critical gap in understanding camelid genetics. This gap poses a significant barrier to advancements in selective breeding and c

First comprehensive genome analysis and antimicrobial resistance profile of Clostridium innocuum from a domestic cat.

This study aimed to investigate the molecular and phenotypic properties of a C. innocuum isolate from a healthy cat, which was previously misidentified as C. difficile by molecular testing (tpi-PCR). To resolve the misidentification and obtain more information on the isolate at the genetic level, whole genome sequencing (WGS) on the long-read sequencing platform Oxford Nanopore PromethION was performed. WGS data were analysed to identify virulence factors, antibiotic resistance, and hypothetical prophage genes. The isolate was identified as C. innocuum as a result of WGS. While several virulen

Functional comparison of control and 3' deletion human NRXN1 isoforms in C. elegans.

Neuropsychiatric conditions including Schizophrenia are historically difficulty to study in vivo due to the extensive genetic heterogeneity present between patients. Recent exome sequencing of a Schizophrenia patient cohort identified a set of 3' deletions present within multiple highly expressed α-isoforms of the synaptic adhesion molecule Neurexin1 (NRXN1), a gene that is broadly implicated across many neurodevelopmental and neuropsychiatric disorders. To understand how isoform differences and mutations within isoforms impact neuronal function and behaviors, we expressed 8 NRXN1 isoform vari

Prospective parents' views on reproductive genetic carrier screening: "You know better, you do better".

Reproductive genetic carrier screening (RGCS) allows screening for hundreds of autosomal recessive and X-linked conditions. Multiple clinical professional bodies recommend that RGCS be offered to all prospective parents. There is some research into attitudes to targeted carrier screening for conditions common in specific populations. However, the attitudes of the public to RGCS for many conditions have not been extensively studied in Australia. The aim of this study was to investigate the views of adults without a personal or familial history of a genetic condition on the inclusion of conditio

Correlation of Genetic Mutation With Outcomes in Children With Hereditary Spherocytosis Undergoing Partial Splenectomy: A Multicentre Study.

PURPOSE: Hereditary Spherocytosis (HS) is a common genetic hematological disorder causing a life-long hemolytic anemia, with sequela of hemolysis. Children with severe HS commonly undergo partial or total splenectomy (PS, TS); PS confers the theoretical advantage of maintaining splenic immune function, but may be associated with regrowth, ongoing hemolysis, and need for completion splenectomy. HS can be caused by 5 different pathogenic gene variants. A rare and severe form is caused by homozygous/compound heterozygous mutations in the SPTA1 gene, coding for alpha spectrin. We hypothesized this

Exploratory DNA methylation analysis in post-mortem heart tissue of sudden unexplained death.

BACKGROUND: Sudden unexplained death (SUD) is a devastating event in the young. Despite efforts to identify causal genetic variants, many cases remain unexplained after genetic screening. This study aimed to investigate an alternative potential contributor to SUD by studying the human methylome using the MethylationEPIC v2.0 BeadChip kit in heart tissue from SUD cases. The genome-wide methylation results of the SUD cases were compared to the results of a control cohort. The SUD cases were divided into three main groups based on their autopsy reports, heart morphology and histopathology (primar
