Developmental dyscalculia (DD) is associated with persistent arithmetic difficulties, but the temporal dynamics of operation-specific arithmetic processing remain unclear. This study examined operation-locked event-related potentials (ERPs) during addition, subtraction, and multiplication in 76 children aged 8-12 years, including 37 children with DD and 39 typically developing (TD) controls. ERPs were time-locked to the onset of the second operand, when the complete arithmetic expression became available. Children with DD showed lower accuracy and longer reaction times across all operations. E

Pediatric Charcot-Marie-Tooth (CMT) disease comprises a heterogeneous group of inherited peripheral nervous system neuropathies that manifest during childhood or adolescence. Clinically, it is characterized by progressive distal muscle weakness, atrophy, areflexia, sensory impairment, and orthopedic deformities such as pes cavus or scoliosis. In early-onset cases, hypotonia, motor delay, and limited independent ambulation may be observed. From a genetic perspective, the most frequent subtypes in the pediatric population include PMP22 duplication (CMT1A), mutations in MPZ (CMT1B), mutations in

Tuberculosis (TB) and its treatment disrupt the gut microbiome, potentially impacting immune regulation and treatment outcomes. However, host-microbial interactions during TB disease are not well characterized. This cross-sectional study investigated the gut microbiome of pulmonary tuberculosis patients—both newly diagnosed and those treated for two months—compared to healthy controls in Tanzania. TB treatment was associated with reduced microbial diversity, distinct clustering, and depletion of Euryarchaeota and Cyanobacteria. Treated patients also showed increased Fusobacteria and Tenericute

High-fat diets (HFDs) have been found to compromise male fertility, with cholesterol dysregulation being a key factor. Sterol regulatory element-binding protein 2 (SREBP2) is a crucial transcription factor that regulates cholesterol biosynthesis and uptake, playing an essential role in maintaining cholesterol homeostasis in the testes. This study investigated the dynamics of SREBP2 and cholesterol levels during rabbit spermatogenesis under HFD conditions. Our findings reveal that SREBP2 expression fluctuates throughout the seminiferous epithelium cycle. However, HFDs induce stage-specific disr

The antidiabetic efficacy of Elaeocarpus grandis, belonging to the Family Elaeocarpaceae, was investigated using the streptozotocin-induced hyperglycemia model. The ethyl acetate and petroleum ether fractions demonstrated a notable hypoglycemic impact in a rat model of type 2 diabetes mellitus (T2DM), compared to the negative control group. The chemical assessment of these fractions resulted in the isolation and identification of six known compounds (17-22). LC-HR-ESI-MS metabolomic profiling yielded the provisional identification of sixteen metabolites (1-16) across various chemical classes.

Toxic oligomeric species are suspected in the etiology of Alzheimer's disease. The full-length Aβ42 can be studied by the fragment Aβ25-35 as it retains neurotoxicity. According to experimental studies, amidation of the Aβ25-35 carboxyl terminal decreases fibrillation activity while retaining its neurotoxic properties. Our molecular dynamics simulation studied the aggregation of the Aβ25-35 trimer from two initial structures (fibril and randomized helical structures) in their amidated and nonamidated forms. Comparing the amidated and nonamidated systems, the results suggest that antiparallel c

A series of novel phenyl naphthalene-2-sulfonate-based thiosemicarbazones (5a-v) were synthesized and evaluated for their inhibitory activity against human carbonic anhydrases I and II (hCA I and hCA II). Compounds 5d and 5p demonstrated the highest inhibitory potency, with IC50 values of 4.32 ± 0.02 nM and 5.24 ± 0.03 nM for hCA I, and 3.89 ± 0.01 nM and 4.72 ± 0.01 nM for hCA II, respectively. Notably, compound 5d exhibited superior potency compared to the reference drug acetazolamide. The structure-activity relationship (SAR) analysis revealed that electron-withdrawing groups, particularly

Infectious diseases cause mass mortalities in wildlife populations globally, but the impact of host sociality on the spread of pathogens is often unknown. While host behaviors drive pathogen transmission, these behaviors vary individually which impacts both individual- and population-level disease outcomes. For example, delphinid species are regularly affected by serious respiratory diseases, but a lack of social behavior data means the structure of vulnerability in these ecosystem sentinels is poorly understood. To assess the role of variable social behavior on disease risk empirically, we co

Herein, we reported a series of quinazoline derivatives bearing amino acids by introducing a rigid pyrimidine structure between the 2 and 3-positions of the biphenyl and establishing an ionic interaction with Lys124 of PD-L1. Evaluation of the PD-1/PD-L1 inhibitory activity identified compound 7, which exhibited the most potent inhibitory activity with an IC50 value of 7.21 nM. Molecular docking was performed to demonstrate that the carboxyl group of amino acid in the tail established an ionic interaction with the ε-NH3+ of Lys124, enhancing the binding. Importantly, molecular dynamics study r

Migrants with refugee backgrounds in the Netherlands face significant reproductive health challenges, including higher rates of unintended pregnancies and limited access to contraception. This study explores how post-migration realities affect the reproductive agency of refugees from Afghanistan, Somalia, Eritrea and Syria. Utilising a participatory approach, eight peer researchers from these communities conducted eight focus-group discussions and 118 in-depth interviews, involving four migrant grassroots organisations and two Dutch non-governmental organisations. The findings reveal that refu

Cardiometabolic diseases encompass a group of conditions characterized by metabolic and inflammatory abnormalities that increase the risk of diabetes and cardiovascular disease. These syndromes involve multiple organs, including the heart, arterial system, brain, skeletal muscle, adipose tissue, hematopoietic system, liver, kidneys, and pancreas. The crosstalk between these organs contributes to the development of disease. Advances in imaging techniques, such as magnetic resonance imaging, magnetic resonance spectroscopy, computed tomography, and positron emission tomography, have revolutioniz

Radiolabeled nanocarriers that can simultaneously deliver therapeutic and diagnostic agents are promising for targeted cancer therapy. In this study, we employed molecular dynamics and metadynamics simulations to investigate how the chemical identity of drug molecules influences the behavior of technetium-99 m (⁹⁹ᵐTc)-loaded carbon nanotube (CNT) nanocarriers at the atomic scale. Four clinically relevant drugs-methotrexate, diclofenac, ketotifen, and piroxicam-were conjugated to a single-walled CNT (SWCNT) via a bifunctional chelating group. Our results revealed that drug structure significant

Dystonias are a group of neurodegenerative disorders that result in altered physiology associated with motor movements. Both the basal ganglia and the cerebellum, brain regions involved in motor learning, sensory perception integration, and reward, have been implicated in the pathology of dystonia, but the cellular and subcellular mechanisms remain diverse and for some forms of dystonia, elusive. The goal of the current review is to summarize recent evidence of cerebellar involvement in different subtypes of dystonia with a focus on Purkinje cell (PC) and cerebellar nuclei (CN) dysfunction, to

Cerebrospinal fluid (CSF) flow may assist the clearance of brain wastes, such as amyloid-β (Aβ) and tau, and thus play an important role in aging and dementias. However, a lack of non-invasive tools to assess the CSF dynamics-related clearance in humans hindered the understanding of the relevant changes in healthy aging. The global infra-slow (<0.1 Hz) brain activity measured by the global mean resting-state fMRI signal (gBOLD) was recently found to be coupled by large CSF movements. This coupling has been found to correlate with various pathologies of Alzheimer's disease (AD), particularly Aβ

Colony-stimulating factor-1-receptor (CSF1R) inhibitors have been widely used to rapidly deplete microglia from the brain, allowing the remaining microglia population to self-renew and repopulate. These new-born microglia are thought to be "rejuvenated" and have been shown to be beneficial in several disease contexts and in normal aging. Their role in Alzheimer's disease (AD) is thus of great interest as they represent a potential disease-modifying therapy. Here, we explored the differential effects of microglial depletion and repopulation during amyloid pathology progression using 5xFAD mice.

To explore the slow-release potential of indole oxygen-containing functional group derivatives in acidic media in order to reduce their negative impact on the environment. We investigated the corrosion inhibition effect of 5-Methoxy-indazole (MIA) and Methyl 1H-indazole-5-carboxylate (MIC) on copper in H2SO4 through electrochemical testing, X-ray photoelectron spectroscopy (XPS), scanning electron microscopy (SEM) analysis, and theoretical calculations. Research has found that MIA and MIC exhibit excellent corrosion inhibition performance, with MIA achieving an efficiency of up to 91.04%. SEM

Ammeline (AM) is a molecule with a very low reputation in the field of supramolecular community, but with a recently proven potential both experimentally and theoretically. In this work, dispersion-corrected density functional theory (DFT-D) computations and molecular dynamics (MD) simulations were employed to understand the aggregation mechanism of AM in chloroform and water media. Our DFT-D and MD analyses show that the most important interactions are those formed by the amine groups (-NH2) with both the pyridine-type nitrogen atoms and the carbonyl groups (C=O). In the more polar solvent, t

Mothers are commonly blamed for the ill-health of their children, and this is well documented in research. However, few studies have considered gendered patterns of blame for hereditary conditions caused by mutations from both parents through dual, shared genetic inheritance. This paper explores the 'gendering' of blame in the context of an inherited blood disorder known as sickle cell disease. The findings are drawn from 18 focus group discussions with 117 caregivers of children with sickle cell disease in Malawi and Uganda. Although one mutation from each parent is required for the disease t
