Digestive enzymes of the mucosa of the small intestine and trypsin and chymotrypsin proteolytic activity of the intestinal contents of germ-free, monocontaminated and conventional rabbits.

The activity of the membrane-bound enzymes of the microvillous zone of the entreocytes (maltase, sucrase, trehalase, lactase, cellobiase, alkaline phosphatase and leucylaminopeptidase) was studied in mucosal smears from the proximal jejunum, ileum, caecum and sigmoid flexure in a group of control (C) (8) and germ-free (GF) (7) rabbits. The trypsin and chymotrypsin activity of the contents of the ileum, caecum and sigmoid flexure was studied in 6 C, 5 GF and 5 monocontaminated (MC) rabbits. In summing up it can be stated that the individual membrane-bound enzymes have a different gradient in th

Megacystis-microcolon-intestinal hypoperistalsis syndrome: a new cause of intestinal obstruction in the newborn. Report of radiologic findings in five newborn girls.

Five newborn girls presented with small intestinal obstruction and microcolon and a giant bladder (megacystis). Organic causes of obstruction were not found, and the gastrointestinal tract failed to function after appropriate diversion. Two died in the postoperative period, two lived several months on central venous hyperalimentation, and one died at 34 months of age following chronic though intermittent hyperalimentation. Pathologic studies showed an abundance of ganglion cells in both dilated and narrowed areas of intestine; the combined small bowel-colon length was one-third of normal in th

A dual-site oral nanovaccine programs anti-colorectal tumor immunity from small intestinal priming to large intestinal boosting.

Oral vaccines hold great promise for colorectal cancer (CRC) immunotherapy but are limited by inefficient mucosal delivery and a lack of spatiotemporal control over immune activation. Here we report a spatiotemporally programmed oral nanovaccine that implements a dual-site vaccination strategy through a sequential and causally linked immune cascade, initiating immune priming in Peyer's patches of the small intestine and subsequently amplifying immunity at colorectal tumor sites. The nanovaccine (DPML@OVA-CpG), composed of antigen- and CpG-loaded mesoporous polydopamine nanoparticles stabilized

Cinnamaldehyde Attenuates Collagen-Induced Arthritis in Rats Through Immunomodulation and Alteration of Intestinal Flora Homeostasis and Repair of Intestinal Barrier Damage.

Rheumatoid arthritis (RA) is a complex autoimmune disease, the present study aimed to investigate the effects of cinnamaldehyde (CA) on collagen-induced arthritis (CIA) in rats, with particular attention to changes in intestinal barrier-related features, gut microbiota composition, and fecal short-chain fatty acids (SCFAs). In CIA rats, CA treatment alleviated arthritis-related signs, reduced inflammatory responses, and attenuated joint pathological damage. CA administration was also associated with restoration of intestinal barrier-related features, including restoration of colonic histopatho

Alginate oligosaccharides attenuate obesity-associated kidney injury through modulation of the intestinal environment with renoprotection transferable by small intestinal contents.

BACKGROUND: High-fat diet (HFD)-induced obesity is closely associated with metabolic imbalance and kidney injury. Alginate oligosaccharides (AOS) are considered promising dietary bioactive compounds; however, whether they protect against obesity-associated renal damage has not been fully clarified. In this study, we investigated whether AOS ameliorates renal injury in HFD-fed mice and further used transplantation of small intestinal contents to explore the possible involvement of gut-derived factors. METHODS: After AOS treatment, metabolic parameters, renal function, kidney histopathology, an

Intestinal-specific ketohexokinase deficiency ameliorates high fructose-induced metabolic dysfunction across the intestinal-liver-kidney axis in mice.

The small intestine, one of the three major sites for ketohexokinase (KHK, also known as fructokinase) expression, plays a crucial role in fructose-related pathological processes, primarily by regulating fructose absorption and its subsequent transport to the liver. However, the direct evidence regarding the importance of intestinal KHK on metabolic syndrome caused by fructose alone remains lacking. In this study, we found that intestinal-specific Khk deletion in mice significantly alleviated metabolic syndrome phenotypes induced by 20% fructose drinking solution. These improvements included r

FMT from IBS-D Rats Impairs Intestinal Barrier Function and is Associated with Increased Intestinal Benzoic Acid and Ruminococcus Abundance.

While the etiology of diarrhea-predominant irritable bowel syndrome (IBS-D) is multifactorial, current studies have converged to identify gut microbiota dysbiosis as a principal contributor. This study aimed to elucidate the role of the gut microbiota in IBS-D progression and to uncover the underlying mechanisms. In this study, a rat model of IBS-D was successfully established, characterized by prominent visceral hypersensitivity and diarrhea. To assess the impact of the gut microbiota, recipient rats pretreated with broad-spectrum antibiotics underwent fecal microbiota transplantation (FMT) f

Proteomic reprogramming of ileal epithelial cells during homologous superimposed intestinal trematode infection reveals coordinated restoration of intestinal homeostasis.

BACKGROUND: Intestinal helminth infections trigger complex host responses, determining parasite survival and tissue homeostasis. Primary Echinostoma caproni infection disrupts epithelial metabolism, differentiation, and repair in an IL-25-deficient environment, as shown in a previous study by our research group; however, the adaptive mechanisms during homologous superimposed infections remain unclear. METHODOLOGY/PRINCIPAL FINDINGS: Male ICR mice were assigned to control, primary infection, and homologous superimposed infection groups, and ileal epithelial cells were isolated for proteomic pr

Human placenta mesenchymal stromal cells alleviate intestinal inflammation and repair intestinal barrier function by activating AMPK-FXR pathway.

Crohn's disease (CD) has a complex pathogenesis; there is currently no effective treatment. Mesenchymal stromal cells (MSCs) are a potential therapeutic option for CD. It is important to systematically evaluate the safety and effectiveness of MSCs and their mechanism for the treatment of CD to support their clinical application. Here, the safety and effectiveness of MSCs from the human placenta (hPMSCs) are evaluated. All the indicators show that hPMSC transplantation is safe and effectively alleviates intestinal inflammation, reduces intestinal apoptosis and fibrosis, and promotes recovery of

Macrophage extracellular traps impair intestinal barrier function in DSS-induced colitis in mice by inducing intestinal epithelial cell apoptosis.

Ulcerative colitis (UC) is a chronic inflammatory bowel disease with recurrent mucosal inflammation in the colon, with macrophages emerging as one of the key effector cells in its pathogenesis. Recently, macrophage extracellular traps (METs), mediated by peptidyl arginine deiminase 4 (PAD4), have emerged as potential contributors to inflammatory disorders. However, the specific mechanism by which METs contribute to the development and progression of UC remains unclear. In this study, we identified a significant presence of METs in the colonic tissues of clinical UC patients. Moreover, we found

X-Ray Irradiation Induces Oxidative Stress and Upregulates Intestinal Nrf2-Mrp2 Pathway, Leading to Decreased Intestinal Absorption of Valsartan.

Background: It has been documented that radiation can influence the pharmacokinetics of chemotherapy drugs, yet the underlying mechanisms remain poorly understood. In clinical practice, a considerable number of cancer patients undergo radiotherapy, and those with comorbid hypertension required antihypertensive drugs, including valsartan, an angiotensin II receptor blocker. However, there is no research investigating whether radiotherapy poses a risk of altering the pharmacokinetics. Objective: The objective of this study is to investigate the impact of X-ray abdominal irradiation on the pharma

Effects of Bacillus amyloliquefaciens F1 postbiotics on growth performance, serum biochemistry, intestinal morphology and intestinal microbiota of yellow-feathered broilers.

The extensive use of antibiotics in poultry production has contributed to antimicrobial resistance, creating a need for safe and stable alternatives. This study evaluated the effects of Bacillus amyloliquefaciens F1 postbiotics (BAP) on growth performance, antioxidant status, intestinal morphology, and cecal microbiota in yellow-feathered broilers. A total of 360 one-day-old male broilers were randomly and evenly assigned to three dietary treatments for 63 days: a basal diet (control, CON), the basal diet separately supplemented with 100 g/t (BAP100) and 150 g/t (BAP150) of BAP. At 21 days, br

Ochratoxin A induces intestinal barrier impairment and concurrent hepatorenal toxicity in broiler chicks via modulation of autophagy, ferroptosis, and the intestinal microbiota.

Ochratoxin A (OTA), a mycotoxin prevalent in contaminated feed, induces systemic toxicity upon intestinal absorption. In this study, broiler chicks were randomly divided into control, vehicle, or OTA-treated groups (0.05, 0.1, and 0.2 mg/kg bw/day). The results indicated that OTA caused histopathological damage to the intestine, liver, and kidney. Notably, OTA at 0.2 mg/kg bw induced hepatocellular injury, as evidenced by significantly elevated serum AST levels and hepatic inflammatory infiltration, and triggered renal damage characterized by increased relative kidney weight and tubular cell f

Monomorphic Epitheliotropic Intestinal T-Cell Lymphoma (MEITL) Causing Small Intestinal Perforation Associated With Adnexal Metastasis: A Case Report.

Monomorphic epitheliotropic intestinal T-cell lymphoma (MEITL) is a rare and aggressive form of peripheral T-cell lymphoma originating from intestinal epithelial lymphocytes and is associated with a poor prognosis. We present the case of a 50-year-old woman who developed a gastrointestinal perforation, initially suspected to be due to ovarian cancer with peritoneal dissemination. The patient underwent laparoscopic adnexal resection and laparotomy for the intestinal perforation. Postoperative pathology confirmed that the cause of the intestinal perforation and enlarged adnexa was MEITL. Prompt
